Multiple sclerosis (MS) is a complex autoimmune disease of the central nervous system characterized by chronic inflammation, demyelination, and axonal damage. As microRNA (miRNA)-dependent alterations in gene expression in hematopoietic cells are critical for mounting an appropriate immune response, miRNA deregulation may result in defects in immune tolerance. In this frame, we sought to explore the possible involvement of miRNAs in MS pathogenesis by monitoring the differential expression of 22 immunity-related miRNAs in peripheral blood mononuclear cells of MS patients and healthy controls, by using a microbead-based technology. Three miRNAs resulted >2 folds up-regulated in MS vs controls, whereas none resulted down-regulated. Interestingly, the most up-regulated miRNA (mir-155; fold change = 3.30; P = 0.013) was previously reported to be up-regulated also in MS brain lesions. Mir-155 up-regulation was confirmed by qPCR experiments. The role of mir-155 in MS susceptibility was also investigated by genotyping four single nucleotide polymorphisms (SNPs) mapping in the mir-155 genomic region. A haplotype of three SNPs, corresponding to a 12-kb region encompassing the last exon of BIC (the B-cell Integration Cluster non-coding RNA, from which mir-155 is processed), resulted associated with the disease status (P = 0.035; OR = 1.36, 95% CI = 1.05-1.77), suggesting that this locus strongly deserves further investigations.  相似文献

Interpretation of structural properties and dynamic behavior of molecules in solution is of fundamental importance to understand their stability, chemical reactivity, and catalytic action. While information can be gained, in principle, by a variety of spectroscopic techniques, the interpretation of the rich indirect information that can be inferred from the analysis of experimental spectra is seldom straightforward because of the subtle interplay of several different effects, whose specific role is not easy to separate and evaluate. In such a complex scenario, theoretical studies can be very helpful at two different levels: (i) supporting and complementing experimental results to determine the structure of the target molecule starting from its spectral properties; (ii) dissecting and evaluating the role of different effects in determining the observed spectroscopic properties. This is the reason why computational spectroscopy is rapidly evolving from a highly specialized research field into a versatile and widespread tool for the assignment of experimental spectra and their interpretation in terms of chemical physical effects. In such a situation, it becomes important that both computationally and experimentally oriented chemists are aware that new methodological advances and integrated computational strategies are available, providing reliable estimates of fundamental spectral parameters not only for relatively small molecules in the gas phase but also for large and flexible molecules in condensed phases. In this Account, we review the most significant methodological contributions from our research group in this field, and by exploiting some recent results of their application to the computation of IR, UV-vis, NMR, and EPR spectral parameters, we discuss the microscopic mechanisms underlying solvent and vibrational effects on the spectral parameters. After reporting some recent achievements for the study of excited states by first principle quantum mechanical approaches, we focus on the treatment of environmental effects by means of mixed discrete-continuum solvent models and on effective methods for computing vibronic contributions to the spectra. We then discuss some new developments, mainly based on time-dependent approaches, allowing us to go beyond the determination of spectroscopic parameters toward the simulation of line widths and shapes. Although further developments are surely needed to improve the accuracy and effectiveness of several items in the proposed approach, we try to show that the first important steps toward a direct comparison between the results obtained in vitro and those obtained in silico have been made, making easier fruitful crossovers among experiments, computations and theoretical models, which would be decisive for a deeper understanding of the spectral behavior associated with complex systems and processes.  相似文献

Engineering levels of porosity in hierarchical zeolites is a vibrant area of research with remarkable application potential. To gain practical relevance, the superior properties observed for the as‐synthesized powders have to be preserved when they are shaped into suitable technical geometries. Herein, mechanically stable millimeter‐sized bodies are prepared by granulation of mesoporous ZSM‐5 zeolite powders using an attapulgite clay binder. Alkaline treatment of conventional zeolite granules is demonstrated to be unsuitable for this purpose. Multiple techniques are applied to characterize mesoporous zeolite granules with respect to their conventional zeolite counterparts, thus establishing the impact of binder inclusion and granulation on their respective properties. The intrinsic structure and acidity of the zeolite are retained post‐structuring. Gas adsorption and mercury porosimetry confirm the presence of interconnected micro‐, meso‐, and macropores. A wide range of imaging techniques permits visualization of the particle properties, phase distribution, and consequent origins of the distinct levels of porosity within the zeolite granules. The superior adsorption properties of the hierarchical ZSM‐5 zeolite granules are demonstrated using cyclohexane, toluene, and isopropyl alcohol as probe molecules.  相似文献

In a gene expression data matrix, a bicluster is a submatrix of genes and conditions that exhibits a high correlation of expression activity across both rows and columns. The problem of locating the most significant bicluster has been shown to be NP-complete. Heuristic approaches such as Cheng and Church's greedy node deletion algorithm have been previously employed. It is to be expected that stochastic search techniques such as evolutionary algorithms or simulated annealing might improve upon such greedy techniques. In this paper we show that an approach based on simulated annealing is well suited to this problem, and we present a comparative evaluation of simulated annealing and node deletion on a variety of datasets. We show that simulated annealing discovers more significant biclusters in many cases. Furthermore, we also test the ability of our technique to locate biologically verifiable biclusters within an annotated set of genes.  相似文献

This paper revisits the theory of operation of field effect transistor in the extremely high frequency scale, where the analysis has gone beyond the conventional cutoff frequency of the transistor. In this range, which is typically the terahertz (THz) and sub-terahertz range, the transistor blocks the high frequency signal and generates a rectified signal related to the input high frequency signal. An analytical model is derived for the channel of the FET in the linear mode of operation in non-resonant THz detection conditions. A transmission line distributed circuit model is applied. This is, from the authors’ point of view, the suitable model for high frequency non-quasi static operation and the characteristic parameters of this model are derived from the differential equation governing the electron gas in the channel. A comparison is presented for the calculated photoresponse with previously published experimental one showing good agreement away from the threshold potential. Finally, the effects of coupling between the present model and the external input circuit have been taken into account including the loading effects of the antenna and a discussion is given for the effect on frequency selectivity of the FET.  相似文献

We present an interpolation algorithm for adaptive color image zooming. The algorithm produces the magnified image in one scan of the input image, and is fully automatic since does not involve any a priori fixed threshold. Given any integer zooming factor n, each pixel of the input image generates an n×n block of pixels in the zoomed image. For the currently visited pixel of the input image, the pixels of its associated block are first assigned tentative values, which are then adaptively updated before building the next block. The method is suggested for RGB images, but can equally be employed in other color spaces. Peak signal to noise ratio (PSNR) and Structural SIMilarity (SSIM) are used to evaluate the performance of the algorithm.  相似文献

This article presents a set of accurate closed-form formulas for the electromagnetic parameters (inductance (L), capacitance (C) and characteristic impedance (Zc)) for squared coaxial lines with circular and square inner conductor. The analytical expressions, deduced from rigorous analysis by the finite element method (FEM), method of moments (MoM) and curves fitting techniques, can be easily implemented in CAD simulation tools, to design components for wireless communication. This study presents accurate and suitable general expressions for all squared coaxial lines with a wide range of outer to inner conductors ratio between 1.2 and 10. As an application, we present the design of 60 GHz branch line couplers.  相似文献

Mobility models of nodes have an important role in the evaluation of data dissemination protocols in vehicular ad hoc networks (VANETs). Many recent researches have used the constant velocity mobility models while vehicles have acceleration and their speeds change as they move. Because of the dynamic nature of VANETs, the network changes from a densely connected to the sparsely connected environment in a short time. In sparsely connected networks, it is said that vehicles move in clusters. In this paper, the constant acceleration mobility calculations for the sparsely connected network and its characteristics have been presented. These characteristics are usable for the evaluation of the data dissemination protocols in VANETs. The results show that acceleration affects the number of viewed clusters during the trip and the number of vehicles within a cluster. In fact, acceleration has a significant impact on the network sparsely connection. This matter shows that the appropriate data dissemination protocol should be used to study the acceleration effect which is applicable in sparse networks. The analysis in this research provides the necessary background for better understanding and accurate calculations for the evaluation of data dissemination in VANETs.  相似文献

This paper presents the concept of intra-, inter- and multiple-domain service level negotiation using thecops-sls protocol.
