Self-controlled case series analyses: Small-sample performance

Second-order expressions for the asymptotic bias and variance of the log relative incidence estimator are derived for the self-controlled case series model in a simplified scenario. The dependence of the bias and variance on factors such as the relative incidence and ratio of risk to observation period are studied. Small-sample performance of the estimator in realistic scenarios is investigated using simulations. It is found that, in scenarios likely to arise in practice, asymptotic methods are valid for numbers of cases in excess of 20-50 depending on the ratio of the risk period to the observation period and on the relative incidence. The application of Monte Carlo methods to self-controlled case series analyses is also discussed.     

Isolation and DNA sequence of the STE14 gene encoding farnesyl cysteine: Carboxyl methyltransferase

We isolated a mutant defective in C-terminal farnesyl cysteine:carboxyl methyltransferase activity from a screen for mutations causing a -specific sterility. A genomic fragment was cloned from a yeast multi-copy library that restored mating. Both the cloned gene and the sterile mutation were allelic to the STE14 gene. A ste14-complementing 2·17 kb BamHI fragment subclone was sequenced and found to encode a 239 amino acid protein with a molecular weight of 27,887 Daltons. The hydrophobicity profile of the methyltransferase reveals the presence of at least five potential transmembrane domains. In comparisons of the C-terminal methyltransferase amino acid sequence with those in the PIR and Swiss protein databases, no significantly similar sequences were found nor were conserved regions from other methyltransferases present.     

The effect of random vaccine response on the vaccination coverage required to prevent epidemics

The response people have to vaccination varies because their immune systems differ and vaccine failures occur. Here we consider the effect that a random response, independent for each vaccinee, has on the vaccination coverage required to prevent epidemics in a large community. For a community of uniformly mixing individuals an explicit expression is found for the critical vaccination coverage (CVC) and the effect of the vaccine response is determined entirely by the mean E(AB), where A and B, respectively, reflect the infectivity and susceptibility of a vaccinated individual. This result shows that the usual concept of vaccine efficacy, which focuses on the amount of protection the vaccine provides the vaccinee against infection, is not adequate to describe the requirements for preventing epidemics when vaccination affect infectivity. The estimation of E(AB) poses a problem because A and B refer to the vaccine response of the same individual. Similar results are found when there are different types of individual, but now the mean E(AB) may differ between types. However, for a community made up of households it is shown that the CVC also depends on other characteristics of the vaccine response distribution. In practice this means that estimating a single measure of vaccine effectiveness is generally not enough to determine the CVC. For a specific community of households it is found that the vaccination coverage required to prevent epidemics decreases as the variation in the vaccine response increases.     

Designing the MPC105 PCI bridge/memory controller

The MPC105 peripheral component interconnection bridge/memory controller provides a platform-specification-compliant bridge between Power PC microprocessors and the PCI bus. With it, designers can create systems using peripherals already designed for a variety of standard PC interfaces. This bridge chip also integrates a secondary cache controller and high-performance memory controller that supports DRAM or synchronous DRAM and ROM or flash ROM     

Hepatic damage produced by long-term alcohol consumption in well-nourished rats

A study of the effect of long-term alcohol consumption on the liver of well-nourished rats is described. Rats fed for 16 weeks on a semipurified diet supplemented with high levels of vitamins and lipotropic factors and alcohol corresponding to 35% of the total caloric intake developed marked fatty changes of the liver. Mild fatty changes were observed in pair-fed controls receiving as isoenergetic equivalent of sucrose instead of alcohol. Intracellular hyaline bodies, corresponding ultrastructurally to giant mitochondria were abundantly found in the hepatocytes of alcoholic rats, while in the controls they were not seen. Te findings in this investigation are postulated to provide further evidence that the long-term intake of alcohol exerts a direct causative role in the pathogenesis of liver damage.
