The present study was designed to investigate the effect of ethanol (EtOH) dose on low density lipoprotein (LDL) and platelet composition. Male squirrel monkeys were divided into three groups designated Control, Low, and High EtOH, and fed isocaloric liquid diets containing 0%, 12%, and 24% of calories as EtOH, respectively. After four months of treatment, monkeys fed the 12% alcohol dose had LDL and platelet cholesterol concentrations similar to Controls. By contrast, platelet membranes from High EtOH animals contained significantly more cholesterol which was associated with higher levels of plasma LDL cholesterol and apolipoprotein B. Blood platelet count, size, and mass were similar for all groups and circulating platelet aggregates were absent in the two alcohol cohorts. Despite elevations in platelet cholesterol mass and thromboxane A2 (TXA2) precursor, phospholipid arachidonate, platelet responsiveness, measured as thromboxane formed in response to a collagen challengein vitro, and the cholesterol/phospholipid molar ration, were not significantly altered by high dose alcohol. Normal platelet activity in High EtOH monkeys may have resulted from a significant increase in the platelet phospholipid polyunsaturated/saturated fatty acid ratio and a non-significant increase in platelet phospholipid mass, both of which would have a fluidizing effect on platelet membranes. Our data indicate that low EtOH intake has no effect on platelet composition and function while unfavorable platelet cholesterol enrichment following consumption of high dose ethanol may arise from elevations in plasma LDL. Potentially thrombogenic (TXA2 formation, platelet aggregate formation) consequences of increased platelet cholesterol in squirrel monkeys fed high levels of alcohol may be averted by compensatory physicochemical changes in platelet membrane lipid composition although persistent elevations in circulating LDL may contribute to increased risk of cardiovascular disease.  相似文献

By adding a gold core to silica nanoparticles (BrightSilica), silica‐like nanoparticles are generated that, unlike unmodified silica nanoparticles, provide three types of complementary information to investigate the silica nano‐biointeraction inside eukaryotic cells in situ. Firstly, organic molecules in proximity of and penetrating into the silica shell in live cells are monitored by surface‐enhanced Raman scattering (SERS). The SERS data show interaction of the hybrid silica particles with tyrosine, cysteine and phenylalanine side chains of adsorbed proteins. Composition of the biomolecular corona of BrightSilica nanoparticles differs in fibroblast and macrophage cells. Secondly, quantification of the BrightSilica nanoparticles using laser ablation inductively coupled plasma mass spectrometry (LA‐ICP‐MS) micromapping indicates a different interaction of silica nanoparticles compared to gold nanoparticles under the same experimental conditions. Thirdly, the metal cores allow the investigation of particle distribution and interaction in the cellular ultrastructure by cryo nanoscale X‐ray tomography (cryo‐XT). In 3D reconstructions the assumption is confirmed that BrightSilica nanoparticles enter cells by an endocytotic mechanism. The high SERS intensities are explained by the beneficial plasmonic properties due to agglomeration of BrightSilica. The results have implications for the development of multi‐modal qualitative and quantitative characterization in comparative nanotoxicology and bionanotechnology.  相似文献

Fatigue crack growth rates (FCGR) for a steel similar to AISI 316LN have been evaluated in air and salt water. At free corrosion potential, a short crack effect appeared for crack lengths shorter than 4  mm. For longer cracks, a segment (called region I) of the corrosion FCGR curve can be described by an equation with the same coefficients as for steels of ordinary resistance to corrosion. Thus, the process controlling crack propagation is presumably the same for both steels. Corrosion FCGR in region II (range of longer crack lengths and higher Δ K values, where the curve cannot be described by the previously mentioned equation) are slower than for ordinary steels. The plateau in FCGR observed at a cathodic potential corresponds to the higher plateau at the free corrosion potential. An interpretation is presented for the shape of the corrosion FCGR curves at both potentials.  相似文献

As the most ambitious concept of isotope separation on line (ISOL) facility, EURISOL aims at producing unprecedented intensities of post-accelerated radioactive isotopes. Charge breeding, which transforms the charge state of radioactive beams from 1+ to an n+ charge state prior to post-acceleration, is a key technology which has to overcome the following challenges: high charge states for high energies, efficiency, rapidity and purity. On the roadmap to EURISOL, a dedicated R&D is being undertaken to push forward the frontiers of the present state-of-the-art techniques which use either electron cyclotron resonance or electron beam ion sources. The results of numerical modeling of the base transit time and collector current of SiGe-base heterojunction bipolar transistors with a Gaussian base doping profile and two Ge profiles (linearly graded and box) are presented for the first time. The importance of including the dependence of minority carrier mobility on the drift field and the dependence of the effective density of states on the Ge concentration along the base is demonstrated through the analysis of base transit time and collector current. A function describing the decrease of the density of states product in strained SiGe layers with increasing Ge concentration is proposed.  相似文献

A theoretical study of the local buckling of thin-walled girders under compound loading is presented. Girders of triangular cross-section with one axis of symmetry are considered. The following load cases are examined: compression, eccentric compression, pure bending and bending with shear. A parabolic distribution of shear stress in the webs is assumed when lateral forces are present.

The problem is solved using a semi-energy method, described in Jakubowski, S., Buckling of thin-walled girders under compound load, Thin-walled Structures,6 (1988) 129-50. Numerical results are presented in the form of diagrams. The analysis of buckling modes for the more complicated cases such as bending with shear has also been performed.  相似文献

Investigated the responses toward young shown by males and nulliparous females in 2 outbred stocks of laboratory rats (214 Sprague-Dawley and 166 Wistar rats) in 3 experiments. Sprague-Dawley females showed maternal behavior either spontaneously (35% of the naive Ss) or through concaveation (92% of the initially neutral virgins). Of the Wistar females, however, only 10% showed maternal behavior spontaneously, and only 29% of the neutral virgins came to behave maternally during 15 days of concaveation. Prepubertal cohabitation with lactating Ss did not facilitate maternal responsiveness in adulthood in the Wistar virgin females. Of the Sprague-Dawley males, 50% showed paternal behavior spontaneously, and only 4% killed the young. Among the Wistar males, however, only 4% showed paternal behavior spontaneously, and 76% killed pups. It is suggested that such profound differences between outbred stocks of Ss may be a source of discrepancies between the results of studies dealing with the induction of parental behavior in nonlactating rats.
