General anesthetics mainly act by modulating synaptic inhibition on the one hand (the potentiation of GABA transmission) or synaptic excitation on the other (the inhibition of NMDA receptors), but they can also have effects on numerous other proteins, receptors, and channels. The effects of general anesthetics on ion channels have been the subject of research since the publication of reports of direct actions of these drugs on ion channel proteins. In particular, there is considerable interest in T-type voltage-gated calcium channels that are abundantly expressed in the thalamus, where they control patterns of cellular excitability and thalamocortical oscillations during awake and sleep states. Here, we summarized and discussed our recent studies focused on the CaV3.1 isoform of T-channels in the nonspecific thalamus (intralaminar and midline nuclei), which acts as a key hub through which natural sleep and general anesthesia are initiated. We used mouse genetics and in vivo and ex vivo electrophysiology to study the role of thalamic T-channels in hypnosis induced by a standard general anesthetic, isoflurane, as well as novel neuroactive steroids. From the results of this study, we conclude that CaV3.1 channels contribute to thalamocortical oscillations during anesthetic-induced hypnosis, particularly the slow-frequency range of δ oscillations (0.5–4 Hz), by generating “window current” that contributes to the resting membrane potential. We posit that the role of the thalamic CaV3.1 isoform of T-channels in the effects of various classes of general anesthetics warrants consideration. 相似文献

The stability of alumina suspensions in the presence of Tiron was studied and discussed in the light of surface properties of alumina powder used, ionization chemistry of Tiron, and sedimentation behavior of the suspensions. The point of zero charge of alumina with and without addition of Tiron was determined using a batch equilibration method. The sedimentation of the suspensions was evaluated via screen sedimentation tests as a function of pH and Tiron concentration. The dissociation constant, pKa1, of the hydroxyl groups of the Tiron molecule was determined by potentiometric titration carried out in water and KNO3 as a background electrolyte. Tiron was shown to adsorb specifically onto the alumina surface in the examined pH range 4–10, but impart stability to the suspensions only at pH ≥ 6. 相似文献

Adequate assessment of human health risk of uranium contamination at hazardous waste sites, which is an important step in determining the cleanup strategy, is based on bioavailability data. Bioavailability of uranium from contaminated soil has not been properly determined yet. Bioaccessibility is an in vitro conservative estimate of bioavailability and is thus frequently used for site-specific risk assessment. Bioaccessibility of uranium was measured in 33 soil samples from the Port Hope area in Ontario, Canada, by the physiologically based extraction test (PBET). Higher bioaccessibility values in the gastric plus intestinal phase, 48.4% ± 16.8%, than in the gastric phase, 20.8% ± 11.7%, are very probably the result of more efficient extraction of uranium from soil by intestinal fluid rich in carbonate ions. The observed variability of measured bioaccessibility values is discussed in light of the results of scanning electron microscope examination of the soil samples. Uranium bioaccessibility values in both gastric (acidic) and gastric plus intestinal (neutral) phases are higher in soil samples with smaller uranium-bearing particles and lower in samples where the uranium-bearing particles are larger. We postulate that the most important reason for variability of measured bioaccessibility values in Port Hope soil samples may be the difference in particle size of uranium-bearing particles. 相似文献

The aim of this paper was to present the energy flows in the life cycle of biogas utilization systems (cogeneration and transportation), as well as their mutual relations, starting from providing the feedstock for digestion through to end-of-life management of biogas system as fertilizer on agricultural land. This study was carried out through the energy analysis of two scenarios (biogas in cogeneration and biogas in transportation) using performance of Mirotin biogas plant (1 MW) in Serbia. Results obtained in this study have shown that the analyzed scenario (biogas in cogeneration and biogas in transportation) have positive energy balances (52,114 and 53,585 GJ) and these scenarios are sustainable from energetic point of view. 相似文献
