Mechanistic links have been suggested between repolarization alternans (RPA) and the onset of ventricular tachycardia (VT) and/or fibrillation. Endocardial detection of RPA may, therefore, be an important step in future device-based treatments of arrhythmias. Here, we investigate if RPA could be detected during acute ischemia using an implantable cardioverter defibrillator (ICD) lead (tip to distal coil) located in the right ventricular apex. In 18 pigs, the right coronary (n = 10) or left anterior descending coronary (n = 8) artery was occluded for 10 min using a balloon catheter, followed by reperfusion for 30 min, and re-occlusion for 30 min. RPA magnitude, computed using the modified moving average (MMA) method, showed a sharp increase in all 18 animals, from a mean baseline level of 1.9 +/- 1.3 mV to 3.0 +/- 1.3 mV during first occlusion (p < 0.001). RPA magnitude showed a prominent increase in 10 animals during re-occlusion, from a mean baseline level of 1.7 +/- 1.0 mV to 3.3 +/- 1.5 mV (p < 0.001). The protocol was terminated during the first two stages of occlusion and reperfusion for the remaining 8 animals due to the occurrence of ventricular fibrillation (VF). These results confirm that RPA increases under ischemic conditions and that it is possible to detect and track RPA dynamics with an ICD lead that is positioned in a clinically realistic location. Such an approach may be useful in formulating improved arrhythmia detection and control algorithms.  相似文献

Data recorded in electroencephalogram (EEG)-based brain-computer interface experiments is generally very noisy, non-stationary, and contaminated with artifacts that can deteriorate discrimination/classification methods. In this paper, we extend the common spatial pattern (CSP) algorithm with the aim to alleviate these adverse effects. In particular, we suggest an extension of CSP to the state space, which utilizes the method of time delay embedding. As we will show, this allows for individually tuned frequency filters at each electrode position and, thus, yields an improved and more robust machine learning procedure. The advantages of the proposed method over the original CSP method are verified in terms of an improved information transfer rate (bits per trial) on a set of EEG-recordings from experiments of imagined limb movements.  相似文献

A multidimensional, batch high-throughput screening (MD-HTS) protocol was developed to investigate the effects of various parameters on the selectivity of ion-exchange protein displacement systems. A variety of molecules were screened, and the results were employed to provided insights into the influence of displacer chemistry and concentration, resin chemistry, and mobile-phase salt counterion on the efficacy and selectivity of these nonlinear chromatographic systems. These results open up the possibility of tailoring the selectivity of displacement separations by choosing appropriate combinations of operating conditions using the MD-HTS technique. The screens were also employed for the identification of displacers and conditions for the separation of a challenging protein mixture by selective displacement chromatography. Column displacements were carried out with potential lead compounds identified from the MD-HTS screens, and the results confirmed that selective displacement could indeed be achieved for this model mixture. Furthermore, the results indicated that this approach is particularly useful when the order of elution is not changed, but the inherent selectivity is increased in the presence of the displacer. The results presented in this paper demonstrate the utility of the MD-HTS technique for rapid method development in protein ion-exchange displacement chromatography.  相似文献

Composite neutron scintillators consisting of neutron-insensitive fluorescent dopant particles (e.g. ZnS:Ag) embedded in a matrix material containing isotopes with high neutron cross sections that emit energetic charged particles (e.g. 6Li) are a popular method for neutron detection in a variety of applications. The size and volume doping fraction of the fluorescent dopant particles and the densities of both dopant particles and the matrix material determine the characteristics of the pulse-height spectrum of emitted light and the probability that capture of a neutron will result in scintillation. In this work, we characterise the effects of these parameters for ZnS:Ag particles in a lithiated glass matrix using a Monte Carlo simulation of composite neutron detectors that we have constructed.  相似文献

The potential of Raman spectroscopy to discriminate between architectural finishes (household paint) has been investigated using a test set of 51 'lilac' paints and three different excitation wavelengths. The spectra obtained with visible excitation typically displayed a series of intense Raman bands on a featureless fluorescence background but the spectra of all the paints studied had essentially identical bands. With 785 nm excitation, although the same bands that dominated the 514 nm spectra were still observed, other bands with comparable intensity also appeared. The two strongest scattering constituents were identified as a dioxazine dye, Violet 23 and beta-Cu(phthalocyanine). A scatter plot of the intensities of marker bands for these constituents (normalized to the strong rutile bands that were always present) showed that, despite the fact that the sample set spanned a wide range of rutile : dioxazine dye : phthalo- cyanine ratios, many of the samples had very similar ratios and could not be discriminated. However, all the samples (even those with similar relative proportions of the main constituents) could be discriminated on the basis of their minor constituents, either by manually measuring band intensities or through the creation and searching of spectral libraries.  相似文献

The National Aeronautics and Space Administration's (NASA's) Ames Research Center's Airborne Sensor Facility (ASF) is responsible for the calibration of several airborne Earth-viewing sensor systems in support of NASA Earth Observing System (EOS) investigations. The primary artifact used to calibrate these sensors in the reflective solar region from 400 to 2500 nm is a lamp-illuminated integrating sphere source. In September 1999, a measurement comparison was made at the Ames ASF Sensor Calibration Facility to validate the radiometric scale, establish the uncertainties assigned to the radiance of this source, and examine its day-to-day repeatability. The comparison was one of a series of validation activities overseen by the EOS Calibration Program to ensure the radiometric calibration accuracy of sensors used in long-term, global, remote-sensing studies. Results of the comparison, including an evaluation of the Ames Sensor Calibration Laboratory (SCL) measurement procedures and assigned radiometric uncertainties, provide a validation of their radiometric scale at the time of the comparison. Additionally, the maintenance of the radiance scale was evaluated by use of independent, long-term, multiyear radiance validation measurements of the Ames sphere source. This series of measurements provided an independent assessment of the radiance values assigned to integrating sphere sources by the Ames SCF. Together, the measurements validate the SCF radiometric scale and assigned uncertainties over the time period from September 1999 through July 2003.  相似文献

Changes in salivary composition correlate with disease susceptibility, disease state, or both. However, use of saliva for diagnostic purposes is complicated by the gland-specific effects of circadian rhythm or diurnal variation. We recently characterized a suite of peptides in the < or =10-kDa fraction of human parotid saliva that included many novel species. In this study, we used novel iTRAQ labeling chemistry to investigate possible diurnal effects on peptide generation. We collected samples produced by gustatory stimulation as the ductal secretions at four time points under conditions that minimized proteolysis, pooled them according to collection time, and isolated the LMW fractions. Samples collected at each collection time were derivatized with a different isobaric iTRAQ reagent. The labeled samples were combined, separated by reversed-phase HPLC, co-spotted with matrix on MALDI targets, and analyzed by MALDI TOF/TOF mass spectrometry. With this approach, we achieved relative quantification of the parotid peptides at four time points. In several cases, abundance during the day changed dramatically. iTRAQ tagging improved the efficiency of MS/MS fragmentation, which in turn allowed the identification of several novel peptides. Our results demonstrated both the utility of this method and the importance of diurnal effects on the composition of the human parotid saliva peptidome.  相似文献

Analyte protectants were previously defined as compounds that strongly interact with active sites in the gas chromatographic (GC) system, thus decreasing degradation, adsorption, or both of coinjected analytes. In this study, we evaluated various combinations of promising analyte protectants for the volatility range of GC-amenable pesticides using GC/quadrupole mass spectrometry (MS) and 1-microL hot splitless injection for sample introduction. A mixture of ethylglycerol, gulonolactone, and sorbitol (at 10, 1, and 1 mg/mL, respectively, in the injected samples) was found to be the most effective in minimizing losses of susceptible analytes and significantly improving their peak shapes (due to reduction of peak tailing). When added to final sample extracts and matrix-free calibration standards alike, these analyte protectants induced a similar response enhancement in both instances, resulting in effective equalization of the matrix-induced response enhancement effect even after a large number of fruit and vegetable extract injections. As compared to matrix-matched standardization, the analyte protectant approach offers a more convenient solution to the problems associated with calibration in routine GC/MS analysis of pesticide residues and possibly other susceptible analyte types in diverse samples. Moreover, the use of analyte protectants also substantially reduced another adverse matrix-related effect caused by gradual build-up of nonvolatile matrix components in the GC system, thus improving ruggedness and, consequently, reducing need for frequent maintenance.  相似文献

The aim of this study was to examine the cardio-respiratory effects of voluntary hyperpnoea using a respiratory muscle trainer (RMT) with three different sized rebreathing bags. In particular, the effects of hyperpnoea on inspired and end-tidal gas concentrations were determined. Seven adult males completed three 30 min bouts of hyperpnoea using optimal, oversized and undersized rebreathing bags. Inspired (F(I)) and expired end-tidal (F(ET)) O2 and CO2 concentrations, arterial O2 saturation (S(AO2)) and heart rate were measured during hyperpnoea. Before and after a bout of hyperpnoea, pulmonary function and blood pressure (BP) were assessed. Data were analysed using a two-way repeated-measures ANOVA, with p < 0.05 considered significant. Three subjects experienced discomfort during hyperpnoea and stopped after 20 min. During hyperpnoea, the F(ETCO2) was maintained at 4.6 +/- 0.7% irrespective of bag size. The increase in F(ICO2) over time reached 0.5 +/- 0.5% at 20 min. The F(IO2) fell to 19.4 +/- 0.8% at 20 min, and S(AO2) decreased to 97%. Heart rate and systolic BP increased slightly, but independently of rebreathing bag volume. No changes in pulmonary function or diastolic BP were found. It is concluded that the RMT maintained a constant F(ETCO2) at the expense of a mild hypoxia. The acute effects of hyperpnoea on the cardio-respiratory system are generally mild, but not always tolerable for 30 min.  相似文献

A software has been developed that is aimed at quantitative analysis of images acquired by isoelectric focusing and double blotting procedures used for recombinant erythropoietin doping control. It represents a unified and easy-to-use tool for Epo doping experts in WADA accredited laboratories. It is based on image segmentation philosophy that enables identification of individual bands whose characteristics are needed for evaluation of the Epo doping positivity criteria. Several modules implemented in the GASepo software include an original know how, in particular, the method of robust calculation of the cut-off line, band segmentation and classification algorithms. GASepo is being used in several doping control laboratories worldwide.  相似文献
