We are all aware that radio communication is not instantaneous. With the coming of satellite communication we have fallen prey to some of the consequences of telephony with long propagation time. So it is that when one person speaks to another via satellite link, he must wait perhaps 600 ms for a rejoinder. Such a delay, of itself, may not be too disconcerting, but in combination with echo it can contribute to poorer quality transmission. In an attempt to decrease the incidence of driving under the influence of alcohol (DUI), some alcohol-serving establishments have installed blood-alcohol level (BAL) feedback devices. These devices are meant to increase awareness of intoxication and result in the avoidance of DUI. However, studies in the US, Canada, and New Zealand (e.g., J. F. Oates, 1976; J. Calvert-Boyanowsky and E. O. Boyanowsky, 1980; P. M. Hurst and P. G. Wright, 1981) indicate that BAL feedback does not deter intoxicated individuals from DUI. Moreover, there is some evidence that it may increase alcohol consumption. For the transient dynamic analysis of structural systems, the direct numerical integration of the equations of motion may be regarded as an alternative to the mode superposition method for linear problems and a necessity for nonlinear problems. When compared to a modal superposition solution, the direct integration approach is attractive in that the eigenvalue problem is avoided. Depending on the amount of information required from the dynamic analysis, e.g. frequencies, frequencies and mode shapes, and/or a complete time history, a direct integration scheme may prove to be more efficient than a modal superposition solution for some linear problems as well.The purpose of this study is to develop and demonstrate a direct integration algorithm which is compatible with an existing high-precision rotational shell finite element. Excellent comparative efficiency for static problems was achieved with this element by the incorporation of the exact geometry, the utilization of high-order interpolation polynomials and, yet, the retention of only a minimum number of nodal variables in the global formulation. Likewise, accurate and efficient results for the free vibration analysis of rotational shells were facilitated by the inclusion of a consistent mass matrix and the utilization of a rationally justified kinematic condensation procedure. The approach to the direct integration stage is strongly tempered by the established characteristics of this element which enable a given shell to be modeled accurately in the spatial domain with a comparatively coarse discretization.The equations of motion for a shell of revolution under conservative loading are derived from Hamilton's variational principle and specialized for the discretization of a rotational shell into curved shell elements. Degrees of freedom in excess of those required to establish minimum (C°) continuity at the nodal circles are eliminated through kinematic condensation. Some guidance as to the proper order of the polynominal approximations for a dynamic analysis is provided by earlier free vibration studies. Whereas the condensation is exact for static problems, it is only approximate for dynamic response and it was found that the accuracy of the eigenvalues obtained for the reduced problem decreases with increasing order of the condensed functions. This tendency is counted by the desirability of using sufficiently high-order interpolations so as to permit accurate stress computations, both at the nodal circles and between nodes since a coarse discretization is necessary to realize maximum efficiency. It was found that cubic polynomials were generally satisfactory from both standpoints, except in localized regions of high stress gradients where quintic polynomials were employed. The finite element discretizations for the direct integration studies were selected on this basis.For the high-precision finite element at hand, the efficiencies achieved in the space domain are demonstrated by the ability to achieve precise solutions with relatively coarse discretization patterns. The resulting comparatively large elements are not subject to accurate representation by diagonal mass matrices so that an implicit, consistent mass approach is followed. Efficiency in the time domain as well rests on the successful modeling of rotational shells subject to dynamic loading using coarse discretitations in space and large increments in time. Computational efficiency and accuracy are demonstrated for various problems documented in the literature, including a shallow spherical cap subject to a step pulse and a hyperboloidal shell under a simulated dynamic wind pressure. 相似文献

The Pichia pastoris PAS4 gene encodes a ubiquitin-conjugating enzyme required for peroxisome assembly 总被引:2,自引:0,他引:2
We report here the cloning and initial characterization of PAS4, a gene required for peroxisome assembly in the yeast Pichia pastoris. The PAS4 gene encodes a 24-kDa protein (Pas4p) that is located on the cytoplasmic surface of peroxisomes and is induced during peroxisome proliferation. Analysis of the Pas4p sequence revealed a high degree of similarity to ubiquitin-conjugating enzymes, particularly in the region surrounding the putative active-site cysteine residue with which ubiquitin forms a thioester bond. As expected for a ubiquitin-conjugating enzyme, substitution of alanine or serine for the conserved active-site cysteine residue abolished PAS4 function. In addition, a small amount of a 32 kDa form of Pas4p (the predicted size of a Pas4p-ubiquitin conjugate) was detected both in vivo and in vitro. This species was eliminated by reducing agents and was not detected in the cysteine to alanine substitution mutant, suggesting that it is a Pas4p-ubiquitin conjugate. Using a yeast strain that overexpresses a Myc-ubiquitin fusion protein, we demonstrate directly that this conjugate contains ubiquitin. We conclude from these observations that PAS4 is a member of the ubiquitin-conjugating enzyme gene family and that one or more ubiquitination reactions are required for peroxisome assembly. 相似文献

This paper presents the general and analytical solution of a fourth-order lumped element model (LEM) to describe human body model (HBM) electrostatic discharge (ESD) testers including the main tester parasitic elements. The analytical fitting to the LEM of experimentally obtained HBM pulse data is a new scientifically justified tool to determine HBM tester parasitic elements. The impact of the test board capacitance on HBM testing is demonstrated and explained. Furthermore, the MIL 883C/3015.71 and EOS/ESD S5.1-19912 Standards on HBM testers are evaluated upon their selectivity to the test board capacitance. Although sprawl is a growing national debate, there have been few efforts to measure or monitor changes in degree of sprawl over time. By using a methodology that employs census data, this sprawl index allows computation of levels of sprawl and examination of temporal and geographic changes. The results show that sprawl has increased over the past decade in many metropolitan areas.
