Environmental pests may serve as reservoirs and vectors of zoonotic pathogens to leafy greens; however, it is unknown whether insect pests feeding on plant tissues could redistribute these pathogens present on the surface of leaves to internal sites. This study sought to differentiate the degree of tissue internalization of Escherichia coli O157:H7 when applied at different populations on the surface of lettuce and spinach leaves, and to ascertain whether lettuce-infesting insects or physical injury could influence the fate of either surface or internalized populations of this enteric pathogen. No internalization of E. coli O157:H7 occurred when lettuce leaves were inoculated with 4.4 log CFU per leaf, but it did occur when inoculated with 6.4 log CFU per leaf. Internalization was statistically greater when spinach leaves were inoculated on the abaxial (underside) than when inoculated on the adaxial (topside) side, and when the enteric pathogen was spread after surface inoculation. Brief exposure (~18 h) of lettuce leaves to insects (5 cabbage loopers, 10 thrips, or 10 aphids) prior to inoculation with E. coli O157:H7 resulted in significantly reduced internalized populations of the pathogen within these leaves after approximately 2 weeks, as compared with leaves not exposed to insects. Surface-contaminated leaves physically injured through file abrasions also had significantly reduced populations of both total and internalized E. coli O157:H7 as compared with nonabraded leaves 2 weeks after pathogen exposure.  相似文献

Animal studies suggest that perfluorocarbons (PFCs) may alter sexual maturation. Relationships of human PFC exposure with puberty are not clear. We conducted a cross-sectional study to investigate whether perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) were associated with indicators of sexual maturation in a 2005-2006 survey of residents with PFOA water contamination from the Mid-Ohio Valley. Participants were 3076 boys and 2931 girls aged 8-18 years. They were classified as having reached puberty based on either hormone levels (total >50 ng/dL and free >5 pg/mL testosterone in boys and estradiol >20 pg/mL in girls) or onset of menarche. We estimated the odds of having reached puberty classified by these criteria and the fitted median age of reaching puberty in relation to serum PFOA and PFOS concentrations measured when puberty status was assigned. For boys, there was a relationship of reduced odds of reached puberty (raised testosterone) with increasing PFOS (delay of 190 days between the highest and lowest quartile). For girls, higher concentrations of PFOA or PFOS were associated with reduced odds of postmenarche (130 and 138 days of delay, respectively). In conclusion, our study showed a later age of puberty in this population correlated with PFC concentrations.  相似文献

BACKGROUND: A transgenic papaya line (TPY10‐4) that is resistant to both papaya ringspot virus (PRSV) and papaya leaf distortion mosaic virus (PLDMV) has been developed in Taiwan. This study investigated the immunomodulatory properties of transgenic TPY10‐4 and its native (TCK) papaya fruits using an ovalbumin (OVA)‐sensitised mouse model. Both green and ripe papaya fruits at low (0.2 g powder kg?1 body weight (BW)) and high (1.6 g powder kg?1 BW) doses were administered to experimental mice by intragastric gavage for 5 weeks. Changes in serum total immunoglobulin A (IgA), IgE, IgG and IgM levels, OVA‐specific IgE, IgG1 and IgG2a titres and Th1/Th2 cytokine secretions using splenocytes were determined. RESULTS: Transgenic TPY10‐4 or native TCK papaya fruit supplementation did not significantly affect body, visceral organ and relative tissue weights, total IgE antibody levels, OVA‐specific IgE and IgG1 antibody titres or OVA‐stimulated interferon‐γ (IFN‐γ), interleukin‐2 (IL‐2), IL‐4, IL‐5 and IL‐10 secretions using splenocytes. However, transgenic papaya fruits markedly increased serum total IgM levels. CONCLUSION: This study suggests that transgenic TPY10‐4 papaya fruits do not increase the allergenic potential of OVA by oral administration but may have a protective immunity via increasing the serum total IgM level. The responses to fertiliser‐N of winter wheat and winter barley grown on sandy soils were measured in 72 experiments in England from 1990 to 1994. Yield without fertiliser‐N (Y0) was c 1.1 t ha−1 greater following root crops than following cereals. Following potato crops given organic manures, Y0 was c 1.2 t ha−1 greater than following unmanured potato crops, but Y0 was no greater following sugarbeet to which organic manures had been applied. Only after the two driest winters was there sufficient variation in soil N supply in spring (SNSs) for this to show a relationship with Y0. However, Y0 increased with increasing N mineralisation during the growing season (AM) in the three years it was measured. There was no consistent effect of sowing date on Y0. Following potatoes, yield at optimum fertiliser‐N (Yopt) decreased as sowing date was delayed, but this was not so after cereals, sugarbeet or overall. There was no increase in Yopt with SNSS or AM, but Yopt decreased with increasing moisture stress (S) in June. The mean yield response to NoptY) was c 0.4 and 0.8 t ha−1 smaller following potatoes and sugarbeet respectively than following cereals, but not consistently so as there were large interactions between site, year and previous crop. Following root crops, ΔY was c 0.6 and 1.4 t ha−1 less after sugarbeet and potatoes respectively that had been given organic manures. Without the addition of organic manures, ΔY following potatoes was similar to that following cereals. Regression on SNSS and AM accounted for 28 and 15% respectively of the variance in ΔY. The optimum economic fertiliser‐N application (Nopt) was similar, at c 140 kg ha−1, following cereals and potatoes. Following sugarbeet, cereal Nopt was only c 110 kg ha−1. The differences according to previous crop reported here are consistent with mineralisation of crop residues on sandy soils being more rapid than on other soils; the potato residues were rapidly mineralised in autumn and lost by leaching over winter. Residues from later‐harvested sugarbeet were mineralised during the growing season of the subsequent cereal crop. Fertiliser‐N requirements were, at c 110–140 kg ha−1, smaller than has been found on other soil types, and less than current recommendations for wheat. Requirements were significantly reduced in years of drought stress. No differences were found in Nopt between wheat and barley. These data do not justify the current advice to invariably reduce fertiliser‐N to cereals following potatoes by 20–25 kg ha−1 on these sandy soils. On average a reduction of c 20 kg ha−1 could be made following sugarbeet, with a further reduction of c 40 kg ha−1 N if manures had been applied to the previous sugarbeet crop. A reduction of 40 kg ha−1 N could also be made where cereals followed a potato crop to which manures had been applied. Further refinements on the basis of measurements of soil mineral N could not be justified. Seasonal variation in N response due to drought stress makes recommendations difficult on these soils. Adopting the fertiliser‐N recommendations proposed here would produce N surpluses to the soil of c 37, 10 and 27 kg ha−1 respectively following cereals, sugarbeet and potatoes when cereal grain is removed but straw incorporated. On farms where straw is removed, N surplus would be largely eliminated. Our recommendation that no reduction in fertiliser‐N application to cereal crops grown on sandy soils should be made following potatoes will not increase fertiliser‐N use and is not expected to increase nitrate leaching. Some reduction in nitrate leaching may be achieved if recommendations following cereal crops and sugarbeet are made in accordance with the results reported here. © 2000 Society of Chemical Industry  相似文献

The potential of mid-infrared (MIR) and near-infrared (NIR) spectroscopy for their ability to differentiate between apple juice samples on the basis of apple variety and applied heat-treatment was evaluated. The heat-treatment involved exposure of juice samples (15 ml) for 30 s in a 900 W microwave oven and the apple varieties used to produce the juice samples were Bramley, Elstar, Golden Delicious and Jonagold. The chemometric procedures applied to the MIR and NIR data were partial least squares regression (PLS1 for differentiation on the basis of heat-treatment, PLS2 for varietal differentiation) and linear discriminant analysis (LDA) applied to principal component (PC) scores. PLS1 and PLS2 gave the highest level of correct classification of the apple juice samples according to heat-treatment (77.2% for both MIR and NIR data) and variety (78.3–100% for MIR data; 82.4–100% for NIR data), respectively.  相似文献

Herbal remedies are increasing in popularity as treatments for metabolic conditions such as obesity and Type 2 Diabetes. One potential therapeutic option is fenugreek seeds (Trigonella foenum-graecum), which have been used for treating high cholesterol and Type 2 diabetes. A proposed mechanism for these benefits is through alterations in the microbiome, which impact mammalian host metabolic function. This study used untargeted metabolomics to investigate the fenugreek-induced alterations in the intestinal, liver, and serum profiles of mice fed either a 60% high-fat or low-fat control diet each with or without fenugreek supplementation (2% w/w) for 14 weeks. Metagenomic analyses of intestinal contents found significant alterations in the relative composition of the gut microbiome resulting from fenugreek supplementation. Specifically, Verrucomicrobia, a phylum containing beneficial bacteria which are correlated with health benefits, increased in relative abundance with fenugreek. Metabolomics partial least squares discriminant analysis revealed substantial fenugreek-induced changes in the large intestines. However, it was observed that while the magnitude of changes was less, significant modifications were present in the liver tissues resulting from fenugreek supplementation. Further analyses revealed metabolic processes affected by fenugreek and showed broad ranging impacts in multiple pathways, including carnitine biosynthesis, cholesterol and bile acid metabolism, and arginine biosynthesis. These pathways may play important roles in the beneficial effects of fenugreek.  相似文献

The finite-element method is used to determine the scattering matrices of open devices in two dimensions. Microwave and optical devices in which the fields are not confined to a finite region can be analyzed with the finite-element method if special boundary conditions are used to absorb outgoing radiation. The absorbing boundary conditions can be imposed by the addition of two terms to the usual functional for the scalar Helmholtz equation. Universal matrices are introduced to allow the additional terms to be easily assembled, for polynomial orders one through four. Results are given for the impedance of a parallel-plate waveguide radiating into free space and for the scattering parameters of three dielectric slab waveguide devices: a rectangular discontinuity, a feed structure, and a junction  相似文献
