Leucine modulates synthetic and degradative pathways in muscle, possibly providing metabolic benefits for both athletes and diseased populations. Leucine has become popular among athletes for improving performance and body composition, however little is known about the metabolic effects of the commonly consumed leucine‐derived metabolite β‐hydroxy‐β‐methyl butyrate (HMB). Our work measured the effects of HMB on metabolic protein expression, mitochondrial content and metabolism, as well as lipid content in skeletal muscle cells. Specifically, cultured C2C12 myotubes were treated with either a control or HMB ranging from 6.25 to 25 μM for 24 h and mRNA and/or protein expression, oxygen consumption, glucose uptake, and lipid content were measured. Contrary to leucine's stimulatory effect on metabolism, HMB‐treated cells exhibited significantly reduced regulators of lipid oxidation including peroxisome proliferator‐activated receptor alpha (PPARα) and PPARβ/δ, as well as downstream target carnitine palmitoyl transferase, without alterations in glucose or palmitate oxidation. Furthermore, HMB significantly inhibited activation of the master regulator of energetics, AMP‐activated protein kinase. As a result, HMB‐treated cells also displayed reduced total mitochondrial content compared with true control or cells equivocally treated with leucine. Additionally, HMB treatment amplified markers of lipid biosynthesis (PPARγ and fatty acid synthase) as well as consistently promoted elevated total lipid content versus control cells. Collectively, our results demonstrate that HMB did not improve mitochondrial metabolism or content, and may promote elevated cellular lipid content possibly through heightened PPARγ expression. Neglected tropical diseases caused by parasitic infections are an ongoing and increasing concern. They are a burden to human and animal health, having the most devastating effect on the world′s poorest countries. Building upon our previously reported triazole analogues, in this study we describe the synthesis and biological testing of other novel heterocyclic acetogenin‐inspired derivatives, namely 3,5‐isoxazoles, furoxans, and furazans. Several of these compounds maintain low‐micromolar levels of inhibition against Trypanosoma brucei, whilst having no observable inhibitory effect on mammalian cells, leading to the possibility of novel lead compounds for selective treatment. 相似文献

The bibliometric indices of the scientific field of geostatistics were analyzed using statistical and spatial data analysis.
The publications and their citation statistics were obtained from the Web of Science (4000 most relevant), Scopus (2000 most
relevant) and Google Scholar (5389). The focus was on the analysis of the citation rate (CR), i.e. number of citations an
author or a library item receives on average per year. This was the main criterion used to analyze global trends in geostatistics
and to extract the Top 25 most-cited lists of the research articles and books in geostatistics. It was discovered that the
average citation rate for geostatisticians has stabilized since 1999, while the authors’ n-index seems to have declined ever
since. One reason for this may be because there are more and more young authors with a lower n-index. We also found that the
number of publications an author publishes explains only 60% of the variation in the citation statistics and that this number
progressively declines for an author with a lower number of publications. Once the geographic location is attached to a selection
of articles, an isotropic Gaussian kernel smoother weighted by the CR can be used to map scientific excellence around the
world. This revealed clusters of scientific excellence around locations such as Wageningen, London, Utrecht, Hampshire, UK,
Norwich, Paris, Louvain, Barcelona, and Zürich (Europe); Stanford, Ann Arbor, Tucson, Corvallis, Seattle, Boulder, Montreal,
Baltimore, Durham, Santa Barbara and Los Angeles (North America); and Canberra, Melbourne, Sydney, Santiago (Chile), Taipei,
and Beijing (other continents). Further correlation with socio-economic variables showed that the spatial distribution of
CRs in geostatistics is independent of the night light image (which represents economic activity) and population density.
This study demonstrates that the commercial scientific indexing companies could enhance their service by assigning the geographical
location to library items to allow spatial exploration and analysis of bibliometric indices. 相似文献

This review identified fourteen reported illness outbreaks attributed to consumption of pathogen-contaminated spice during the period 1973–2010. Countries reporting outbreaks included Canada, Denmark, England and Wales, France, Germany, New Zealand, Norway, Serbia, and the United States. Together, these outbreaks resulted in 1946 reported human illnesses, 128 hospitalizations and two deaths. Infants/children were the primary population segments impacted by 36% (5/14) of spice-attributed outbreaks. Four outbreaks were associated with multiple organisms. Salmonella enterica subspecies enterica was identified as the causative agent in 71% (10/14) of outbreaks, accounting for 87% of reported illnesses. Bacillus spp. was identified as the causative agent in 29% (4/10) of outbreaks, accounting for 13% of illnesses. 71% (10/14) of outbreaks were associated with spices classified as fruits or seeds of the source plant. Consumption of ready-to-eat foods prepared with spices applied after the final food manufacturing pathogen reduction step accounted for 70% of illnesses. Pathogen growth in spiced food is suspected to have played a role in some outbreaks, but it was not likely a contributing factor in three of the larger Salmonella outbreaks, which involved low-moisture foods. Mycobacterium avium subsp. paratuberculosis (M. paratuberculosis) causes Johne's disease in ruminants (including cattle, sheep and goats) and other animals, and may contribute to Crohn's disease in humans. This possibility, and the fact that M. paratuberculosis may be present in raw milk, make it important to ensure that the heat treatment specified for pasteurization of milk will give acceptable inactivation of this bacterium, with an adequate margin of safety. Published studies of the heat resistance of this bacterium in milk have given widely differing results. Possible reasons for these differences, and the technical problems involved in the work, are reviewed. It is concluded that there is a need (i) for the adoption of an agreed Performance Criterion for pasteurization of milk in relation to this bacterium, (ii) a need for definitive laboratory experiments to understand and determine the heat resistance of M. paratuberculosis, and (iii) a need for an assessment of whether the minimum heat treatments specified at present for pasteurization of milk (Process Criteria) will meet the Performance Criterion for M. paratuberculosis. Measures are also required to ensure that commercial processes deliver continually the specified heat treatment, and to ensure that post-pasteurization contamination is avoided. 相似文献

Inferior temporal cortex of squirrel monkeys consists of caudal (ITC), intermediate (ITI), and rostral (ITR) subdivisions, possibly homologous to TEO, posterior TE, and anterior TE of macaque monkeys. The present study compared visual learning in squirrel monkeys with ablations of ITC; ITI and ITR (group ITRd); or ITI, ITR, and more ventral cortex, including perirhinal cortex (group ITR+), with visual learning in unoperated controls. The ITC monkeys had significant impairments on pattern discriminations and milder deficits on delayed nonmatching to sample (DNMS) of objects. The ITRd monkeys had deficits on some pattern discriminations but not on DNMS. The ITRd monkeys were significantly impaired on DNMS and some pattern discriminations. The iron and steel industry is under considerable pressure to continuously reduce the levels of generated dioxins and other harmful emissions from process offgas. In the early 1990s this industry commenced with the investigation of new offgas cleaning systems for sinter plants and electric arc furnaces (EAF) which resulted in a considerably improved offgas cleaning efficiency. The global tendency in the steel industry towards an increased application of the electric arc furnace for the production of steel and especially the introduction of scrap-pre-heating technologies in the late 1980s resulted in increased demands for VOC and dioxin reduction technologies.
