The pivotal roles of miRNAs in carcinogenesis, metastasis, and prognosis have been demonstrated recently in various cancers. This study intended to investigate the specific roles of hsa-miR-654-5p in lung cancer, which is, in general, rarely discussed. A series of closed-loop bioinformatic functional analyses were integrated with in vitro experimental validation to explore the overall biological functions and pan-cancer regulation pattern of miR-654-5p. We found that miR-654-5p abundance was significantly elevated in LUAD tissues and correlated with patients’ survival. A total of 275 potential targets of miR-654-5p were then identified and the miR-654-5p-RNF8 regulation axis was validated in vitro as a proof of concept. Furthermore, we revealed the tumor-suppressing roles of miR-654-5p and demonstrated that miR-654-5p inhibited the lung cancer cell epithelial-mesenchymal transition (EMT) process, cell proliferation, and migration using target-based, abundance-based, and ssGSEA-based bioinformatic methods and in vitro validation. Following the construction of a protein–protein interaction network, 11 highly interconnected hub genes were identified and a five-genes risk scoring model was developed to assess their potential prognostic ability. There is a great demand for improving the effective utilization of pesticides and reducing their application for sustainable agriculture, and polymeric nanoparticles have provided strong technical support for the efficient delivery of pesticides. To this context, we tried to construct a relatively safe imidaclothiz nano-delivery system for enhanced plant uptake, reduced pesticide residue and improved bioactivity toward green peach aphids. The imidaclothiz could be assembled with the hydrophobic core of SPc through hydrophobic association, which led to the self-assembly of nanoscale imidaclothiz/SPc complex consisting of nearly spherical particles. The SPc decreased the contact angle of imidaclothiz drops and remarkably increased the plant uptake. Furthermore, the bioactivity and control efficacy of imidaclothiz were significantly improved with the help of SPc in both laboratory and field. Excitingly, the residue of imidaclothiz decreased with the help of SPc 7 d after the treatment due to the faster degradation of nanoscale imidaclothiz/SPc complex, which exhibited no negative effects on agronomic traits of tobacco plants. The standing column well for ground source heat pump systems is a promising technology with high efficiency and environmental benefit, where groundwater is drawn from the bottom of a well and then re-injected to its top after transferring heat with heat pumps. Heat transfer analysis of great significance and aquifer involves complex problems. Determining the groundwater seepage flow is a precondition to solve the energy equation describing the heat transfer of the system. Only when piezometric head is obtained, the seepage velocity can be determined according to Darcy’s law. In this article the groundwater seepage flow in an axial symmetrical geometry was studied under the assumption that gross groundwater flow is neglected. An analytical solution of the groundwater seepage flow for a confined aquifer was acquired by using the integral transform method, which may provide a foundation for heat transfer analysis of the standing column well system. 相似文献

诺如病毒(NoVs)是世界范围内引起非细菌性胃肠炎的主要病原体。为研究不同压力条件的超高压处理对牡蛎中NoVs消减控制的影响,将GⅡ.4型NoVs分别置于牡蛎消化腺匀浆和PBS缓冲液中,采用压力分别为200,300,400,500MPa,加压初始温度5℃,保压时间5 min进行处理,并以常压(0.1 MPa)处理为对照组。采用PMAxx-RT-qPCR与透射电子显微镜检测超高压处理后的NoVs,评价不同压力对牡蛎消化腺匀浆和PBS缓冲液中GⅡ.4型NoVs的消减效果;测定超高压处理后牡蛎丙二醛含量和蛋白质巯基含量,评价超高压处理对牡蛎中脂肪和蛋白质的影响。研究结果显示:PMAxx可有效区分感染性NoVs;qPCR检测发现经不同压力的超高压处理后,牡蛎消化腺匀浆和缓冲液中NoVs的RNA拷贝数均显著减少,当压力为500 MPa时,牡蛎和缓冲液中的RNA拷贝数减少量分别>3.49 lg(copies/μL)和>3.61 lg(copies/μL),均降至检测限以下;超高压处理对PBS缓冲液中NoVs的消减效果优于牡蛎消化腺匀浆,还可使牡蛎中脂肪氧化和蛋白质变性,然而,改变程度小... 相似文献
