Palmitoyl-CoA hydrolase [EC 3.1.2.2.] activity in rat liver was found to be enhanced by high fat diets. Partially hydrogenated marine oil and high-erucic acid rapeseed oil diets produced a greater increase than a diet containing soybean oil. With diets containing from 5 to 30% (w/w) of partially hydrogenated marine oil the increase in palmitoyl-CoA hydrolase activity was similar to the increase observed in peroxisomal β-oxidation activity (correlation coefficient r=0.94). A positive correlation (r=0.86) also was observed between the activity of palmitoyl-CoA hydrolase and previously determined levels of long-chain acyl-CoA. The results presented may suggest a common “induction” mechanism for palmitoyl-CoA hydrolase and peroxisomal β-oxidation enzymes, possibly exerted through an increased cellular level of long-chain acyl-CoA.  相似文献

Exposure of primary rat hepatocytes and human HepG2 cells to water-soluble garlic extracts resulted in the concentration-dependent inhibition of cholesterol biosynthesis at several different enzymatic steps. At low concentrations, sterol biosynthesis from [14C]acetate was decreased in rat hepatocytes by 23% with an IC50 (half-maximal inhibition) value of 90μg/mL and in HepG2 cells by 28% with an IC50 value of 35 μg/mL. This inhibition was exerted at the level of hydroxymethylglutaryl-COA reductase (MHG-CoA reductase) as indicated by direct enzymatic measurements and the absence of inhibition if [14C]mevalonate was used as a precursor. At high concentrations (above 0.5 mg/mL), inhibition of cholesterol biosynthesis was not only seen at an early step where it increased considerably with dose, but also at later steps resulting in the accumulation of the precursors lanosterol and 7-dehydrocholesterol. No desmosterol was formed which, however, was a major precursor accumulating in the presence of triparanol. Thus, the accumulation of sterol precursors seem to be of less therapeutic significance during consumption of garlic, because it requires concentrations one or two orders of magnitude above those affecting HMG-CoA reductase. Alliin, the main sulfur-containing compound of garlic, was without effect itself. If converted to allicin, it resulted in similar changes of the sterol pattern. This suggested that the latter compound might contribute to the inhibition at the late steps. In contrast, nicotinic acid and particularly adenosine caused moderate inhibition of HMG-CoA reductase activity and of cholesterol biosynthesis suggesting that these compounds participate, at least in part, in the early inhibition of sterol synthesis by garlic extracts. Indication and Identification of Archaeologic Fats In comparison to the food chemistry, the special tasks of fat indication in archaeologic discoveries consist in regarding possible changes of the fatty acid chains by soil storage. But changes of the composition expressed as percentages need not inevitably occur. There are many examples especially from zoologically determined subrecent bones that the total fat amount is indeed highly reduced, but the remaining residue seems unsignificantly altered. In case of alteration on an empiric basis a solution possibility can be achieved from two sides: pure fatty acid methylates are left to the effects of the natural environment. After some time the occurred changes are analysed. Since because of the relatively short period only introducing changes can be determined, you start from the other end: For archaeological findings enough time for changes has been available. Fats are extracted from objects which were identified by an archaeobotanist as seeds of certain plants or by an archaeozoologist as bones of certain animals and compared with recent fats. Ways of conversion get visible especially by a graduation of the findings according to the age.  相似文献

The work describes adsorption experiments from aqueous solutions on activated carbon using a counter-current adsorber. Phenol and paranitrophenol were used for single component adsorption and molasses colour for multicomponent adsorption experiments. Isotherms were determined using simple equilibrium experiments. Three different techniques were used for determination of the kinetics. These were finite bath, infinite bath and fixed bed experiments. A mathematical model for countercurrent adsorption was tested. The model includes the effect of film diffusion and pore diffusion as well as surface diffusion, and can handle systems with a nonlinear isotherms. Kinetic data in the form of an average diffusivity in the carbon determined from the finite bath experiments and isotherm data did not predict the operation of the countercurrent column well. Data from the fixed bed experiments predicted countercurrent column operation much better. This is deemed to be due to inhomogeneities in the carbon used, resulting in a fairly quick initial uptake of the main part of the solute and a slower uptake of a minor amount.  相似文献

Conventional gasification processes use coal not only as feedstock to be gasified but also for supply of energy for reaction heat, steam production, and other purposes. With a nuclear high temperature reactor (HTR) as a source for process heat, it is possible to transform the whole of the coal feed into gas. This concept offers advantages over existing gasification processes: saving of coal, as more gas can be produced from coal; less emission of pollutants, as the HTR is used for the production of steam and electricity instead of a coal-fired boiler; and a lower production cost for the gas. However, the process has the disadvantage that the temperature is limited to the outlet temperature (950 °C max) of the helium cooling gas of the HTR. Therefore the possibility of catalytic steam gasification was examined. Model calculations based on experimental results show that use of 3–4 wt% relative to coal of K2CO3 catalyst increases the throughput of a large scale nuclear gasification plant by ≈65%, while gas production costs decrease by ≈15%. Corrosion by catalysts is not significant at low concentration (< 5 wt%) and low temperature (< 900 °C).  相似文献

We propose a system that affords real-time sound synthesis of footsteps on different materials. The system is based on microphones, which detect real footstep sounds from subjects, from which the ground reaction force (GRF) is estimated. Such GRF is used to control a sound synthesis engine based on physical models. Two experiments were conducted. In the first experiment, the ability of subjects to recognize the surface they were exposed to was assessed. In the second experiment, the sound synthesis engine was enhanced with environmental sounds. Results show that, in some conditions, adding a soundscape significantly improves the recognition of the simulated environment.  相似文献

Reconfigurable computing offers a wide range of low cost and efficient solutions for embedded systems. The proper choice of the reconfigurable device, the granularity of its processing elements and its memory architecture highly depend on the type of application and their data flow. Existing solutions either offer fine grain FPGAs, which rely on a hardware synthesis flow and offer the maximum degree of flexibility, or coarser grain solutions, which are usually more suitable for a particular type of data flow and applications. In this paper, we present the MORPHEUS architecture, a versatile reconfigurable heterogeneous System-on-Chip targeting streaming applications. The presented architecture exploits different reconfigurable technologies at several computation granularities that efficiently address the different applications needs. In order to efficiently exploit the presented architecture, we implemented a complete software solution to map C applications to the reconfigurable architecture. In this paper, we describe the complete toolset and provide concrete use cases of the architecture.  相似文献
