The biomimetic coating method was used for fabricating calcium phosphate fibres for biomedical applications such as bone defect fillers. Natural cotton substrate was pre-treated with phosphorylation and a Ca(OH)2 saturated solution. The pre-treated samples were then soaked in simulated body fluid (SBF) of two different concentrations, 1.5 times and 5.0 times the ion concentration of blood plasma. The cotton was then burnt out via sintering of the ceramic coating at 950C, 1050C, 1150C, and 1250C. The results demonstrated that osteoblastic cells were able to cover the entire surface cotton fibres, and the cell coverage appeared to be independent of surface roughness and Ca/P ratio of fibres.  相似文献

A new generation of platinum nanoelectrodes for constant-distance mode scanning electrochemical microscopy (CD-SECM) has been prepared, characterized, and used for high spatial resolution electrochemical measurements and visualization of electrochemically induced concentration gradients in microcavities. The probes have long (1-2 cm), narrow quartz tips that were conically polished and have a Pt nanoelectrode that is slightly offset from center. Because of the size and location of the electrode on the probe, it does not exhibit SECM feedback while approaching the analyzed sample surfaces even to distances within a few hundred nanometers. The probe was positioned near the surface while scanning and performing electrochemical measurements through use of nonoptical shear force control of the tip-to-sample distance. Test structures consisted of cylindrically shaped microcavities that are 50 microm in diameter with three individually addressable electrodes: a gold disk at 8-microm depth, a crescent-shaped gold ring at 4-microm depth along the wall, and a top gold electrode at the rim. Different electrodes within the microcavity were used to reduce and oxidize redox species in 250 microL of a solution of 5 mM hexaamineruthenium(III) chloride and 0.1 M potassium chloride, protected from evaporation by mineral oil, while the SECM tip followed the topography of the structures and monitored the current from the oxidation of [Ru(NH3)6]2+. Electrochemically generated concentration profiles were obtained from these complex test structures that are not possible with any other SECM technology at this time.  相似文献

Salinization of surface water from sodium chloride (road salt) applied to paved roads is a widely recognized environmental concern in the northern hemisphere, yet practical information to improve winter road management to reduce the environmental impacts of this deicer is lacking. The purpose of our study was to provide such information by developing baseline concentrations for sodium and chloride for lakes in watersheds without paved roads, and then determining the relationship between these ions and density, type, and proximity of paved roads to shoreline. We used average summer (June-September) sodium and chloride data for 138 lakes combined in a watershed based analysis of paved road networks in the Adirondack Park of New York, U.S.A. The watersheds used in our study represented a broad range in paved road density and type, 56 of which had no paved roads. Median lake sodium and chloride concentrations in these 56 watersheds averaged 0.55 and 0.24 mg/L, respectively. In contrast, the median sodium and chloride concentrations for the 82 lakes in watersheds with paved roads were 3.60 and 7.22 mg/L, respectively. Paved road density (lane-km/km2) was positively correlated with sodium and chloride concentrations, but only state roads were significantly correlated with sodium and chloride while local roads were not. State road density alone explained 84 percent of the variation in both ions. We also successfully modeled the relationship between road proximity to shoreline and sodium and chloride concentrations in lakes, which allowed us to identify sections of road that contributed more to explaining the variation in sodium and chloride in lakes. This model and our approach could be used as part of larger efforts to identify environmentally sensitive areas where alternative winter road management treatments should be applied.  相似文献

In this study, 20 young steers received no beta-agonist (C), 100 animals all received zilpaterol hydrochloride (Z), with 1 group only receiving Z while the other 4 groups received zilpaterol and vitamin D3 at the following levels (IU/animal/day) and durations before slaughter: 7 million for 3 days (3D7M); 7 million for 6 days (6D7M); 7 million for 6 days with 7 days no supplementation (6D7M7N) and 1 million for 9 days (9D1M). Left carcass sides were electrically stimulated (ES) and the right side not stimulated (NES). Samples were aged for 3 or 14 days post mortem. Parameters included Warner-Bratzler shear force (WBSF), myofibril filament length, sarcomere length and calpastatin and calpain enzyme activity. Both ES and prolonged aging reduced WBSF (P<0.001). 6D7M, 6D7M7N and Z remained significantly tougher than C (P<0.001), while 3D7M and 9D1M improved WBSF under NES conditions. ES is more effective to alleviate beta-agonist induced toughness than high vitamin D3 supplements.  相似文献

本文探讨了运用python处理食品生物化学实验中还原糖浓度测定的新方法。分别将稀释成不同浓度的还原糖检测样品放在96孔酶标板上,利用扫描仪对其扫描,得到tiff格式的颜色位图,以利用本课题组自行开发的在线分析平台上的RGB分析模块读取样品溶液的RGB颜色数据。通过考察样品取样量、最适图片分辨率等条件,建立一种基于RGB色彩体系的食品中还原糖快速检测方法。检测葡萄糖的结果,在葡萄糖浓度相同,样品取样量为100μL,图片分辨率为1200 dpi的条件下,拟合系数大于0.98;RGB颜色数据与葡萄糖浓度之间的关系准确度更高,该结果与分光光度法结果相近,但本方法更方便、快速、可多通道处理。与此同时,通过测定实际样品中还原糖的含量,并与分光光度法测定结果比对,验证了利用python/RGB色彩数据分析平台快速测定食品中还原糖浓度的可靠性和可行性。  相似文献

Lactobacillus sakei 1 is a food isolate that produces a heat-stable antimicrobial peptide (sakacin 1, a class IIa bacteriocin) inhibitory to the opportunistic pathogen Listeria monocytogenes. Bacterial isolates with antimicrobial activity may be useful for food biopreservation and also for developing probiotics. To evaluate the probiotic potential of L. sakei 1, it was tested for (i) in vitro gastric resistance (with synthetic gastric juice adjusted to pH 2.0, 2.5, or 3.0); (ii) survival and bacteriocin production in the presence of bile salts and commercial prebiotics (inulin and oligofructose); (iii) adhesion to Caco-2 cells; and (iv) effect on the adhesion of L. monocytogenes to Caco-2 cells and invasion of these cells by the organism. The results showed that L. sakei 1 survival in gastric environment varied according to pH, with the maximum survival achieved at pH 3.0, despite a 4-log reduction of the population after 3 h. Regarding the bile salt tolerance and influence of prebiotics, it was observed that L. sakei 1 survival rates were similar (P > 0.05) for all de Man Rogosa Sharpe (MRS) broth formulations when tests were done after 4 h of incubation. However, after incubation for 24 h, the survival of L. sakei 1 in MRS broth was reduced by 1.8 log (P < 0.001), when glucose was replaced by either inulin or oligofructose (without Oxgall). L. sakei 1 was unable to deconjugate bile salts, and there was a significant decrease (1.4 log) of the L. sakei 1 population in regular MRS broth plus Oxgall (P < 0.05). In spite of this, tolerance levels of L. sakei 1 to bile salts were similar in regular MRS broth and in MRS broth with oligofructose. Lower bacteriocin production was observed in MRS broth when inulin (3,200 AU/ml) or oligofructose (2,400 AU/ml) was used instead of glucose (6,400 AU/ml). L. sakei 1 adhered to Caco-2 cells, and its cell-free pH-neutralized supernatant containing sakacin 1 led to a significant reduction of in vitro listerial invasion of human intestinal Caco-2 cells.  相似文献

Capillaries with inner diameters of 550 microm have successfully been packed with 1.5-microm octadecyl silica particles using frits made of macroporous polymers by the UV photopolymerization of a solution of glycidyl methacrylate and trimethylolpropane trimethacrylate. This type of frit is found superior to one made of low-melting point poly(styrene-co-divinylbenzene) beads. Bubble formation is not observed to occur within these capillary columns under our experimental conditions. Separations can be achieved with sample injection volumes as high as 1 microL. To demonstrate its semipreparative use, a mixture of 500 nL of taxol (20 mM) and its precursor, baccatin III (30 mM), is separated using such a column with a Tris buffer.  相似文献
