Endothelial and epithelial barrier function is crucial for the maintenance of physiological processes. The barrier paracellular permeability depends on the composition and spatial distribution of the cell-to-cell tight junctions (TJ). Here, we provide an experimental workflow that yields several layers of physiological data in the setting of a single endothelial cell monolayer. Human umbilical vein endothelial cells were grown on Transwell filters. Transendothelial electrical resistance (TER) and 10 kDa FITC dextran flux were measured using Alanyl-Glutamine (AlaGln) as a paracellular barrier modulator. Single monolayers were immunolabelled for Zonula Occludens-1 (ZO-1) and Claudin-5 (CLDN5) and used for automated immunofluorescence imaging. Finally, the same monolayers were used for single molecule localization microscopy (SMLM) of ZO-1 and CLDN5 at the nanoscale for spatial clustering analysis. The TER increased and the paracellular dextran flux decreased after the application of AlaGln and these functional changes of the monolayer were mediated by an increase in the ZO-1 and CLDN5 abundance in the cell–cell interface. At the nanoscale level, the functional and protein abundance data were accompanied by non-random increased clustering of CLDN5. Our experimental workflow provides multiple data from a single monolayer and has wide applicability in the setting of paracellular studies in endothelia and epithelia. 相似文献

The intelligent recycling of plastics waste is a major concern. Because of the widespread use of polyethylene terephtalate, considerable amounts of PET waste are generated that are ideally re‐introduced into the material cycle by generating second generation products without loss of materials performance. Chemical recycling methods are often expensive and entail environmentally hazardous by‐products. Established mechanical methods generally provide materials of reduced quality, leading to products of lower quality. These drawbacks can be avoided by the development of new recycling methods that provide materials of high quality in every step of the production cycle. In the present work, oligomeric ethylene terephthalate with defined degrees of polymerization and defined molecular weight is produced by melt‐mixing PET with different quantities of adipic acid as an alternative pathway of recycling PET with respect to conventional methods, offering ecofriendly and economical aspects. The materials typically used for oxygen transport membranes, Ba0.5Sr0.5Co0.8Fe0.2O3?δ (BSCF) and La0.6Sr0.4Co0.2Fe0.8O3 (LSCF) tend to decompose due to their low thermochemical stability under reducing atmosphere. Fe- and Co-doped SrTiO3 (SrTi1-x-yCoxFeyO3-δ, x + y ≤ 0.35) (STCF) materials showing an oxygen transport comparable to LSCF have great potential for application in ion-transport-devices. In this study, the thermochemical stability of pure perovskite-structured STCF was investigated after annealing in a syngas atmosphere at 600–900 °C. The phase composition of the materials after annealing was characterized by means of X-ray diffraction (XRD). The thermodynamic activities of SrO, FeO, and CoO in the STCF materials were evaluated using Knudsen effusion mass spectrometry (KEMS). Co-doped SrTiO3 (STC) materials were not stable after annealing in the syngas atmosphere above 5 mol% Co-substitution. Ruddlesden-Popper-like phases and SrCO3 were detected after annealing at 600 °C. In contrast, Fe substitution (STF) showed good stability after annealing in syngas upto 35 mol% substitution. 相似文献

A subsea flexible pipe containing isotropic poly(vinylidene fluoride) (PVDF) layers is subjected to rapid decompression after being put in pressurized equilibrium with a fluid containing supercritical CO2. The PVDF layers have flowed radially into gaps of adjacent metallic coils forming whitened noses. Microbeam small‐angle X‐ray scattering (SAXS) scanning reveals a considerable morphological gradient in the polymer layers. PVDF layers (inner: wear, outer: barrier) in two zones (undamaged and damaged) are scanned. Far from noses and damage zone, the samples are isotropic without voids. Their morphological parameters are determined and compared to virgin material. Approaching the noses, the structure turns into highly oriented microfibrils perpendicular to the local flow into the gaps. Here, voids extend parallel to the microfibrils. At edges of the metallic structure, they turn more perpendicular to the layer. Crystallite orientation extends out to both sides of the whitened nose regions, but in the undamaged samples, tilting of the orientation direction and void‐formation are restricted to the white regions: successive mechanisms of cold drawing are mapped into space. Two series of aromatic polyimides containing various linkage groups based on 2,7‐bis(4‐aminophenoxy)naphthalene or 3,3′‐dimethyl‐4,4′‐diaminodiphenylmethane and different aromatic dianhydrides, namely 4,4′‐(4,4′‐isopropylidenediphenoxy)bis(phthalic anhydride), 4,4′‐(hexafluoroisopropylidene)bis(phthalic anhydride), 3,3′,4,4′ benzophenonetetracarboxylic dianhydride, 9,9‐bis[4‐(3,4‐dicarboxyphenoxy)phenyl]fluorene dianhydride and 4,4′‐(4,4′‐hexafluoroisopropylidenediphenoxy)bis(phthalic anhydride), were synthesized and compared with regard to their thermal, mechanical and gas permeation properties. All these polymers showed high thermal stability with initial decomposition temperature in the range 475–525 °C and glass transition temperature between 208 and 286 °C. Also, the polymer films presented good mechanical characteristics with tensile strength in the range 60–91 MPa and storage modulus in the range 1700–2375 MPa. The macromolecular chain packing induced by dianhydride and diamine segments was investigated by examining gas permeation through the polymer films.
