Rapidly increasing information about the human genome requires a fast and simple method for the detection of single-nucleotide polymorphisms (SNPs). To date, the conventional SNP detection technologies have been unable to identify all possible SNPs and needed further development in cost, speed, and sensitivity. Here we describe a novel method to discriminate and code all possible combinations. SNPs were coded by monitoring the changes in the electrochemical signal of the monobase-modified colloidal gold (Au) nanoparticles. First, a chitosan layer was formed on the alkanethiol self-assembled monolayer-modified Au nanoparticle. The monobases were then attached onto the chitosan-coated Au nanoparticles through their 5' phosphate group via the formation of a phosphoramidate bond with the free amino groups of chitosan. The size of the surface-modified Au nanoparticle was found to be 8.46 +/- 1.53 nm by using atomic force microscopy. If there is a SNP in DNA and the mismatched bases are complementary to the monobase, Au nanoparticles accumulate on the electrode surface in the presence of DNA polymerase I (Klenow fragment), thus resulting in a significant change in the Au oxide wave. In this report, monobase-modified Au nanoparticles show not only the presence of a SNP, but also identify which bases are involved within the pair. Especially, the identification of a transversion SNP, which contains a couple of the same pyrimidine or purine bases, is greatly simplified. A model study was performed by using a synthetic 21-base DNA probe related to tumor necrosis factor (TNF-alpha) along with its all possible mutant combinations. This versatile nanoparticle-based electrochemical protocol is a promising candidate for coding all mutational changes.  相似文献

In this paper, we demonstrated optical fiber-based stereolithography of alumina ceramics using concentrated slurry with low monomer content for enabling rapid firing of the printed green objects. Photo-cross-linkable alumina slurries were designed using a partial complex of polyethyleneimine and oleic acid (PEI-OA) as reactive polymer dispersants and multifunctional acrylates (MA) as monomers. The effects of the processing parameters, such as oleic acid contents, MA contents, particle contents, and particle sizes, on photocuring properties were systematically investigated. We found that the reactive amine sites of PEI-OA attached to particles and MA volume concentrations in the liquid phase were the most dominant factors to improve the photocuring properties. Therefore, a slurry composed of larger alumina particles with higher solid contents and PEI-OA with lower OA contents was more favorable for achieving photocuring under reduced MA content. The designed photocurable slurry was then applied to ceramic stereolithography using optical fiber. 3D alumina green parts were drawn by passing blue laser beam through an optical fiber in a droplet of photocuring slurry and dense sintered alumina were successfully fabricated by rapid drying (<10 min) and debinding (<1 h) without any structural collapse.  相似文献

A broader characterization of industrial wastewaters, especially in respect to hazardous compounds and their potential toxicity, is often necessary in order to determine the best practical treatment (or pretreatment) technology available to reduce the discharge of harmful pollutants to the environment or publicly owned treatment works. Using a toxicity-directed approach, this paper sets the base for a rational treatability study of polyester resin manufacturing. Relevant physical and chemical characteristics were determined. Respirometry was used for toxicity reduction evaluation after physical and chemical effluent fractionation. Of all the procedures investigated, only air stripping was significantly effective in reducing wastewater toxicity. Air stripping in pH 7 reduced toxicity in 18.2%, while in pH 11 a toxicity reduction of 62.5% was observed. Results indicated that toxicants responsible for the most significant fraction of the effluent's instantaneous toxic effect to unadapted activated sludge were organic compounds poorly or not volatilized in acid conditions. These results led to useful directions for conducting treatability studies which will be grounded on actual effluent properties rather than empirical or based on the rare specific data on this kind of industrial wastewater.  相似文献

This paper describes a fast rendering algorithm for verification of spectacle lens design. Our method simulates refraction corrections of astigmatism as well as myopia or presbyopia. Refraction and defocus are the main issues in the simulation. For refraction, our proposed method uses per-vertex basis ray tracing which warps the environment map and produces a real-time refracted image which is subjectively as good as ray tracing. Conventional defocus simulation was previously done by distribution ray tracing and a real-time solution was impossible. We introduce the concept of a blur field, which we use to displace every vertex according to its position. The blurring information is precomputed as a set of field values distributed to voxels which are formed by evenly subdividing the perspective projected space. The field values can be determined by tracing a wavefront from each voxel through the lens and the eye, and by evaluating the spread of light at the retina considering the best human accommodation effort. The blur field is stored as texture data and referred to by the vertex shader that displaces each vertex. With an interactive frame rate, blending the multiple rendering results produces a blurred image comparable to distribution ray tracing output.  相似文献

The effect of the flash event (FE) on microcrack healing behavior in 8 mol% yttria-stabilized zirconia was examined at healing temperatures of 1040 and 1230°C under the direct and alternating (DC and AC) electric fields. The crack healing behavior changed depending on the factors of the electric field, healing temperature, and crack length. Although the crack healing proceeded with the temperature, the healing rate increased with the crack length, suggesting that the external energy stored as crack surface energy would provide a driving force for the crack healing. Although the crack healing occurs even under the static annealing without the electric field, the healing rate was accelerated by FE significantly more under the AC field than under the DC field. The microcracks with a length of ≈20 μm were fully healed at 1230°C only for 10 min by the FE treatment under the AC field, and the flash healing behavior was four times faster than that of the static annealing. These results suggest that the enhanced healing behavior cannot be explained only by thermal effects, and the accelerated diffusivity caused additionally by nonthermal effect under FE might contribute to the enhanced healing behavior, especially in the AC electric field.  相似文献

The effects of the co-doping and the resultant co-segregation of 2 mol% TiO2 and 2 mol% GeO2 on the ionic conductivity and on the chemical bonding state in a tetragonal ZrO2 polycrystal were investigated. The conductivity data and grain boundary microstructure showed that the doped Ti4+ and Ge4+ cations segregate along the grain boundary, and this segregation causes a reduction in the conductivity of both the grain interior and grain boundary and an increase in the activation energy of the grain boundary conductivity. Overall, the data indicate that the segregation retards the diffusion of oxygen anions. A first-principle molecular orbital calculation explains the retarded diffusion of the oxygen anion from a change in the covalent bonds around the dopant cations; an increase in the strength of the covalent bond between the oxygen and doped cation should work to suppress the diffusion of the oxygen anion.  相似文献

A number of investigations have examined the impact of the use of biodiesel on the emissions of carbon dioxide and regulated emissions, but limited information exists on the chemical composition of particulate matter from diesel engines burning biodiesel blends. This study examines the composition of diesel particulate matter (DPM) emissions from a commercial agriculture tractor burning a range of biodiesel blends operating under a load that is controlled by a power take off (PTO) dynamometer. Ultra-low sulfur diesel (ULSD) fuel was blended with soybean and beef tallow based biodiesel to examine fuels containing 0% (B0), 25% (B25), 50% (B50), 75% (B75), and 100% (B100) biodiesel. Samples were then collected using a dilution source sampler to simulate atmospheric dilution. Diluted and aged exhaust was analyzed for particle mass and size distribution, PM2.5 particle mass, PM2.5 organic and elemental carbon, and speciated organic compounds. PM2.5 mass emissions rates for the B25, B50, and B75 soybean oil biodiesel mixtures had 20%–30% lower emissions than the petroleum diesel, but B100 emissions were about 40% higher than the petroleum diesel. The trends in mass emission rates with the increasing biodiesel content can be explained by a significant decrease in elemental carbon (EC) emissions across all blending ranges and increasing organic carbon (OC) emissions with pure biodiesel. Beef tallow biodiesel blends showed similar trends. Nevertheless, it is important to note that the study measurements are based on low dilution rates and the OC emissions changes may be affected by ambient temperature and different dilution conditions spanning micro-environments and atmospheric conditions. The results show that the use of biodiesel fuel for economic or climate change mitigation purposes can lead to reductions in PM emissions and a co-benefit of EC emission reductions. Detailed speciation of the OC emissions were also examined and are presented to understand the sensitivity of OC emissions with respect to biodiesel fuel blends.

The aim of this study was to examine whether salivary exosomal miRNAs could be identified as aging biomarkers. Fifteen young healthy volunteers (median age, 21.0 years) and 13 old individuals (median age, 66.0 years) were recruited. Unstimulated whole saliva was collected, salivary exosomes were isolated, and total RNA was extracted. In a microarray, 242 miRNAs were commonly detected in these two mixed samples. Based on the cut-off values of 2- or 0.5-fold changes (FC) and regulatory power for aging process, six candidate miRNAs (miR-24-3p, miR-371a-5p, miR-3175, miR-3162-5p, miR-671-5p, and miR-4667-5p) were selected. After comparing each total RNA obtained by the 15 young and 13 old individuals to validate the FC values using quantitative real-time PCR, miR-24-3p was identified as a novel candidate aging biomarker. This pilot study suggested that salivary exosomal miRNAs could be identified as candidate aging biomarkers. To confirm whether miR-24-3p in salivary exosomes are suitable biomarkers of aging, further validation research is required.  相似文献

With the aid of the transient hot‐wire method, the thermal conductivity of molten B2O3, B2O3–SiO2, Na2O–SiO2, and Na2O–B2O3 systems was measured along with their temperature and composition. It was observed that the thermal conductivity of pure B2O3 increased with temperature, until about 1400 K, and then decreased subsequently. Using the MAS‐NMR, 3Q‐MAS, and Raman spectroscopy, the structure of B2O3 and SiO2 in the B2O3–SiO2 system was confirmed. Findings show that an addition of B2O3 into the pure SiO2 system causes a significant decrease in thermal conductivity, due to the formation of boroxol rings. The thermal conductivity of the Na2O–SiO2 system was measured and its phonon mean free path was calculated. In addition, a positive linear relation between viscosity and thermal conductivity was observed. In the Na2O–B2O3 system, it was found that a change in the relative fraction of 4‐coordinated boron has an influence on the thermal conductivity when the concentration of Na2O is between 10 and 30 mol%, in which case the tetraborate unit is dominant.  相似文献

Using the transient hot‐wire method, the thermal conductivity properties of the molten Li2O–B2O3 and K2O–B2O3 systems were measured. The thermal conductivity increases with decreasing the temperature due to the borate structure change. In addition, calculations of the one‐dimensional Debye temperature and the phonon mean free paths as a function of temperature of the alkali borate systems were made. At a fixed temperature of 1273 K, the effect of the alkali oxide concentration on the thermal conductivity was evaluated. Within a range of 10–30 mol% Li2O (or K2O), a positive relationship between the thermal conductivity and 4‐coordinate boron was obtained. However, below 10 mol% Li2O (or K2O), the change in the intermediate‐range order of the borate structure had a more dominant effect on the thermal conductivity. Finally, the effect of cations on the thermal conductivity in the various molten R2O–B2O3 (R=Li, Na and K) systems was considered. Depending on the type of cation, the change in the ionization potential had an effect on the thermal conductivity and also resulted in a change in the bond strength.  相似文献
