A mixture of oil/ethanol (1∶3, w/w) was shaken at 30°C with 4% immobilized Candida antarctica lipase by weight of the reaction mixture. The reaction regiospecifically converted FA at the 1- and 3-positions to FA ethyl esters, and the lipase acted on C14−C24 FA to a similar degree. The content of 2-MAG reached a maximum after 4 h; the content was 28–29 mol% based on the total amount of FA in the reaction mixture at 59–69% ethanolysis. Only 2-MAG were present in the reaction mixture during the first 4 h, and 1(3)-MAG were detected after 7 h. After removal of ethanol from the 4-h reaction mixture by evaporation, 2-MAG were fractionated by silica gel column chromatography. The contents of FA in the 2-MAG obtained by ethanolysis of several oils coincided well with FA compositions at the 2-position, which was analyzed by Grignard degradation. It was shown that ethanolysis of oil with C. antarctica lipase can be applied to analysis of FA composition at the 2-position in TAG.  相似文献

TAG (MLM) with medium-chain FA (MCFA) at the 1,3-positions and long-chain FA (LCFA) at the 2-position, and TAG (LMM) with LCFA at the 1(3)-position and MCFA at 2,3(1)-positions are a pair of TAG regioisomers. Large-scale preparation of the two TAG regioisomers was attempted. A commercially available FFA mixture (FFA-CLA) containing 9-cis, 11-trans (9c, 11t)- and 10t,12c-CLA was selected as LCFA, and caprylic acid (C8FA) was selected as MCFA. The MLM isomer was synthesized by acidolysis of acyglycerols (AG) containing two CLA isomers with C8FA: A mixture of AG-CLA/C8 FA (1∶10, mol/mol) and 4 wt% immobilized Rhizomucor miehei lipase was agitated at 30°C for 72 h. The ratio of MLM to total AG was 51.1 wt%. Meanwhile, LMM isomer was synthesized by acidolysis of tricaprylin with FFA-CLA: A mixture of tricaprylin/FFA-CLA (1∶2, mol/mol) and 4 wt% immobilized R. miehei lipase was agitated at 30°C for 24 h. The ratio of LMM to total AG was 51.8 wt%. MLM and LMM were purified from 1,968 and 813 g reaction mixtures by stepwise short-path distillation, respectively. Consequently, MLM was purified to 92.3% with 49.1% recovery, and LMM was purified to 93.2% with 52.3% recovery. Regiospecific analyses of MLM and LMM indicated that the 2-positions of MLM and LMM were 95.1 mol% LCFA and 98.3 mol% C8 FA, respectively. The results showed that a process comprising lipase reaction and short-path distillation is effective for large-scale preparation of high-purity regiospecific TAG isomers.  相似文献

Specialist bacteria can synthesize nanoparticles from various metal ions in solution. Metal recovery with high efficiency can be achieved by metal-tolerant microorganisms that proliferate in a concentrated metal solution. In this study, we isolated bacteria (Pseudomonas sp. strain KKY-29) from a bacterial library collected from water near an abandoned mine in Komatsu City, Ishikawa Prefecture, Japan. KKY-29 was maintained in nutrient medium with lead acetate and synthesized hydrocerussite and pyromorphite nanoparticles inside the cell; KKY-29 also survived nanoparticle synthesis. Quantitative PCR analysis of genes related to phosphate metabolism showed that KKY-29 decomposed organic phosphorus to synthesize lead phosphate. KKY-29 also deposited various metal ions and synthesized metal nanoparticles when incubated in various metal salt solutions other than lead. The present study considers the development of biotechnology to recover lead as an economically valuable material.  相似文献

As tongue cancer is one of the major malignant cancers in the world, understanding the mechanism of maintenance of lingual epithelial tissue, which is known to be the origin of tongue cancer, is unquestionably important. However, the actual stem cells that are responsible for the long-term maintenance of the lingual epithelium have not been identified. Moreover, a simple and convenient culture method for lingual epithelial stem cells has not yet been established. Recently, we have shown that Bmi1-positive cells, residing at the second or third layer of the epithelial cell layer at the base of the interpapillary pit (IPP), were slow-cycling and could supply keratinized epithelial cells for over one year, indicating that Bmi1-positive cells are long-term lingual epithelial stem cells. In addition, we have developed a novel lingual epithelium organoid culture system using a three-dimensional matrix and growth factors. Here, we discuss current progress in the identification of lingual stem cells and future applications of the lingual culture system for studying the regulatory mechanisms of the lingual epithelium and for regenerative medicine.  相似文献

We developed a novel spherical carbon material. The spherical carbon is composed of a high density of carbon nanotubes or nanofilaments, and includes an oxidized diamond particle as a core. Syntheses of this carbon in high volume with high selectivity may be possible. The hollow cylinder shear test is somewhat controversial due to its non-uniform stresses and strains, and has been analyzed by simple theoretical methods and two-dimensional FE calculations. In this paper, the hollow cylinder test under strain control was carried out numerically by treating the specimen as a three-dimensional initial-boundary value problem considering the inertial forces. At first, besides the known nonuniform strain, the non-uniformities of excess pore water pressure and overconsolidation ratio have been shown to benefit from a soil–water coupled analysis that employs the SYS Cam-clay model. Then, the influence of the specimen geometries, including wall thicknesses, heights and outer diameters on the non-uniformity was investigated sequentially. A new method for evaluating non-uniformity was proposed, which is suitable for the three-dimensional analysis. The response under a uniform deformation field, which is indicated by “the perfect path”, was presented to draw a comparison with the apparent behaviors, with non-uniformities taken into consideration. It should be noted that there is a critical height to prevent failure at the specimen ends according to the apparent behavior. Finally, the torque-controlled experiment indicated that 4 ribs could not transfer the torque reliably while 6 or 8 ribs were feasible.  相似文献

This paper presents results from a behavioral demand response trial targeted to both grid and residential peak hours, in which weekly feedback via paper-based reports, real-time feedback via an in-home display, 30-minute tiered rate, and email prompts are adopted to almost 230 residential customers of a condominium in Funabashi, a city located in Greater Tokyo. Through a randomized experiment, we find that the peak saving impact during grid peak hours (1–4 pm, weekdays only) was 11.6 %, given that all the four interventions provided all at once. In addition, the results show that the variation in peak saving effects by household characteristics exists, and the variation differs among packages of peak saving interventions. Furthermore, we analyzed how much informational elements in weekly reports are considered as useful by residential customers. These results suggest that feedback-based approaches for peak saving can promote households’ energy conservation behavior.  相似文献

We investigate deformation-induced martensitic transformation behavior in cold-rolled and cold-drawn specimens of type 316 stainless steel. Deformation-induced martensite preferentially nucleates at the twin boundary between the austenite matrix and a deformation twin. In the cold-rolled specimen, martensite formed at the twin boundary has a Kurdjumov–Sachs (K–S) relationship with both the austenite matrix and the deformation twin (“double K–S relationship”). In the cold-drawn specimen, two kinds of deformation twins with different twin planes are typically formed, and therefore deformation-induced martensites are formed where the deformation twin boundaries intersect: martensite thus has an imperfect “triple K–S relationship” with the austenite matrix and the two deformation twins. The complicated crystallographic orientation relationship between austenite and martensite grains strongly restricts the formation of some variants of deformation-induced martensites. Because of the difference in number of nucleation sites in the cold-drawn and cold-rolled specimens, martensitic transformation is more enhanced in the former than in the latter.  相似文献

The conditions of convergence in a modified Monte Carlo power iteration method to generate the eigenfunction with the second largest criticality eigenvalue, which was originally proposed by Booth, have been defined with a different approach. In this work, the first and second eigenvectors composed of two volume-integrated fission source intensities defined in two-partitioned regions are used for deriving the convergence conditions. The conditions of convergence as shown by Booth are found to be true in the limit of a small amplitude of the first eigenfunction. Following the method that uses two estimates of the second eigenvalue defined in two-partitioned regions, a new method for removing the fundamental mode eigenfunction from the fission source distributions has been developed. Because of the explicit removal of the first eigenfunction, the validity of this method is convincing as a technique for obtaining the second eigenfunction. Although this method needs the first eigenfunction and eigenvalue, and the subtraction of the first eigenfunction from the fission source distribution, it has the advantage that the adjoint mode calculation which is in general difficult for continuous energy Monte Carlo codes is not required.  相似文献
