As wind turbines continue to grow in size, it becomes increasingly important to ensure that they are as structurally efficient as possible to ensure that wind energy can be a cost‐effective source of power generation. A way to achieve this is through weight reductions in the blades of the wind turbine. In this study, topology optimization is used to find alternative structural configurations for a 45 m blade from a 3 MW wind turbine. The result of the topology optimization is a layout that varies along the blade length, transitioning from a structure with trailing edge reinforcement to one with offset spar caps. Sizing optimization was then performed on a section with the trailing edge reinforcement and was shown to offer potential weight savings of 13.8% when compared with a more conventional design. These findings indicate that the conventional structural layout of a wind turbine blade is sub‐optimal under the static load conditions that were applied, suggesting an opportunity to reduce blade weight and cost. New drugs are urgently needed for the treatment of tropical parasitic diseases such as leishmaniasis and human African trypanosomiasis (HAT). This work involved a high-throughput screen of a focussed kinase set of ~3400 compounds to identify potent and parasite-selective inhibitors of an enzymatic Leishmania CRK3-cyclin 6 complex. The aim of this study is to provide chemical validation that Leishmania CRK3-CYC6 is a drug target. Eight hit series were identified, of which four were followed up. The optimisation of these series using classical SAR studies afforded low-nanomolar CRK3 inhibitors with significant selectivity over the closely related human cyclin dependent kinase CDK2.  相似文献

This paper investigates the effect of heat treatment upon the corrosion morphology and mechanism of ZE41 alloy. The results of optical and scanning electron microscopy (SEM) together with potentiodynamic polarisation reveal the importance of the microstructure in the initiation and propagation of corrosion in an aqueous environment. The corrosion of the heat-treated alloy is significantly altered due to changes in the microstructure, specifically the Zr-rich regions and the grain boundary T-phase.  相似文献

Chemical recognition systems are crucial for maintaining the unity of social insect colonies. It has been proposed that colonies form a common chemical signature, called the gestalt odor, which is used to distinguish colony members and non-members. This chemical integration is achieved actively through social interactions such as trophallaxis and allogrooming, or passively such as through exposure to common nest material. When colonies are infiltrated by social parasites, the intruders often use some form of chemical mimicry. However, it is not always clear how this chemical mimicry is accomplished. Here, we used a three-species nesting symbiosis to test the differences in chemical integration of mutualistic (parabiotic) and parasitic ant species. We found that the parasite (Solenopsis picea) obtains chemical cues from both of the two parabiotic host ant species. However, the two parabiotic species (Crematogaster levior and Camponotus femoratus) maintain species-specific cues, and do not acquire compounds from the other species. Our findings suggest that there is a fundamental difference in how social mutualists and social parasites use chemicals to integrate themselves into colonies.  相似文献

The diagnosis of ocular disease is increasingly important in optometric practice and there is a need for cost effective point of care assays to assist in that. Although tears are a potentially valuable source of diagnostic information difficulties associated with sample collection and limited sample size together with sample storage and transport have proved major limitations. Progressive developments in electronics and fibre optics together with innovation in sensing technology mean that the construction of inexpensive point of care fibre optic sensing devices is now possible. Tear electrolytes are an obvious family of target analytes, not least to complement the availability of devices that make the routine measurement of tear osmolarity possible in the clinic. In this paper we describe the design, fabrication and calibration of a fibre-optic based electrolyte sensor for the quantification of potassium in tears using the ex vivo contact lens as the sample source. The technology is generic and the same principles can be used in the development of calcium and magnesium sensors. An important objective of this sensor technology development is to provide information at the point of routine optometric examination, which would provide supportive evidence of tear abnormality.  相似文献

Excessive vibration, such as chatter, is a common problem in machining processes. Meanwhile, numerous hard, brittle metals have been shown to form segmented chips, also known as sawtooth chips, during machining. In the literature, a cyclic cutting force has been demonstrated where segmented chips are formed, with the force cycle corresponding to the formation of segments. Segmented chip formation has been shown to be linked to high vibration levels in turning and milling processes. Additionally, it has been proposed that the amplitude of chatter vibrations can be limited by interference between the tool flank and wavy workpiece surface, a phenomenon known as tool-flank process damping. In this contribution, a model is proposed to predict the amplitude of forced vibration arising due to the formation of segmented chips during turning. The amplitude of vibration was calculated as a function of cutting parameters. It was demonstrated that the model can be extended to account for the effect of tool-flank process damping. For validation, titanium Ti6Al4V alloy was turned using a flexible toolholder, with surface speed ranging from 10 to 160 m/min, feed rate from 0.1 to 0.7 mm/rev and width of cut from 0.35 to 4 mm. In the experimental validation, 25 of 68 test cuts exhibited high-amplitude vibration. In 16 of these cases, the main cause was concluded to be chip segmentation, which can be predicted by the model. The model is thus considered of practical value to machinists.  相似文献

The effects of three different extraction methods, namely hydrodistillation, supercritical CO2 extraction (SCE) and hexane extraction on the yield, chemical composition and antimicrobial and antioxidant activities of lavender essential oil were investigated in this study. SCE produced a yield of 6.7 % (dry weight), which was comparable to that of solvent extraction (7.6 %), but significantly higher than that of hydrodistillation (4.6 %). The chemical composition of the oils showed considerable variations among the extraction methods, with linalool, linalyl acetate, camphor and borneol making up approximately 80 % of identified components in all extracts. Hexane extraction produced oils with the presence of waxes, colour pigments and albuminious materials with semi-solid consistency, while hydrodistillation extracts showed signs of thermal degradation. The SCE extracts had an aroma with the closest resemblance to the starting material, showing negligible thermal degradation, and exhibited significantly higher antioxidant activity than the hydrodistillation and hexane extracts. Oils produced by SCE and hydrodistillation had antimicrobial activities higher than hexane extracts. The results of this study demonstrate that SCE is a promising process for the extraction of lavender essential oil.  相似文献

Due to increasing customer and political pressures, and more stringent environmental regulations, sediment and other blockage issues are now a high priority when assessing sewer system operational performance. Blockages caused by sediment deposits reduce sewer system reliability and demand remedial action at considerable operational cost. Consequently, procedures are required for identifying which parts of the sewer system are in most need of proactive removal of sediments. This paper presents an exceptionally long (7.5 years) and spatially detailed (9658 grid squares - 0.03 km2 each - covering a population of nearly 7.5 million) data set obtained from a customer complaints database in Bogotá (Colombia). The sediment-related blockage data are modelled using homogeneous and non-homogeneous Poisson process models. In most of the analysed areas the inter-arrival time between blockages can be represented by the homogeneous process, but there are a considerable number of areas (up to 34%) for which there is strong evidence of non-stationarity. In most of these cases, the mean blockage rate increases over time, signifying a continual deterioration of the system despite repairs, this being particularly marked for pipe and gully pot related blockages. The physical properties of the system (mean pipe slope, diameter and pipe length) have a clear but weak influence on observed blockage rates. The Bogotá case study illustrates the potential value of customer complaints databases and formal analysis frameworks for proactive sewerage maintenance scheduling in large cities.  相似文献

To probe the mechanism of the photosensitized loss of phenols by humic substances (HS), the dependence of the initial rate of 2,4,6-trimethylphenol (TMP) loss (R(TMP)) on dioxygen concentration was examined both for a variety of untreated as well as borohydride-reduced HS and C(18) extracts from the Delaware Bay and Mid-Atlantic Bight. R(TMP) was inversely proportional to dioxygen concentration at [O(2)] > 50 μM, a dependence consistent with reaction with triplet excited states, but not with (1)O(2) or RO(2). Modeling the dependence of R(TMP) on [O(2)] provided rate constants for TMP reaction, O(2) quenching, and lifetimes compatible with a triplet intermediate. Borohydride reduction significantly reduced TMP loss, supporting the role of aromatic ketone triplets in this process. However, for most samples, the incomplete loss of sensitization following borohydride reduction, as well as the inverse dependence of R(TMP) on [O(2)] for these samples, suggests that there remains another class of oxidizing triplet sensitizer, perhaps quinones.  相似文献

South-central Arkansas (AR) is home to major manufacturing facilities for brominated flame retardant chemicals (BFRs) in the U.S. Unintended release during production may have caused accumulation of the BFRs in the local environment. In this work, sediment cores were collected from six water bodies in AR, including three located close to the BFR manufacturing facilities in El Dorado and Magnolia, to investigate past and recent deposition histories. A total of 49 polybromodiphenyl ethers (PBDEs) and decabromodiphenyl ethane (DBDPE) were detected, with concentrations as high as 57?000 and 2400 ng/g dry weight for decabromodiphenyl ether (BDE209) and DBDPE, respectively. Log-log regression of BDE209 and DBDPE surface concentrations versus distance to known BFR manufacturing facilities fit the Gaussian Plume Dispersion model, and showed that, if the distance is shortened by half, concentrations of BDE209 and DBDPE would increase by 5-fold. The spatial distribution and temporal trend of the contamination indicate that the manufacturing of PBDEs and DBDPE is the primary source for these compounds in the environment of southern Arkansas. Interestingly, the occurrence of debromination of PBDEs in the sediments of a previously used wastewater sludge retention pond in Magnolia is indicated by the presence of congeners that had not been detected in any commercial PBDE mixtures and by increased fractions of lower brominated congeners relative to higher brominated congeners. Two unknown brominated compounds were detected in the sediments, and identified as nonabromodiphenyl ethanes.  相似文献
