This study was performed to estimate the pool of SCFA produced in the digestive tract of growing pigs after the consumption of diets containing dif-ferent sources of resistant starch (RS) (15 g per meal) (potato starch (PoS), high amylose maize starch (HS) and retrograded extruded HS (RHS)). SCFA production in vitro was respectively 19, 23 and 21 mmol g−1 of fermented PoS, HS and RHS. The highest butyric acid production was observed with PoS. Fifty six, 36 and 19% respectively of ingested PoS, HS and RHS escaped digestion in the small intestine, whereas residual starch was completely fermented in the large bowel. Luminal total SCFA in the caeco-colon 7 h after the experimental meal were respectively 33, 78 and 105 mmol for PoS, HS and RHS, with PoS providing the highest production of butyric acid. Theoretical in vivo SCFA production, calculated on the basis of results for in vitro fermentation and ileal digestibility, differed from in vivo results for PoS (+65%) and HS (-13%), but not for RHS. This study reveals different rates of disappearance of RS both in vitro and in vivo. © 1998 SCI.  相似文献

Electron spin resonance (ESR) was used in order to study the mechanism of interaction of potato starch with lipids in systems with different water contents (40, 50 and 60% w/w) during heating and cooling. Different spin probes were used, on the one hand spin‐labelled stearic acids (5‐DSA and 16‐DSA), which limited lipids, and on the other hand the water‐soluble probe 4‐hydroxy‐2,2,6,6‐tetramethylpiperidine‐1‐oxyl (Tempol), which was sensitive to changes in the dynamic properties of the water phase associated with the temperature‐induced starch structural transformations. Whatever the water content in the hydration range considered, Tempol was observed to be relatively mobile in the presence of potato starch, whereas a strong absorption of spin‐labelled stearic acids on potato starch granules took place at room temperature for all investigated starch–water systems. Spin‐labelled stearic acids were expected to stick on the starch granules. The mobility of the spin‐labelled stearic acid was less upon cooling than upon heating as a result of starch gelation. The mobility of spin probes decreased with increasing starch concentration. Introduction of a doxyl fragment at position 16 of the hydrocarbon chain leads to a lower degree of immobilisation of the spin probe as compared with 5‐DSA. This result suggests that there is a stronger interaction of polar sites of stearic acid with starch granules. © 2000 Society of Chemical Industry  相似文献

The effect of a controlled lactic fermentation on textural and ultrastructural properties of salmon fillets was evaluated. When compared with a non‐inoculated cured sample, fermented salmon had unique textural properties since it exhibited significantly higher force (p < 0.05) and work (p < 0.001) values in a penetration test on two sampling days but a consistently lower hardness (p < 0.05) in a Texture Profile Analysis (TPA) rheological test. Study of the myofibrillar ultrastructure showed that basic structures were drastically damaged during processing and storage of fermented and cured salmon. Nevertheless, Z‐lines were better preserved in fermented samples. It is suggested that the slight protective effect of lactic fermentation with the starter L sake may be the consequence of a lower pH, by putatively inhibiting neutral or alkaline proteolytic enzymes. © 2000 Society of Chemical Industry  相似文献

The synthetic polycyclic musks HHCB and AHTN are potential chemical markers for domestic wastewater contamination of surface waters. Understanding their environmental behavior is important to evaluate their suitability as markers. This study focuses on the quantification of the processes that lead to an elimination in lakes. Rate constants for all relevant processes were estimated based on laboratory studies and models previously described. In lake Zurich, during winter time, both compounds are eliminated primarily by outflowing water and due to losses to the atmosphere. In summer, direct photolysis represents the predominant elimination process for AHTN in the epilimnion of lake Zurich (quantum yield, 0.12), whereas for HHCB, photochemical degradation is still negligible. HHCB and AHTN were then measured in effluents of Swiss wastewater treatment plants (WWTPs), in remote and anthropogenically influenced Swiss surface waters, and in Mediterranean seawater using an analytical procedure based on SPE and GC-MS-SIM with D6-HHCB as internal standard (LODs for natural waters, 2 and 1 ng/L, respectively). In winter, concentrations of HHCB and AHTN in lakes (<2-47 and <1-18 ng/L, respectively) correlated with the anthropogenic burden by domestic wastewater (ratio population per water throughflow), demonstrating the suitability of these compounds as quantitative, source-specific markers. In summer, however, no such correlations were observed. Vertical concentration profiles in lake Zurich indicated significant losses in the epilimnion during summer, mainly for AHTN, and could be rationalized with a lake modeling program (MASASlight), considering measured, average loads from WWTP effluents (0.80 +/- 0.22 and 0.32 +/- 0.11 mg person(-1) d(-1) for HHCB and AHTN, respectively) and the estimated rate constants for elimination processes.  相似文献

The influence of polyesters end-groups on cyclic oligoester formation is investigated using a series of hydroxy-, carboxy- and methylester-terminated aliphatic polyesters, in the presence of various ester interchange catalysts. The presence of hydroxy end-groups is the preponderant factor on cyclodepolymerization kinetics. This indicates that the main reaction is the intramolecular hydroxy–ester interchange reaction between hydroxy end-groups and ester functions in the chain. Carboxy-ester and ester–ester interchanges play a minor role, as the cycle-chain equilibrium is reached only very slowly when carboxy- or ester-terminated polyesters are reacted. High temperature and the presence of tin catalysts are also favorable factors, while, as expected, dilution shifts the equilibrium toward the formation of high yields of cyclic oligoesters. A mechanism is proposed, based on the reverse of the “coordination-insertion” mechanism established for the ring-opening polymerization of lactones.  相似文献

Liver cells are an essential target for drug delivery in many diseases. The hepatocytes express the asialoglycoprotein receptor (ASGPR), which promotes specific uptake by means of N‐acetylgalactosamine (GalNAc) recognition. In this work, we designed two different chemical architectures to treat Wilson's disease by intracellular copper chelation. Two glycoconjugates functionalized with three or four GalNAc units each were shown to enter hepatic cells and chelate copper. Here, we studied two series of compounds derived from these glycoconjugates to find key parameters for the targeting of human hepatocytes. Efficient cellular uptake was demonstrated by flow cytometry using HepG2 human heptic cells that express the human oligomeric ASGPR. Dissociation constants in the nanomolar range showed efficient multivalent interactions with the receptor. Both architectures were therefore concluded to be able to compete with endogeneous asialoglycoproteins and serve as good vehicles for drug delivery in hepatocytes.  相似文献

The illumination of overhead lines by a nuclear electromagnetic pulse (NEMP) in presence of a perfectly conducting ground gives rise to a scattering problem leading to phenomenons both studied here in time and frequency domains. This problem is resolved with the help of an integral equation established in space-time domain and making possible (without approximation) the determination of the time dependance of the current induced on each point of the wire structure. Moreover, the passage in frequency domain allows to express a transfer function defined in one point of the excited line, pointing out its spectral characteristics. Results clearly show the insufficiency of the classic transmission line theory concealing some phenomenons which are described here and for which an interpretation is proposed.  相似文献

Alkaline phosphatase (ALP) is widely used as an indicator of proper pasteurization in bovine milk. Due to interest in the use of equine ALP as a time/temperature integrator (TTI) for evaluation of the efficacy of thermal processing of equine milk, its inactivation kinetics were evaluated in whole and skimmed equine milk. Experimentally determined decimal reduction times showed that equine ALP is more readily inactivated in equine milk than its bovine counterpart. Thus, considering the required 6 D reduction of pathogens and the rather low enzyme level present in equine milk, equine ALP will not be suitable as indicator for correct pasteurization of equine milk under the conditions currently used in the reference method for the determination of ALP in milk-based products.  相似文献

A biofilm model for flowing systems in the food industry   总被引:1,自引:0,他引:1  
When bacteria attach to the walls of pipelines, they can form biofilms, which can cause the recontamination of food products. In order to quantify such recontamination, a one-dimensional biofilm model was developed taking into account adsorption, desorption, and the growth of cells. The model consisted of two mass balances describing increases in biofilm formation at the wall and the accumulation of cells in the liquid phase. The necessary parameters for the model were obtained in laboratory biofilm experiments. These experiments involved a flowing system and the use of Staphylococcus aureus as a model pathogen and silicon tubing as a testing material. S. aureus was inoculated into the system for 2 h, and then the system was changed to a sterile medium. Both biofilm formation and the release of cells into the flowing liquid were measured until steady-state conditions were reached (for up to 9 days). The experiments were performed in duplicate for different flow conditions (i.e., for Reynolds numbers of 3.2, 32, and 170). It was shown that at higher Reynolds numbers, the biofilm developed faster, probably owing to an increase in the transfer of nutrients to the surface. The proposed biofilm model was capable of describing the data obtained for the three different flow conditions with the use of the specific growth rate in the biofilm and the desorption coefficient as fit parameters. The specific growth rates were 0.16, 0.27, and 0.49 h(-1) for Reynolds numbers of 3.2, 32, and 170, respectively, and the desorption coefficients were about 1% of these values.  相似文献
