Vein wall inflammation associated with venous thrombosis is mediated by an imbalance in proinflammatory as compared with antiinflammatory molecules. We hypothesize that IL-10 is an important antiinflammatory cytokine that influences vein wall inflammation and thrombus propagation during venous thrombosis. To test this hypothesis a model of inferior vena caval thrombosis was used. Studies were performed at sacrifice 2 days after thrombus induction and included leukocyte morphometrics, myeloperoxidase activity, vein wall permeability, thrombus weight, and IL-10 ELISA analysis from the vein wall. IL-10 was elevated in the vein wall during venous thrombosis. Neutralization of IL-10 increased inflammation, while supplementation with rIL-10 demonstrated a dose- and time-dependent decrease in inflammation. Interestingly, a low 2.5-microg rIL-10 dose given at time of initiation of thrombosis most significantly decreased inflammation. Thrombus weight was importantly diminished by reconstitution of IL-10. These studies support an important role for IL-10 in the regulation of thrombus-associated inflammation and thrombosis and suggest that IL-10 could be used as a therapeutic agent in the treatment of venous thrombosis.  相似文献

Using an isolated, blood-perfused atrium preparation, the effects of temperature on SA nodal pacemaker activity were investigated in 9 preparations. The PP interval decreased as temperature was raised. Regular sinus rhythm and atrial contraction were maintained above approximately 26 degrees C. Below 26 degrees C, sinus depolarization still showed a regular rate, although atrial contractions had ceased. At about 24 degrees C, atrial rhythm became irregular. Below 20 degrees C, atrial depolarization disappeared. Chronotropic responses to norepinephrine were suppressed at decreased temperatures, not only with respect to maximum PP shortening but also to the threshold dose for inducing sinus acceleration. Overdrive suppression was not influenced significantly by decreasing temperature. These results indicate that a temperature decrease causes suppression of SA nodal pacemaker activity, although the SA node continues to function regularly until about 25 degrees C.  相似文献

The variation in the length of gestation, the period from mating until parturition, was studied in 77 dogs of different breeds; the time for mating was determined by measuring peripheral blood progesterone levels. The mean length of gestation was 62.1 +/- 0.2 (S.E.M.) days, with a variation of 11 days. The number of pups appeared to influence the length of gestation. Length of gestation was negatively correlated (r = -0.96, P < 0.001, n = 44) with litter size in litters with 7 or fewer pups. The intra-breed variation in length of gestation in the five breeds represented by five or more bitches was 3-6 days. The mean gestation of Alsatians (60.1 +/- 0.5, n = 9) was shorter (P < 0.005) than that of the other breeds combined (62.3 +/- 0.3, n = 68). The primiparous/multiparous status of the bitch did not influence the length of gestation.  相似文献

We propose a model to explain experimentally observed effects of Gz onset rates and levels on the time of occurrence of loss of vision and/or consciousness. The model is based on the existence of two generally accepted parameters: a G limit beyond which cerebral perfusion ceases, and a buffer time between loss of perfusion and loss of function. When applied to ramp onset G profiles, the model predicts a generally hyperbolic locus of endpoints, similar to the well-known Stoll curve, except for the dip. The advantage of the model is its applicability to any G onset profile. Data from the literature support the assumptions of the model and its results, including the absence of the dip in the locus for a ramp onset. The results call into question some concepts used to design G avoidance inflight strategies and the usefulness of some experimental centrifuge methods. The model may enable an increase in the accuracy of predictions of the time of visual or cerebral loss of function under various G profiles.  相似文献
