Videogames and their specific devices can be used to improve learning process since they are very attractive for children. In this way, pupils increase their cognitive skills, the time dedicated to learn, their motivation for learning, their concentration and their attention while they are working/playing. The subject of “learning by play” is behind the introduction of recreational educational techniques in the classroom. If we also consider the increasing presence of new technologies in society in general and in classrooms in particular, we encounter a new way of teaching/learning. Moreover, several studies in the area of computer supported collaborative learning (CSCL) have proved that learning in a group environment (both actively and interactively) is much more productive for pupils than traditional education.Our main objective is to reduce the complexity of introducing the collaborative learning techniques into development of educational videogames. So, in this paper we analyze the use of videogames as a particular case of new technologies in the classroom and we present a set of design guidelines to enable us to incorporate the features of collaborative learning in the videogame development process. We also explore how these guidelines affect the videogame architecture and how they can be applied when designing a videogame. As a practical example of using our proposal we have designed an educational videogame with group activities which aim is to learn the vowels.  相似文献

The aim of this study was to assess the hygienic quality of raw milk used in the manufacture of S?o Jorge, a Protected Denomination of Origin Portuguese semihard cheese, as well as to ascertain the sanitary conditions prevailing during its processing. Viable counts of Enterobacteriaceae and Micrococcaceae were accordingly obtained, pertaining to 21 independent batches (including samples of raw milk, curd, and cheeses after 1, 3, and 4 months of ripening), from 7 dairy farms. Standard plate counts (log CFU per milliliter or per gram) ranged from 6.1 to 8.6 in raw milk, whereas they ranged from 7.0 to 8.0 in 4-month-old cheeses. Viable counts of Enterobacteriaceae ranged between 5.9 and 7.0 in raw milk and between 0.0 and 1.3 in 4-month-old cheeses. Species identified within this family encompassed Klebsiella oxytoca, Klebsiella cloacae, Klebsiella pneumoniae, Enterobacter sakazakii, and Escherichia coli; Klebsiella ornithinolytica, Klebsiella terrigena, and Serratia odorifera were detected only in raw milk. No Salmonella whatsoever could be detected in any of the samples. Viable counts of Micrococcaceae ranged between 4.7 and 5.9 and between 1.3 and 3.3 in raw milk and 4-month-old cheeses, respectively. Species identified within this family encompassed Staphylococcus sciuri, Staphylococcus epidermidis, Staphylococcus saprophyticus (which was found mainly in ripened cheeses), and Staphylococcus aureus (which was not detected in 4-month-old cheeses). Accompanying physicochemical analyses included determination of moisture, salt, and pH. Statistical analyses revealed a negative correlation between salt content and viable numbers of Enterobacteriaceae in cheese, whereas in the case of Micrococcaceae, a more negative correlation was found between viable numbers and moisture content than between viable numbers and pH. The results of our study indicate, in general, poor milk handling conditions in all farms, given that the indicators total mesophile and Enterobacteriaceae counts were high, between 100- and 1,000-fold those enforced by international standards pertaining to the matrices in question. However, by the time of regular consumption (i.e., after 4 months of ripening), S?o Jorge cheeses exhibit low levels of contamination by Enterobacteriaceae and S. aureus, as well as absence of Salmonella.  相似文献

Heptadecenoic acid (17:1) is a minor constituent of ruminant fats and its isomeric definition remains undefined in most reports on ruminant milk and intramuscular fat. Samples of milk and intramuscular fat of bovine, ovine, and caprine origin were analyzed by gas chromatography (GC) using 3 capillary columns with and without addition of 17:1 cis-10. Additionally, cis isomers of ovine milk fat samples were isolated as methyl esters by preparative thin-layer chromatography and analyzed by GC. The structural analysis of 17:1 present in samples was achieved by chemical ionization tandem mass spectrometry techniques. The isomer 17:1 cis-9 is the overwhelming heptadecenoic isomer in ruminant milk and intramuscular fat; 17:1 cis-10 is virtually absent. Moreover, current GC methods were able to resolve cis-9 from cis-10 and cis-8 isomers, so reports on 17:1 contents in ruminant fat should define its isomeric composition.  相似文献

The main purpose of this work was to optimize and validate a new isocratic high-performance liquid chromatography method for thiamine analysis in wheat flours. Thiamine was previously derivatized into thiochrome and determined by fluorescence detection (excitation wavelength, 366 nm; emission wavelength, 435 nm). The mobile phase consisted of buffer phosphate (0.02 M) and organic modifier (methanol 88 %; acetonitrile 12 %). Optimization was done using a Box–Behnken design (three factors at three levels) where organic modifier proportion (A), varying from 20 to 90 %, mobile phase pH (B), varying from 5.8 to 7.8, and flow rate (C), varying from 0.5 to 1 mL min?1, were the studied variables. Peak width was the studied response. Optimal chromatographic conditions that minimized response were A?=?90 %, B?=?6.00, and C?=?1 mL min?1. Using optimal conditions, thiochrome extractions with isobutanol and solid-phase extraction (SPE) employing styrene-divinylbenzene adsorbent cartridges were compared. Method validation, using thiamine standard solutions and both types of thiochrome isolation, included linearity studies, limits of detection and quantification, and calibration and analytical sensitivity quantifications. Standard addition method was employed to quantify thiamine in wheat flours. Good precision results were obtained by both extraction methods. Recoveries ranged from 63 to 65 % for isobutanol and 84 to 101 % for SPE, including sample extraction, thiamine derivatization, thiochrome purification, and chromatographic separation. Taking into account better obtained SPE recoveries, acceptable precision, and isobutanol irritant action on the eyes and skin of operator, SPE using styrene-divinylbenzene adsorbent could be chosen for routine thiamine analysis in wheat flour samples.  相似文献

Few studies have considered immune-mediated inflammatory disorders (IMID) together, which is necessary to adequately understand them given they share common mechanisms. Our goal was to investigate the expression of vasoactive intestinal peptide (VIP) and its receptors VPAC1 and VPAC2 in selected IMID, analyze the effect of biological therapies on them, and identify miRNA signatures associated with their expression. Serum VIP levels and mRNA of VPAC and miRNA expression in peripheral blood mononuclear cells were analyzed from 52 patients with psoriasis, rheumatoid arthritis, Graves’ disease, or spondyloarthritis and from 38 healthy subjects. IMID patients showed higher levels of VIP and increased expression of VPAC2 compared to controls (p < 0.0001 and p < 0.0192, respectively). Receiver operating characteristic curve analysis showed that the levels of VIP or VPAC2 expression were adequate discriminators capable of identifying IMID. Treatment of IMID patients with anti-TNFα and anti-IL12/23 significantly affected serum VIP levels. We identified miRNA signatures associated with levels of serum VIP and VPAC2 expression, which correlated with IMID diagnosis of the patients. The results indicate that the expression of VIP/VPAC2 is able of identify IMIDs and open up a line of research based on the association between the VIP/VPAC axis and miRNA signatures in immune-mediated diseases.  相似文献

Until now, few studies have proposed analytical short-cut methods for reliably designing multicomponent reactive distillation columns. Therefore, in this study we have improved and extended a design methodology for the design of RD columns of multicomponent systems. We have developed a graphical design method, based on distillation lines and tray-by-tray calculations defined in terms of reaction-invariant composition variables, to determine RD design parameters such as the number of theoretical stages, operating reflux ratio, the feed tray location and the top or bottom flow. In this note, we report our extended and improved method, which is analytical and useful for reliably determining the design parameters of multicomponent RD systems. We study the synthesis of TAME with inert components (with different feed thermal conditions) as case of study to show the effectiveness of the proposed strategy. Results obtained with our strategy show a significant agreement with those obtained using a rigorous model of commercial simulator AspenONE Aspen Plus®.  相似文献

Signaling mediated by cytokines and chemokines is involved in glaucoma-associated neuroinflammation and in the damage of retinal ganglion cells (RGCs). Using multiplexed immunoassay and immunohistochemical techniques in a glaucoma mouse model at different time points after ocular hypertension (OHT), we analyzed (i) the expression of pro-inflammatory cytokines, anti-inflammatory cytokines, BDNF, VEGF, and fractalkine; and (ii) the number of Brn3a+ RGCs. In OHT eyes, there was an upregulation of (i) IFN-γ at days 3, 5, and 15; (ii) IL-4 at days 1, 3, 5, and 7 and IL-10 at days 3 and 5 (coinciding with downregulation of IL1-β at days 1, 5, and 7); (iii) IL-6 at days 1, 3, and 5; (iv) fractalkine and VEGF at day 1; and (v) BDNF at days 1, 3, 7, and 15. In contralateral eyes, there were (i) an upregulation of IL-1β at days 1 and 3 and a downregulation at day 7, coinciding with the downregulation of IL4 at days 3 and 5 and the upregulation at day 7; (ii) an upregulation of IL-6 at days 1, 5, and 7 and a downregulation at 15 days; (iii) an upregulation of IL-10 at days 3 and 7; and (iv) an upregulation of IL-17 at day 15. In OHT eyes, there was a reduction in the Brn3a+ RGCs number at days 3, 5, 7, and 15. OHT changes cytokine levels in both OHT and contralateral eyes at different time points after OHT induction, confirming the immune system involvement in glaucomatous neurodegeneration.  相似文献

Ouabain is a cardiac glycoside that has been described as a hormone, with interesting effects on epithelial physiology. We have shown previously that ouabain induces gap junctional intercellular communication (GJIC) in wild, sensitive cells (MDCK-S), but not in cells that have become insensitive (MDCK-I) by modifying their Na+-K+-ATPase. We have also demonstrated that prostaglandin E2 (PGE2) is able to induce increased GJIC by a mechanism other than ouabain, that does not depend on Na+-K+-ATPase. In this work we show, by dye transfer assays, that when MDCK-S and MDCK-I are randomly mixed, to form monolayers, the latter stablish GJIC, because of stimulation by a compound released to the extracellular media, by MDCK-S cells, after treatment with ouabain, as evidenced by the fact that monolayers of only MDCK-I cells, treated with a conditioned medium (CM) that is obtained after incubation of MDCK-S monolayers with ouabain, significantly increase their GJIC. The further finding that either (1) pre-treatment with COX-2 inhibitors or (2) addition to CM of antagonists of EP2 receptor abolish CM’s ability to induce GJIC in MDCK-I monolayers indicate that PGE2 is the GJIC-inducing compound. Therefore, these results indicate that, in addition to direct stimulation, mediated by Na+-K+-ATPase, ouabain enhances GJIC indirectly through the paracrine production of PGE2.  相似文献

In the last decades, the need of cleaner and renewable fuels has increased the use of alcohols to power internal combustion engines. The ABE (acetone–butanol–ethanol) method to produce purified biobutanol and bioethanol is the most known and well-studied method to produce those alcohols. Nevertheless, it is not yet clear how the bioethanol’s content in the biobutanol stream could affect the engine’s biofueled performance. Thus, this study focuses on (1) the performance of an internal combustion engine and (2) on the different compositions of n-butanol/ethanol mixtures, up to 20 vol%, to assess the operative issues of the use of both alcohols instead of pure n-butanol. In order to get the most reliable results of the engine’s performance, we conducted three different experiments for each alcohols’ composition, including conventional gasoline testing as reference. The experimental tests were carried out in a CT 150 Internal Combustion Engine Basic Module and conducted at various speeds—from 700 to 1500 rpm—resulting in load variation. In general, the changes in the composition of ethanol in n-butanol do not seem to affect the performance of the engine considerably, as variations occur at an approximate rate of 3.7% in effective power, 3.3% in torque and 5.4% in specific fuel consumption.  相似文献
