Background
The exposure to pollutants such as diesel exhaust particles (DEP) is associated with an increased incidence of respiratory diseases. However, the mechanisms by which DEP have an effect on human health are not completely understood. In addition to their action on macrophages and airway epithelial cells, DEP also modulate the functions of dendritic cells (DC). These professional antigen-presenting cells are able to discriminate unmodified self from non-self thanks to pattern recognition receptors such as the Toll like Receptors (TLR) and Scavenger Receptors (SR). SR were originally identified by their ability to bind and internalize modified lipoproteins and microorganisms but also particles and TLR agonists. In this study, we assessed the implication of SR in the effects of DEP associated or not with TLR agonists on monocyte-derived DC (MDDC). For this, we studied the regulation of CD36, CXCL16, LOX-1, SR-A1 and SR-B1 expression on MDDC treated with DEP associated or not with TLR2, 3 and 4 ligands. Then, the capacity of SR ligands (dextran sulfate and maleylated-ovalbumin) to block the effects of DEP on the function of lipopolysaccharide (LPS)-activated DC has been evaluated. 相似文献42.

The isoquinoline alkaloid berberine, present in nine different plant families was found to be phototoxic to mosquito larvae. In the presence of near UV the LC50 for acute 24-hr toxicity was 8.8 ppm compared to 250 ppm for dark controls. Mosquito larvae that were treated with 10 ppm berberine plus near UV for 24 hr and then transferred to berberine-free water showed decreased larval survival and resulted in a smaller cumulative number of pupae and adults as compared to controls, during a subsequent 4-week development period. Berberine was found to be a singlet O2 generator in experiments with the chemical trap 2,5-dimethyl furan. A slight increase in chromosome aberrations in Chinese hamster cells was also observed with berberine plus near UV treatment. The significance of the phototoxicity of berberine is discussed in relation to plant-insect relations. 相似文献

The allocation of water resources between different users is a hard task for water managers because they must deal with conflicting objectives. The main objective is to obtain the most accurate distribution of the resource and the associated circulating flows through the system. This induces the need for a river basin optimization model that provides optimized results. This article presents a network flow optimization model to solve the water allocation problem in water resource systems. Managing a water system consists in providing water in the right proportion, at the right place and at the right time. Time expanded network allows to take into consideration the temporal dimension in the decision making. Since linear cost functions on arcs present many limitations and are not realistic, quadratic convex cost functions on arcs are considered here. The optimization algorithm developed herein extend the cycle canceling algorithm developed for linear cost functions. The methodology is applied to manage the three reservoirs of La Haute-Vilaine’s watershed located in the north west of France to protect a three vulnerable areas from flooding. The results obtained with the algorithm are compared to a reference scenario which consists in considering reservoirs transparent. The results show that the algorithm succeeds in managing the reservoir releases efficiently and keeps the flow rates below the vigilance flow in the vulnerable areas. 相似文献

An ultra-performance liquid chromatography (UPLC®) method has been developed for the simultaneous determination of deoxynivalenol (DON) and nivalenol (NIV) in wheat. Ground sample was extracted with water and the filtered extract was cleaned up through an immunoaffinity column containing a monoclonal antibody specific for DON and NIV. Toxins were separated and quantified by UPLC® with photodiode-array detector (λ?=?220 nm) in less than 3 min. Mean recoveries from blank wheat samples spiked with DON and NIV at levels of 100–2,000 μg/kg (each toxin) ranged from 85 to 95 % for DON and from 81 to 88 % for NIV, with relative standard deviations less than 7 %. Similar recoveries were observed from spiked samples when methanol/water (80:20, v/v) was used as extraction solvent. However, by using a wheat sample naturally contaminated with DON and NIV, the one-way analysis of variance (Student–Newman–Keuls test) between different extraction solvents and modes showed that water extraction provided a significant increase (P?<?0.001) in toxin concentrations (mean values of six replicate analyses) with respect to methanol/water (80:20, v/v). No significant difference was observed between shaking (60 min) and blending (3 min). The limit of detection (LOD) of the method was 30 μg/kg for DON and 20 μg/kg for NIV (signal-to-noise ratio 3:1). The immunoaffinity columns showed saturation of DON/NIV binding sites at levels higher than 2,000 ng in blank wheat extracts spiked with the corresponding amount of mycotoxin, as single mycotoxin or sum of DON and NIV. The range of applicability of the method was from LOD to 4,000 μg/kg, as single mycotoxin or sum of DON and NIV in wheat. The analyses of 20 naturally contaminated wheat samples showed DON contamination in all analyzed samples at level ranging from 30 to 2,700 μg/kg. NIV was detected in two samples at negligible toxin levels (up to 46 μg/kg). In this study, skim milk (9.5% w/v solid content) was supplemented with 1-3% (w/v) lentil flour or skim milk powder, inoculated with a yogurt culture, fermented and stored at 4 °C. Acid production during the fermentation, microbial growth, physical properties (pH, syneresis, and color), rheological properties (dynamic oscillation temperature sweep test at 4-50 °C), during 28 days of refrigerated storage and also sensory properties (flavor, mouth feel, overall acceptance and color) after production, were studied. Milk supplementation with 1-3% lentil flour enhanced acid production during fermentation, but the microbial population (CFU) of both S. thermophilus and L. delbrueckii subsp. bulgaricus were in the same range in all lentil flour and skim milk powder supplemented yogurts. The average pH of samples decreased from 4.5 to 4.1 after 28 days storage. Syneresis in 1-2% lentil flour supplemented yogurts was significantly higher than all other samples; however, greater lentil supplementation (3%) resulted in the lowest syneresis during the 28 days storage. With respect to color, “a” and “L” values did not significantly differ in all samples and remained constant after 28 days whereas “b” value increased as a result of lentil supplementation. Yogurt with 3% lentil flour showed higher storage (G') and loss (G?) moduli in comparison with samples supplemented with 1-3% skim milk powder and the non-supplemented control yogurt. Storage modulus (G') was higher than loss modulus (G?) in all samples and at all temperatures between 4 and 50 °C and they showed a hysteresis loop over this temperature range when the samples were heated and cooled. 1-2% lentil flour supplemented yogurt showed comparable sensory properties in comparison with 1-2% skim milk powder supplemented yogurt and the control sample. 相似文献

The impact of induced lipolysis on the composition of plasma lipids is analyzed by proton magnetic resonance spectroscopy (MRS) in humans. The variations of the methylene and methyl resonances from lipids in lipoproteins are studied under two sets of lipolytic conditions: acute endovascular lipolysis induced by an intravenous injection of heparin and subacute lipolysis induced by short fasting. During acute lipolysis, the degradation of the very low density lipoproteins structures is well correlated to the modifications observed in the areas of CH2 and CH3 MRS signals. The comparison of regular spectra, spectra with water signal suppression, and spectra recorded with a spin-echo sequence provides information on the behavior of the different parts of the lipoproteins, that is, the neutral core, little affected by heparine-induced lipoprotein lipase (LPL) activation, and the surface layer supplying substrates to LPL. Due to the exponential increase in aircraft traffic in recent decades, the role of soot particles emitted by aircraft engines on the radiative forcing needs to be addressed, and especially their interaction with clouds has to be better understood and quantified. In this work, we investigate the hygroscopic properties of fresh and aged soot sampled on line in a kerosene flame. The activated fraction (Fa) of size selected soot is measured by means of a variable supersaturation condensation nucleus counter at several heights above the burner (HAB), thereby probing soot particles with different residence times in the flame, i.e., different degrees of maturity. In order to simulate atmospheric aging, the activity of soot as cloud condensation nuclei is measured as a function of ozone exposure. We show that fresh soot is hydrophobic (Fa~0), while Fa increases when soot is exposed to ozone. The measurements depend on the HAB at which soot particles are sampled showing that activation of soot particles is related to their chemical composition. This study brings new results on the link between atmospheric aging of soot and its hygroscopic properties, which is of great interest for understanding the role of soot in the cloud formation.
