Measurement of cell volume in living epithelial cells has become an important technique in studies of membrane transport processes that function in cell volume regulation. Planimetry of video images of optical sections enables the measurement of the cross sectional area of each section. Cell volume is calculated from the measured area of each section and the known focus displacements. In the past the measurement of cross section area has been done by manual positioning of a cursor superimposed on the video image. Each experiment generates approximately 200 images in which two or more cells may be analysed. We have developed a computer-based method that uses one image as a template, and allows automated area determination of successive images by template matching and digital image processing. This new method is comparable to the older method in speed and accuracy, but requires much less effort from the experimenter.  相似文献

The purpose of this study was to evaluate the association between four distinct histopathological features: (1) tumor infiltrating lymphocytes, (2) mucinous differentiation, (3) tumor-stroma ratio, plus (4) tumor budding and two gene expression-based classifiers—(1) consensus molecular subtypes (CMS) plus (2) colorectal cancer intrinsic subtypes (CRIS). All four histopathological features were retrospectively scored on hematoxylin and eosin sections of the most invasive part of the primary tumor in 218 stage II and III colon cancer patients from two independent cohorts (AMC-AJCC-90 and AC-ICAM). RNA-based CMS and CRIS assignments were independently obtained for all patients. Contingency tables were constructed and a χ2 test was used to test for statistical significance. Odds ratios with 95% confidence intervals were calculated. The presence of tumor infiltrating lymphocytes and a mucinous phenotype (>50% mucinous surface area) were strongly correlated with CMS1 (p < 0.001 and p = 0.008) and CRIS-A (p = 0.006 and p < 0.001). The presence of mucus (≥ 10%) was associated with CMS3: mucus was present in 64.1% of all CMS3 tumors (p < 0.001). Although a clear association between tumor-stroma ratio and CMS4 was established in this study (p = 0.006), still 32 out of 61 (52.5%) CMS4 tumors were scored as stroma-low, indicating that CMS4 tumors cannot be identified solely based on stromal content. Higher budding counts were seen in CMS4 and CRIS-B tumors (p = 0.045 and p = 0.046). No other associations of the measured parameters were seen for any of the other CRIS subtypes. Our analysis revealed clear associations between histopathologic features and CMS or CRIS subtypes. However, identification of distinct molecular subtypes solely based on histopathology proved to be infeasible. Combining both molecular and morphologic features could potentially improve patient stratification.  相似文献

In addition to their therapeutic potential in regenerative medicine, human corneal stromal stem cells (CSSCs) could serve as a powerful tool for drug discovery and development. Variations from different donors, their isolation method, and their limited life span in culture hinder the utility of primary human CSSCs. To address these limitations, this study aims to establish and characterize immortalized CSSC lines (imCSSC) generated from primary human CSSCs. Primary CSSCs (pCSSC), isolated from human adult corneoscleral tissue, were transduced with ectopic expression of hTERT, c-MYC, or the large T antigen of the Simian virus 40 (SV40T) to generate imCSSC. Cellular morphology, proliferation capacity, and expression of CSSCs specific surface markers were investigated in all cell lines, including TNFAIP6 gene expression levels in vitro, a known biomarker of in vivo anti-inflammatory efficacy. SV40T-overexpressing imCSSC successfully extended the lifespan of pCSSC while retaining a similar morphology, proliferative capacity, multilineage differentiation potential, and anti-inflammatory properties. The current study serves as a proof-of-concept that immortalization of CSSCs could enable a large-scale source of CSSC for use in regenerative medicine.  相似文献

Partial order techniques enable reducing the size of the state space used for model checking, thus alleviating the “state space explosion” problem. These reductions are based on selecting a subset of the enabled operations from each program state. So far, these methods have been studied, implemented, and demonstrated for assertional languages that model the executions of a program as computation sequences, in particular the linear temporal logic. The present paper shows, for the first time, how this approach can be applied to languages that model the behavior of a program as a tree. We study here partial order reductions for branching temporal logics, e.g., the logics CTL and CTL* (with the next time operator removed) and process algebra logics such as Hennesy–Milner logic (withτactions). Conditions on the selection of subset of successors from each state during the state-space construction, which guarantee reduction that preserves CTL* properties, are given. The experimental results provided show that the reduction is substantial.  相似文献

There are currently no epidemiological studies on health effects of long-term exposure to ultrafine particles (UFP), largely because data on spatial exposure contrasts for UFP is lacking. The objective of this study was to develop a land use regression (LUR) model for UFP in the city of Amsterdam. Total particle number concentrations (PNC), PM10, PM2.5, and its soot content were measured directly outside 50 homes spread over the city of Amsterdam. Each home was measured during one week. Continuous measurements at a central urban background site were used to adjust the average concentration for temporal variation. Predictor variables (traffic, address density, land use) were obtained using geographic information systems. A model including the product of traffic intensity and the inverse distance to the nearest road squared, address density, and location near the port explained 67% of the variability in measured PNC. LUR models for PM2.5, soot, and coarse particles (PM10, PM2.5) explained 57%, 76%, and 37% of the variability in measured concentrations. Predictions from the PNC model correlated highly with predictions from LUR models for PM2.5, soot, and coarse particles. A LUR model for PNC has been developed, with similar validity as previous models for more commonly measured pollutants.  相似文献

Summary 1. In eight series of triplicate tests on persons in good health and with normal secretory responses who were fed test meals of foods cooked with fat, the day to day variations in gastric evacuation times ranged from 0 to 30 minutes with an average deviation from the mean of ±17 minutes. 2. Experiments with meals of foods in which moderate quantities of fats were incorporated intimately by either baking or frying according to good culinary procedure revealed no relationship between the fat contents of the foods and the times required for complete evacuation of the foods from the subjects’ stomachs. 3. The experiments indicated a parallelism between the protein contents of the test meals and the gastric evacuation times. 4. No significant differences were observed between the influences of a hydrogenated vegetable fat and of butter upon the rates of evacuation from the subjects’ stomachs of meals of potatoes in which these fats had been incorporated intimately by good culinary technic. Within the limits of error of the experimental procedure, French fried and Lyonnaise potatoes containing moderate amounts of fat were evacuated from the stomachs as rapidly as boiled potatoes. 5. Gastric emptying times for meals of doughnuts did not differ significantly from gastric evacuation times for meals of bread and butter supplying equivalent amounts of fat. 6. The addition to a test meal of glucose in water of either hydrogenated vegetable fat or butter fat in amounts equivalent to two-thirds the weight of the glucose retarded the evacuation of the glucose meal from the stomach. 7. Excess fat, added to potatoes by pan-frying in hydrogenated vegetable fat or butter to the extent of making the potatoes “greasy,” such as may occur in poor culinary practice, prolonged the emptying time of the stomach beyond the period for boiled potatoes. 8. An explanation has been suggested for this retardation of the emptying of the stomach effected both by digestible fats added to glucose solutions and by foods containing fats in excess of that which is incorporated intimately in these foods by good culinary methods. ABSTRACT: BACKGROUND: Diesel exhaust particles (DEP) contribute substantially to ambient particulate matter (PM) air pollution in urban areas. Inhalation of PM has been associated with increased incidence of lung disease in susceptible populations. We have demonstrated that the glutathione Stransferase M1 (GSTM1) null genotype could aggravate DEP-induced airway inflammation in human subjects. Given the critical role airway epithelial cells play in the pathogenesis of airway inflammation, we established the GSTM1 deficiency condition in primary bronchial epithelial cells from human volunteers with GSTM1 sufficient genotype (GSTM1+) using GSTM1 shRNA to determine whether GSTM1 deficiency could exaggerate DEP-induced expression of interleukin-8 (IL-8) and IL-1beta proteins. Furthermore, the mechanisms underlying GSTM1 regulation of DEP-induced IL-8 and IL-1beta expression were also investigated. METHODS: IL-8 and IL-1beta protein levels were measured using enzyme-linked immunosorbent assay. GSTM1 deficiency in primary human bronchial epithelial cells was achieved using lentiviral GSTM1 shRNA particles and verified using real-time polymerase chain reaction and immunoblotting. Intracellular reactive oxygen species (ROS) production was evaluated using flow cytometry. Phosphorylation of protein kinases was detected using immunoblotting. RESULTS: Exposure of primary human bronchial epithelial cells (GSTM1+) to 25-100 mug/ml DEP for 24 h significantly increased IL-8 and IL-1beta protein expression. Knockdown of GSTM1 in these cells further elevated DEP-induced IL-8 and IL-1beta expression, implying that GSTM1 deficiency aggravated DEP-induced pro-inflammatory response. DEP stimulation induced the phosphorylation of extracellular signal-regulated kinase (ERK) and Akt, the downstream kinase of phosphoinositide 3-kinase (PI3K), in GSTM1+ bronchial epithelial cells. Pharmacological inhibition of ERK kinase and PI3K activity blocked DEP-induced IL-8 and IL-1beta expression. DEP-induced ERK and Akt phosphorylation could be increased by GSTM1 knockdown. In addition, pretreatment of HBEC with the antioxidant N-acetyl cysteine significantly inhibited DEP-induced ERK and Akt phosphorylation, and subsequent IL-8 and IL-beta expression. CONCLUSION: GSTM1 regulates DEP-induced IL-8 and IL-1beta expression in primary human bronchial epithelial cells by modulation of ROS, ERK and Akt signaling.  相似文献
