Arsenic is naturally associated with gold mineralisation and elevated in some soils and mine waste around historical gold mining activity in Victoria, Australia. To explore uptake, arsenic concentrations in children's toenail clippings and household soils were measured, and the microdistribution and speciation of arsenic in situ in toenail clipping thin sections investigated using synchrotron-based X-ray microprobe techniques. The ability to differentiate exogenous arsenic was explored by investigating surface contamination on cleaned clippings using depth profiling, and direct diffusion of arsenic into incubated clippings. Total arsenic concentrations ranged from 0.15 to 2.1 µg/g (n = 29) in clipping samples and from 3.3 to 130 µg/g (n = 22) in household soils, with significant correlation between transformed arsenic concentrations (Pearson's r = 0.42, P = 0.023) when household soil was treated as independent. In clipping thin sections (n = 2), X-ray fluorescence (XRF) mapping showed discrete layering of arsenic consistent with nail structure, and irregular arsenic incorporation along the nail growth axis. Arsenic concentrations were heterogeneous at 10 × 10 µm microprobe spot locations investigated (< 0.1 to 13.3 µg/g). X-ray absorption near-edge structure (XANES) spectra suggested the presence of two distinct arsenic species: a lower oxidation state species, possibly with mixed sulphur and methyl coordination (denoted As≈ III(-S, -CH3)); and a higher oxidation state species (denoted As≈ V(-O)). Depth profiling suggested that surface contamination was unlikely (n = 4), and XRF and XANES analyses of thin sections of clippings incubated in dry or wet mine waste, or untreated, suggested direct diffusion of arsenic occurred under moist conditions. These findings suggest that arsenic in soil contributes to some systemic absorption associated with periodic exposures among children resident in areas of historic gold mining activity in Victoria, Australia. Future studies are required to ascertain if adverse health effects are associated with current levels of arsenic uptake.  相似文献

Based on the rapid increase in incidence of inflammatory bowel disease (IBD), the identification of susceptibility genes and cell populations contributing to this condition is essential. Previous studies suggested multiple genes associated with the susceptibility of IBD; however, due to the analysis of whole-tissue samples, the contribution of individual cell populations remains widely unresolved. Single-cell RNA sequencing (scRNA-seq) provides the opportunity to identify underlying cellular populations. We determined the enrichment of Crohn’s disease (CD)-induced genes in a publicly available Crohn’s disease scRNA-seq dataset and detected the strongest induction of these genes in innate lymphoid cells (ILC1), highly activated T cells and dendritic cells, pericytes and activated fibroblasts, as well as epithelial cells. Notably, these genes were highly enriched in IBD-associated neoplasia, as well as sporadic colorectal cancer (CRC). Indeed, the same six cell populations displayed an upregulation of CD-induced genes in a CRC scRNA-seq dataset. Finally, after integrating and harmonizing the CD and CRC scRNA-seq data, we demonstrated that these six cell types display a gradual increase in gene expression levels from a healthy state to an inflammatory and tumorous state. Together, we identified cell populations that specifically upregulate CD-induced genes in CD and CRC patients and could, therefore, contribute to inflammation-associated tumor development.  相似文献

Oxidative stress has been directly implicated in hypertension and myocardial remodelling, two pathologies fundamental to the development of chronic heart failure. Selenium (Se) can act directly and indirectly as an antioxidant and a lowered Se status leads to a higher risk of cardiovascular disease. This study examined the role of Se on the development of hypertension and subsequent progression to chronic heart failure in spontaneously hypertensive rats (SHR). Three dietary groups were studied: (i) Se‐free; (ii) normal Se (50 μg Se/kg food); and (iii) high Se (1000 μg Se/kg food). Systolic blood pressure and echocardiography were used to detect cardiac changes in vivo. At study end, cardiac tissues were assayed for glutathione peroxidase activity, thioredoxin reductase activity, and protein carbonyls. The major finding of this study was the high heart failure‐related mortality rate in SHRs fed an Se‐free diet (70%). Normal and high levels of dietary Se resulted in higher survival rates of 78 and 100%, respectively. Furthermore, high dietary Se was clearly associated with lower levels of cardiac oxidative damage and increased antioxidant expression, as well as a reduction in disease severity and mortality in the SHR.  相似文献

Crystalline titanium dioxide (TiO2) coatings have been widely used in photo-electrochemical solar cell applications. In this study, TiO2 and carbon-doped TiO2 coatings were deposited onto unheated titanium and silicon wafer substrates using a DC closed-field magnetron sputtering system. The resultant coatings had an amorphous structure and a post-deposition heat treatment is required to convert this amorphous structure into the photoactive crystalline phase(s) of TiO2. This study investigates the use of a microwave plasma heat treatment as a means of achieving this crystalline conversion. The treatment involved placing the sputtered coatings into a 2.45 GHz microwave-induced nitrogen plasma where they were heated to approximately 550 °C. It was observed that for treatment times as short as 1 min, the 0.25-μm thick coatings were converted into the anatase crystalline phase of TiO2. The coatings were further transformed into the rutile crystalline phase after treatments at higher temperatures. The doping of TiO2 with carbon was found to result in a reduction in this phase transformation temperature, with higher level of doping (up to 5.8% in this study) leading to lower anatase-to-rutile transition temperature. The photoactivity performance of both doped and un-doped coatings heat-treated using both furnace and microwave plasma was compared. The carbon-doped TiO2 exhibited a 29% increase in photocurrent density compared to that observed for the un-doped coating. Comparing carbon-doped coatings heat-treated using the furnace and microwave plasma, it was observed that the latter yielded a 19% increase in photocurrent density. This enhanced performance may be correlated to the differences in the coatings' surface morphology and band gap energy, both of which influence the coatings' photoabsorption efficiency.  相似文献

Notes that recovery from combat-related posttraumatic stress disorder (PTSD) is often complicated by unacknowledged problems with alcohol and anger. 102 males combat veterans (aged 42–63 yrs) entering a residential PTSD rehabilitation program completed University of Rhode Island Change Assessment and process-of-change questionnaires based on J. O. Prochaska and C. C. DiClemente's transtheoretical model (TTM; J. O. Prochaska et al, 1992). Separate assessments were made for alcohol abuse and anger control. Four motivational subtypes were identified for both problems. Motivation to change alcohol problems was independent of that for anger. Relative to less-motivated peers highly motivated patients were more like to spontaneously identify alcohol or anger as problems in their life and made greater use of change strategies specified by the TTM. The impact of accelerated boning on tenderness, ageing rate and meat quality of pork longissimus muscle was investigated. Accelerated boning of eight Large White×Landrace pork carcasses at 30 min post-mortem followed by chilling at 0°C resulted in tougher pork with higher drip loss and a darker surface lightness relative to conventional boning at 24 h post-mortem. The increased toughness was still observed at 4 days post-mortem, a time by which 80% of ageing generally had occurred as seen in experiment 2. The increased toughness could be attributed to cold shortening and a reduction in proteolysis.  相似文献

The effects of stress on mothers' recall for a major hurricane were studied. Stress was objectively defined as low, moderate, or high according to the severity of home damage. This study of 96 mothers was conducted concurrently with L. E. Bahrick, J. F. Parker, R. Fivush, and M. Levitt (1998), allowing the authors to compare child and adult recall as a function of the same stressor. There was a quadratic relationship between storm severity and total recall for adults, similar to their children. Mothers' recall increased from low to moderate severity, but recall at moderate severity did not differ from high severity. These findings help clarify the effects of stress on the amount and type of information adults recall in retrospective accounts of naturalistic, temporally extended events. A finite-difference technique is used to compute exact solutions to the diffusion equation describing fission gas release from UO2 nuclear fuel during steady reactor operation. The resolution of gas atoms from grain-boundary bubbles is treated in two alternative ways, and the results of the parallel calculations compared. Predictions of gas release using simple analytical models are compared with the numerical results and are found in general to describe the process very accurately.  相似文献

The analysis of the element pattern in an array of waveguide elements on a cone is approached by systematically exploiting the geometrical symmetry of the structure. The method consists of decomposing the arbitrary array excitation into fundamental excitations (eigenexcitations) for which the solution of the electromagnetic problem is simpler than in the general case. Following Bailin and Silver, the fields external to the cone are represented as a superposition of modes TE and TM to the radial direction. For each eigenexcitation the fields in the waveguide elements are represented as a superposition of normal waveguide modes. The enforcement of the continuity of the electromagnetic fields at the conical interface provides complete information about the radiation from the elements and the reflection coefficients of the modes in the waveguide. The array element pattern is obtained by the superposition, with suitable weights, of the patterns of the eigenexcitations. The communication presents the computational procedures followed to evaluate with high speed and precision the roots of the Legendre functions necessary to represent the fields external to the conical structure. Numerical results for the realized element patterns of a particular conical array are presented in the final section.  相似文献

Proper linear models are those in which predictor variables are given weights such that the resulting linear composite optimally predicts some criterion of interest; examples of proper linear models are standard regression analysis, discriminant function analysis, and ridge regression analysis. Research summarized in P. Meehl's (1954) book on clinical vs statistical prediction and research stimulated in part by that book indicate that when a numerical criterion variable (e.g., graduate GPA) is to be predicted from numerical predictor variables, proper linear models outperform clinical intuition. Improper linear models are those in which the weights of the predictor variables are obtained by some nonoptimal method. The present article presents evidence that even such improper linear models are superior to clinical intuition when predicting a numerical criterion from numerical predictors. In fact, unit (i.e., equal) weighting is quite robust for making such predictions. The application of unit weights to decide what bullet the Denver Police Department should use is described; some technical, psychological, and ethical resistances to using linear models in making social decisions are considered; and arguments that could weaken these resistances are presented.
