Searching for adequate and effective compounds displaying antimicrobial activities, especially against Gram-positive bacteria, is an important research area due to the high hospitalization and mortality rates of these bacterial infections in both the human and veterinary fields. In this work, we explored (E)-4-amino-3-((3,5-di-tert-butyl-2-hydroxybenzylidene)amino) benzoic acid (SB-1, harboring an intramolecular hydrogen bond) and (E)-2-((4-nitrobenzilidene)amino)aniline (SB-2), two Schiff bases derivatives. Results demonstrated that SB-1 showed an antibacterial activity determined by the minimal inhibitory concentration (MIC) against Staphylococcus aureus, Enterococcus faecalis, and Bacillus cereus (Gram-positive bacteria involved in human and animal diseases such as skin infections, pneumonia, diarrheal syndrome, and urinary tract infections, among others), which was similar to that shown by the classical antibiotic chloramphenicol. By contrast, this compound showed no effect against Gram-negative bacteria (Klebsiella pneumoniae, Escherichia coli, and Salmonella enterica). Furthermore, we provide a comprehensive physicochemical and theoretical characterization of SB-1 (as well as several analyses for SB-2), including elemental analysis, ESMS, 1H and 13C NMR (assigned by 1D and 2D techniques), DEPT, UV-Vis, FTIR, and cyclic voltammetry. Evidence from dental-related stem cells (DRSCs) suggests an enhanced potential for ectodermal lineage differentiation due to their neural crest origin. Growing evidence that DRSC cultures can produce cells with a neural crest-derived stem cell (NCSC)-like phenotype supports their potential for future therapeutic approaches for neurodegenerative diseases and nerve injuries. However, most of the evidence is limited to the characterization of DRSCs as NCSCs by detecting the expression of neural crest markers. Only a few studies have provided proof of concept of an improved neuro-glial differentiation or direct applicability in relevant models. In addition, a current problem is that several of the existing protocols do not meet manufacturing standards for transferability to a clinical scenario. This review describes the current protocols to obtain NCSCs from DRSCs and their characterization. Also, it provides important considerations from previous work where DRSCs were established and characterized as mesenchymal stromal cells but studied for their neuro-glial differentiation potential. The therapeutic advancement of DRSCs would depend on establishing protocols that can yield a neural crest-like phenotype efficiently, using appropriate manufacturing standards and testing them in relevant models of disease or injury. Aggregation of high affinity IgE Fc receptors (Fc epsilon RI) on RBL-2H3 cells results in tyrosine phosphorylation of 33-, 42-, 44-, 72-, 80-, 90-, 125-kDa proteins. The 42 and 44 kDa proteins were identified as mitogen-activated protein (MAP) kinases with immunoblotting of anti-MAP kinase antibody. The effects of an antiallergic drug, pemirolast potassium (TBX) on Ag-induced protein tyrosine phosphorylation and MAP kinase activation were investigated. When RBL-2H3 cells were stimulated with Ag in the presence of TBX, tyrosine phosphorylation of three proteins (33, 42 and 44 kDa) was inhibited concentration-dependently (0.1-10 micrograms/ml). Inhibition of Ag-induced tyrosine phosphorylation of 33 kDa protein, which could be a beta subunit of Fc epsilon RI, suggests that TBX may prevent the activation of Fc epsilon RI. TBX suppressed activation of MAP kinases (42 and 44 kDa) in response to Ag as well as phorbol myristate acetate (100 nM) or calcium ionophore A23187 (500 nM), implying that the drug acts on signal transduction component(s) between the second messengers and MAP kinases. However, TBX had no effects on protein tyrosine phosphorylation and MAP kinase activation in MC3T3-E1 osteoblastic cells. These results indicate that TBX may affect Fc epsilon RI and also may act as a step distal of Ca2+ mobilization and protein kinase C activation leading to MAP kinase activation in RBL-2H3 cells. 相似文献

In this study, we proposed an in vitro tumor model to simulate the mechanical microenvironment and investigate the effect of compressive stress on the invasion process of malignant tumors. It has been pointed out that the biomechanical environment, as well as the biochemical environment, could affect the transformation of cancer cell migration, invasion, and metastasis. We hypothesized that the solid stress caused by the exclusion of surrounding tissue could transform tumor cells from noninvasive to invasive phenotypes. Colorectal cell spheroids were embedded and cultured in agarose gels of varying concentrations to simulate the earliest stages of tumor formation and invasion. The spheroids embedded in gels at higher concentrations showed peculiar growth after 72 h of culture, and the external compressive loading imposed on them caused peculiar growth even in the gels at lower concentrations. In conclusion, the mechanical microenvironment caused the transformation of tumor cell phenotypes, promoting the growth and invasion of tumor cell spheroids. 相似文献

Emerging deep learning-based applications in precision medicine include computational histopathological analysis. However, there is a lack of the required training image datasets to generate classification and detection models. This phenomenon occurs mainly due to human factors that make it difficult to obtain well-annotated data. The present study provides a curated public collection of histopathological images (DeepHP) and a convolutional neural network model for diagnosing gastritis. Images from gastric biopsy histopathological exams were used to investigate the performance of the proposed model in detecting gastric mucosa with Helicobacter pylori infection. The DeepHP database comprises 394,926 histopathological images, of which 111 K were labeled as Helicobacter pylori positive and 283 K were Helicobacter pylori negative. We investigated the classification performance of three Convolutional Neural Network architectures. The models were tested and validated with two distinct image sets of 15% (59K patches) chosen randomly. The VGG16 architecture showed the best results with an Area Under the Curve of 0.998%. The results showed that CNN could be used to classify histopathological images from gastric mucosa with marked precision. The effects of the SAR (14 and 38) and of the methodology of introduction of nickel (wetness impregnation and ion exchange) on catalyst activation were investigated in Ni/USY model catalysts submitted to two activation treatments: reduction (A) and reduction/calcination/reduction, successively (B). the catalysts prepared by wetness impregnation, a marked increase in the catalytic activity after treatment B was observed. On the basis of the catalyst efficiency toward ethane hydrogenolysis, the lower SAR catalyst presents a threefold higher activity. On the contrary, the catalysts prepared by ion exchange, as well as a model catalyst prepared by wetness impregnation over a USY exhaustively exchanged with nickel ions, do not present measurable increasing of the catalytic activity under the same activation conditions. Monitoring the catalyst activation by EXAFS indicates the formation of nickel clusters in the impregnated catalysts after activation treatment B, which should be responsible for the high catalytic activity. However, TEM images reveal a bimodal particle size distribution, with large (ca. 20 nm) and small nickel particles. Temperature-programmed reduction performed under the same conditions of catalyst activation suggests that only the small nickel particles are activated under the experimental conditions adopted in this work. 相似文献

Changing the pore morphology of hydrogels can be an effective strategy to modulate their drug release profiles. Herein, Pluronic F127 was used to change the three-dimensional pore morphology of crosslinked poly(N-isopropylacrylamide-co-acrylic acid) (P[NIPAm-co-AAc]) hydrogels. F127 reduced the pore diameters from 20 ± 4 to 2.9 ± 0.4 μm and from 11 ± 1 to 1.4 ± 0.4 μm in hydrogels synthesized at 8 and 30°C, respectively. Small-angle X-ray scattering indicates that the segregation of the F127 during the polymerization process induces F127 phase transitions from unimers (at 8°C) or cubic-packed micelles (at 30°C) to a lamellar structure. P(NIPAm-co-AAc) hydrogels charged with S-nitrosoglutathione (GSNO), released nitric oxide (NO) spontaneously during hydration. The decrease in the pore diameter led to a twofold to threefold increase in the rate of water absorption and a fourfold to sixfold increase in the rate of NO release of the hydrogels. F127 can be used to change the pore morphology of P(NIPAm-co-AAc) hydrogels, with concomitant changes in their rate of hydration and NO release from GSNO, opening a new perspective for their use in topical NO delivery. 相似文献

When a benzene/cyclohexane mixture was permeated through poly(dimethyl acrylamide-random-methyl methacrylate) (DMAA-r-MMA)
and poly(dimethyl acrylamide)-graft-poly(methyl methacrylate) (DMAA-g-MMA) membranes by pervaporation, the benzene-permselectivity
of the DMAA-r-MMA membrane changed from the diffusivity selectivity to the solubility selectivity with increasing DMAA content
but DMAA-g-MMA membranes with a high DMAA content had the higher apparent diffusivity selectivity than the apparent solubility
selectivity. Furthermore, the apparent solubility selectivity for a benzene/cyclohexane mixture between the DMAA-r-MMA membrane
and the DMAA-g-MMA membrane with a high DMAA content was remarkably different. These results were attributed to the difference
of structure between the copolymers.

The high-speed melt spinning of poly(ethylene 2,6-naphthalene dicarboxylate) (PEN) was performed up to the take-up velocity of the ultra-high-speed region, 9 km/min. From the investigations of the structure and physical properties of the as-spun fibers, the high-speed spinning of PEN was divided into three regions in terms of the mechanism of fiber structure formation. The first region is the take-up velocity of up to 2.5 km/min and the birefringence of up to 0.08 where only a slight increase in molecular orientation was attained. At the take-up velocity of 2.5–4.5 km/min and the birefringence of 0.08–0.25, although some experimental evidences indicated that the orientation-induced crystallization did not occur, there was an increase in the fiber density which suggested the formation of some ordered structure. At the take-up velocity > 4.5 km/min and birefringence > 0.25, the orientation-induced crystallization occurred. The fibers obtained in this region were characterized by the formation of the crystalline structure dominated by the β form. The presence of the necklike deformation in the spinning line was also confirmed. The solidification temperature of the spinning line analyzed from the diameter profile suggested that the formation of β modification crystals occurred at relatively low crystallization temperatures in comparison with that in an isotropic state. Therefore it was indicated that the presence of elongational stress in the spinning line promoted the formation of the β modification crystals. © 1997 John Wiley & Sons, Inc.
