This study quantified Escherichia coli (EC) and enterococci (ENT) in beach waters and dominant source materials, correlated these with ambient conditions, and determined selected EC genotypes and ENT phenotypes. Bathing-water ENT criteria were exceeded more frequently than EC criteria, providing conflicting interpretations of water quality. Dominant sources of EC and ENT were bird feces (10(8)/d/bird), storm drains (10(7)/d), and river water (10(11)/d); beach sands, shallow groundwater and detritus were additional sources. Beach-water EC genotypes and ENT phenotypes formed clusters with those from all source types, reflecting diffuse inputs. Some ENT isolates had phenotypes similar to those of human pathogens and/or exhibited high-level resistance to human-use antibiotics. EC and ENT concentrations were influenced by collection time and wind direction. There was a 48-72-h lag between rainfall and elevated EC concentrations at three southern shoreline beaches, but no such lag at western and eastern shoreline beaches, reflecting the influence of beach orientation with respect to cyclic (3-5 d) summer weather patterns. In addition to local contamination sources and processes, conceptual or predictive models of Great Lakes beach water quality should consider regional weather patterns, lake hydrodynamics, and the influence of monitoring method variables (time of day, frequency). 相似文献

In an earlier study, we showed that feeding CLA immediately after weaning prolonged survival of NZB/W F1 mice after onset
of proteinuria. In the present study, the feeding of CLA was delayed until mice had developed proteinuria. Thirty NZB/W F1
mice were fed a regular rodent chow after weaning. Urine samples were collected to detect proteinuria. Once a mouse was proteinuria
positive, it was then randomly assigned to a 0.5% CLA supplement semipurified diet or a control diet (supplement 0.5% corn
oil). The next proteinuria positive mouse was then assigned to the opposite diet to which the first mouse was assigned. Mice
fed the control diet lost 25% more body weight (13.0 g) than mice fed the CLA diet (9.7 g). Moreover, CLA-fed mice survived
an average 1.7-fold longer (148 d) than mice fed the control diet (89 d) after the onset of proteinuria. This follow-up study
confirmed that dietary CLA had a beneficial effect in the autoimmune NZB/W F1 mouse. In summary, the cachectic symptom of

We report a new method that gives atomic resolution in the reconstruction of simulated holograms in theoretical low energy electron point source (LEEPS) microscopy, and that uses a screen size that is commensurate with screen sizes used in experimental LEEPS. The method exploits the spherical symmetry in the electron waves emerging from the source. We compare holograms obtained by rotating the screen about an axis passing through the point source as opposed to rotating the atomic cluster in the opposite sense about the same axis. We show that, by generating and combining simulated holograms obtained by rotating the cluster, with the screen held fixed, a composite hologram, comprised of the individual holograms, captures enough information that atomic resolution in the reconstructions is obtained. A key feature is to choose the rotations to optimize the collective interference pattern on the composite hologram. This results in sharper resolution while using a considerably smaller screen size; results are reported for a screen size about ten times smaller than screen sizes typically used in theoretical LEEPS. The method used gives commensurate or better resolution on comparison to results obtained using the larger screen size. Possible implications for experimental LEEPS are briefly discussed. 相似文献

Toxicity to organisms is usually expressed in terms of an observable effect on individuals from which a summary endpoint (such as the NOEC or ECx) is derived for risk assessment and environmental quality standards. However, toxicity evaluated in terms of a demographic endpoint may be more relevant to such regulatory applications. In this paper the effect of toxicity on population growth rate r is explored in tandem with a 'double bootstrap' to incorporate uncertainty. Exemplifying the approach with a set of individualized life table response data obtained for Daphnia magna exposed to zinc sulphate solution, the influence of increasing concentrations is assessed. A demographic-based metric for r, the ErCx (effect on r concentration percentage), is defined to permit alternative population level estimation of a 'safe effect' concentration. 相似文献

The electron-beam (EB) degradation of volatile aromatics (benzene, toluene, ethylbenzene, xylenes: BTEX) in groundwater strip gas, which in the present work has been modeled by the introduction of the desired aromatic(s) to a stream of air or another gas, such as oxygen, is initiated essentially by the addition of *OH radicals to the aromatic ring, giving rise to hydroxycyclohexadienyl radicals, which form the corresponding peroxyl radicals upon addition of oxygen. As studied in some detail with benzene as a BTEX representative, various reactions of these lead to numerous oxidation products in a cascade of reactions, including the decomposition of products under the prevailing conditions of high turnover of the initial aromatic. Importantly, hydroxycyclohexadienylperoxyl radical formation is partly reversible, and the reactions of the hydroxycyclohexadienyl radicals, which thus have a significant presence in these systems, must therefore also be taken into consideration. In the gas phase, in contrast to the aqueous phase (see Part 1), the reactions of the hydroxycyclohexadienyl radicals lead to oligomeric products that appear to contribute, in addition to ionic clusters, to nucleation for the aerosols observed. Various nitrated products, among them nitrophenols, are observed when air is used for the stripping. However, these studies did not clear the pilot plant stage, since BTEX degradation using a bioreactor carried out in parallel was so successful that the EB technology was judged to be noncompetitive. As for the latter, expensive equipment consisting of a stripper, the EB machine, and an aerosol precipitator would be required. In this study, the interaction of a bioluminescence-labeled Escherichia coli strain with growing spinach plants was assessed. Through bioluminescence profiles, the direct visualization of E. coli growing around the roots of developing seedlings was accomplished. Subsequent in situ glucuronidase (GUS) staining of seedlings confirmed that E. coli had become internalized within root tissue and, to a limited extent, within hypocotyls. When inoculated seeds were sown in soil microcosms and cultivated for 42 days, E. coli was recovered from the external surfaces of spinach roots and leaves as well as from surface-sterilized roots. When 20-day-old spinach seedlings (from uninoculated seeds) were transferred to soil inoculated with E. coli, the bacterium became established on the plant surface, but internalization into the inner root tissue was restricted. However, for seedlings transferred to a hydroponic system containing 10(2) or 10(3) CFU of E. coli per ml of the circulating nutrient solution, the bacterium was recovered from surface-sterilized roots, indicating that it had been internalized. Differences between E. coli interactions in the soil and those in the hydroponic system may be attributed to greater accessibility of the roots in the latter model. Alternatively, the presence of a competitive microflora in soil may have restricted root colonization by E. coli. The implications of this study's findings with regard to the microbiological safety of minimally processed vegetables are discussed. 相似文献

This is the final report of a panel set up by the U.S. Department of Energy (DOE) Fusion Energy Sciences Advisory Committee (FESAC) in response to a charge letter from Dr. Ray Orbach (Appendix A). In that letter, Dr. Orbach asked FESAC for an assessment of the present status of inertial fusion energy (IFE) research carried out in contributing programs. These programs include the heavy ion (HI) beam, the high average power laser (HAPL), and Z-Pinch drivers and associated technologies, including fast ignition (FI). This paper characterizes factors affecting the accuracy of the correlation coefficient (CC) template matching algorithm, as applied to motion tracking from two-dimensional real-time coronary artery magnetic resonance images. The performance of this algorithm is analyzed in the presence of both random and systematic error. In the presence of random error, it is shown that a necessary and sufficient condition for accurate motion tracking is a large CC difference-to-noise ratio (CCDNR). The CCDNR itself is in turn affected by five factors: image and template size, image and template structure, and the magnitude of the noise. Techniques are introduced for manipulating some of these factors in order to increase the CCDNR for greater motion tracking accuracy. In the presence of superimposed systematic error it is shown that, while large CCDNR is necessary, it alone is not sufficient to ensure accurate motion tracking. Techniques are developed for improving motion tracking accuracy that minimize the effects of systematic error, while maintaining an adequate CCDNR level. The ability of these techniques to improve motion tracking accuracy is demonstrated both in phantoms and in coronary artery images. 相似文献

The majority of organic carbon in the oceans is present as dissolved organic matter (DOM); therefore understanding the distribution and dynamics of DOM is central to understanding global carbon cycles. Describing the time-space variability in colored dissolved organic matter (CDOM) has been difficult, as standard spectrophotometric methods for CDOM determination are laborious and susceptible to methodological biases. Previously, measurements of CDOM absorption in discrete water samples by use of a liquid-waveguide capillary cell (LWCC) compared favorably with measurements made with a benchtop spectrophotometer. Given this, we focused on automating the LWCC technique to improve our spatial and temporal sampling capabilities for CDOM. We found strong correlations between CDOM absorption spectra collected from discrete water samples using standard methods and selected corresponding CDOM spectra collected by the automated LWCC system. The near-continuous measurements by the LWCC system made it possible to map the temporal, spatial, and spectral variability of CDOM absorption along the ship track. 相似文献
