ABSTRACT

We describe the construction and operation of a cross-correlation phase and modulation fluorometer which uses the harmonic content of a high repetition rate mode-locked laser as the excitation source.

A mode-locked argon ion laser is used to synchronously pump a dye laser. The pulse train output from the dye laser is amplitude modulated by an acousto-optic modulator and then frequency doubled with an angle tuned frequency doubler. With the particular dye utilized in these studies, the ultraviolet light obtained was continuously tunable over the range 280-310 nm. In the frequency domain the high repetition rate pulsed source gives a large series of equally spaced harmonic frequencies. The frequency spacing of the harmonics is determined by the repetition frequency of the laser. Amplitude modulation of the pulse train permits variation of the frequency quasi-continuously from a few hertz to gigahertz. Use of cross-correlation techniques permits precise isolation of individual frequencies. The cross-correlation frequency required for the analysis of the phase delay and modulation ratio is obtained using coupled frequency synthesizers. In the present instrument three synthesizers are used. One drives the pump mode-locker head, a second drives the acousto-optic modulator and the third is used to modulate the response of the photomultiplier tubes which detect the signal. The accuracy, reproducibility and sensitivity of the instrumentation have been determined. Experimental data are provided to show use of this high frequency cross-correlation phase-modulation fluorometer for the determination of fluorescence lifetimes and rotational motions of tryptophan in solution and in proteins.  相似文献

The demilitarization of ammunition that has reached the end of life (or become obsolete) has to be carried out with minimum energy and environmental impacts. The Portuguese Armed Forces have significant amounts of ammunition that need to be eliminated. In order to assess and improve ammunition demilitarization, a life‐cycle approach must be adopted. The main goal of this article is to present a comprehensive life‐cycle assessment (LCA) of the ammunition demilitarization performed by the Portuguese company IDD (Industria de Desmilitarização e Defesa). A life‐cycle model was developed for the entire demilitarization process, which involves ammunition dismantling, discharging, the incineration of energetic material, and the subsequent flue gas treatment. A detailed inventory was based on data collected from the IDD. A life‐cycle impact assessment was carried out, based on three complementary methods used to assess a total of ten impact categories: cumulative energy demand (primary energy); CML 2001 (six environmental impact categories) and USEtox (three toxicological categories). The results show that the main contributor in nine out of the ten impact categories is the incineration and gas treatment process, due to the high energy requirements (electricity and propane). Nevertheless, equipment manufacture also has a significant impact in the Human Toxicity (non‐cancer) category, mainly related to the manufacture of the static kiln. These findings enhance our understanding of demilitarization using a static kiln, showing that the associated impacts are significant and should be reduced.  相似文献

In the present paper, the extraction of passion fruit oil with acetone, ethanol and isopropanol of passion fruit seeds under three different techniques (ultrasound, shaker and soxhlet) were studied. The influence of seed to solvent ratio, time and type of solvent were investigated during ultrasound, shaker and soxhlet process. Hexane was used as well, as an extraction solvent to evaluate the extraction of oil in all extraction process studied to make a comparison among the employed solvents. The temperature was kept isoterminally at 40 °C in both shaker and ultrasound process. The highest yield when green solvents were was 23.8%, during ultrasound process, with acetone was used. Our results indicate that ultrasound can greatly replace the conventional extraction.  相似文献

Poly(ethylene‐co‐vinyl acetate) (EVA) is the primary polymer used for encapsulation of photovoltaic (PV) modules. Its degree of cross‐linking (gel content) is taken as a major quality reference. The EVA gel content is normally measured by Soxhlet extraction and more recently also by Differential Scanning Calorimetry (DSC). The DSC method is proven here to be fast and effective but is, as the Soxhlet extraction method, destructive to the PV module. With the aim of developing a fast and non‐destructive method to determine the gel content, a number of analytical techniques are presented. The most promising method is ultraviolet/visible/near‐infrared (UV/Vis/NIR) optical transmission. The measured diffuse transmission reflects the EVA crystallite size, which is related to the EVA gel content. This opens the possibility to apply an in‐line analysis of every PV module immediately after the lamination step and could significantly contribute to the process quality control that is needed in future high‐throughput production lines of PV modules. This work focused on the study of the impact event on molded parts in the framework of automotive components. The influence of the impact conditions and processing parameters on the mechanical behavior of talc‐filled polypropylene specimens was analyzed. The specimens were lateral‐gate discs produced by injection molding, and the mechanical characterization was performed through instrumented falling weight impact tests concomitantly assisted with high‐speed videography. Results analyzed using the analysis of variance (ANOVA) method have shown that from the considered parameters, only the dart diameter and test temperature have significant influence on the falling weight impact properties. Higher dart diameter leads to higher peak force and peak energy results. Conversely, higher levels of test temperatures lead to lower values of peak force and peak energy. By means of high‐speed videography, a more brittle fracture was observed for experiments with higher levels of test velocity and dart diameter and lower levels of test temperature. The injection‐molding process conditions assessed in this study have an influence on the impact response of moldings, mainly on the deformation capabilities of the moldings. POLYM. ENG. Ethyl carbamate is an impurity present in distilled beverages. Given the risk of it being a carcinogenic substance, Brazilian legislation has determined that its presence in distilled beverages, such as ‘aguardente’ and ‘cachaça’ (two types of sugarcane spirits), should be limited to a maximum of 150 µg/L. Ordinary spirits usually contain variable amounts of ethyl carbamate, although in lower concentrations than the maximum determined by law. The finding that commercial spirits had a much lower concentration of this impurity (around 50 µg/L) led the authors to research the reasons for the differences, and these are explored in this paper, with a focus on the speed of the distillation process and its influence on the spirit's composition. The team conducted research in a sugarcane distillery producing ‘aguardente’ using a simple pot still and measured the influence of fast and slow distillation on the presence of ethyl carbamate and non‐alcohol components in the process. The results demonstrated that the speed of distillation was proportionally related to the concentration of ethyl carbamate and secondary components in the beverage's composition. The aim of this study was to assess the food hygiene knowledge of professional food handlers from an institutional catering company which manufactures and distributes meals to the canteens of schools, kindergartens and nursing homes in Portugal. A total of 101 food handlers from 18 geographically distributed business units were assessed. Data collection employed a previously used, multiple-choice questionnaire, aimed at exploring the food safety knowledge and practices of individual respondents. The average score of questions answered correctly was 13 out of 23 (56.5%) with a standard deviation (SD) of 3.22. However, the percentage of correct answers varies with the issues questioned, being significantly lower on issues such as temperature control (p < 0.001) and sources of contamination/high-risk foods (p < 0.001). The level of knowledge was influenced by the level of formal education (p = 0.025) of respondents. The results reinforce the importance of conducting a preliminary assessment of training needs and evaluating the effectiveness of training.  相似文献

Wireless sensor networks have been more and more deployed for monitoring and controlling real environments. However, because of sensor nodes?? limitations, their radios are more susceptible to noise and interference than in other wireless technologies. These are critical factors in the rigorous industrial environment, whose effects may not be the same on all channels of a sensor network. In this paper, we present some experimental studies about the characteristics of IEEE 802.15.4 channels in ISM band. We are particularly interested in the differences of implementing wireless sensor networks in academic and industrial environments. A common problem of wireless sensor network implementations is that most of them are purely academic or initially prepared in laboratory. Our study shows that it is very difficult to predict the quality and stability of the different channels in sensor networks, as the environment conditions greatly influence the network performance. Empirical approaches are suitable methods for choosing the appropriate channels to deploy WSNs in noisy environments. We conduct several tests in both laboratory and in a real world environment, particularly in an oil refinery as this is our case study in the context of the FP7 GINSENG project (Ginseng 2010).  相似文献

A new method for the investigation of the inhibition efficiency against corrosion by organic films adsorbed on metals based on the measurement of Z-approach curves by AC-SECM is proposed. Preliminary measurements conducted on four copper-inhibitor systems exposed to aqueous solutions support that a characteristic frequency can be defined, which corresponds to the frequency of the AC potential signal applied to the SECM tip for which a transition between negative- and positive-feedback behaviours is observed in the approach curves. From the shift of this characteristic frequency towards higher values, the enhancement of the corrosion protection efficiency of the inhibitor system can be established. Furthermore, the effects of inhibitor nature, concentration, and pre-treatment duration for the formation of the surface films, can be readily investigated.  相似文献
