Bacterial transport experiments were conducted using intact sediment cores collected from sites on the Delmarva Peninsula near South Oyster, VA, to delineate the relative importance of physical and chemical heterogeneity in controlling transport of an adhesion-deficient bacterial strain. Electron microscopy revealed that the sediments consisted of quartz and feldspar with a variable amount of clay and iron and aluminum hydroxide coatings on the grains. A nonmotile, gram-negative indigenous groundwater strain, designated as Comamonas sp. DA001, was injected into the cores along with a conservative tracer bromide (Br). DA001 cells were 1.2 x 0.6 microm in size with a hydrophilic surface and a slightly negative surface charge. Bacterial breakthrough preceded that of Br. This differential advection phenomenon can be accounted for by reduction of the effective porosity for the bacteria relative to Br. The distribution of cells remaining in the core as determined by scintillation counting and phosphor imaging techniques was variable, ranging from nearly uniform concentrations throughout the core to exponentially decreasing concentrations with distance from the point of injection. The fraction of bacterial retention in the core was positively correlated with the abundance of the metal hydroxides and negatively correlated with grain size. Because grain size was inversely correlated with the abundance of the metal hydroxide coatings, it was necessary to separate the effects of grain size and mineralogy. The fraction of the bacterial retention accounting for the effect of grain size, the collision efficiency, exhibited no correlation with the abundance of the metal hydroxides, indicating that the bacterial retention was primarily controlled by grain size. Reasons for the lack of influence of mineralogy on bacterial transport include (i) the slightly negatively charged bacterial surfaces; (ii) an insufficient heterogeneity of sediment surface properties; and (iii) the masking of the positive charge of the metal hydroxide surfaces by adsorbed organic carbon (up to 1180 ppm). This study demonstrates that the laboratory-based bacterial transport experiments are effective in delineating physical versus chemical controlling factors and provide an important link to field-based bacterial transport studies. 相似文献

A stepwise mobilization strategy has been developed for the elution of complex protein mixtures, separated by capillary isoelectric focusing (CIEF) for detection using on-line electrospray ionization mass spectrometry (ESI-MS). Carrier polyampholytes are used to establish a pH gradient as well as to control the electroosmotic flow arising from the use of uncoated fused-silica capillaries. Elution of focused protein zones is achieved by controlling the mobilization pressure and voltage, leaving the remaining protein zones focused inside the capillary. Protein zones are stepwise eluted from the capillary by changing the mobilization conditions. Stepwise mobilization improves separation resolution and simplifies coupling with multistage MS (i.e., MSn) analysis since it allows more effective temporal control of protein elution from the CIEF capillary. We also describe a modified configuration for coupling CIEF with ESI-MS using a coaxial sheath flow interface that facilitate the automation of on-line CIEF-ESI-MS analyses. The stepwise mobilization strategy is demonstrated for the analysis of standard protein mixtures and soluble E. coli lysate proteins using CIEF-ESI-MS. These results indicate that inlet pressure or voltage programming to control the elution of the protein zones from the capillary (i.e., gradient mobilization) may allow for the optimization of the mobilization conditions and provide higher resolution for CIEF separation of complex mixtures with on-line MS. 相似文献

A “fast-drying” method to accelerate the fluid bed drying process is presented. It relies on concepts of heat and mass transfer with real-time near-infrared (NIR) monitoring of moisture. Triplicate trials show that fast drying can reduce granulation drying time by half over single-temperature cycles. The product is equivalent in every way tested to material made using a conventional cycle even though the inlet temperature throughout the constant-rate stage was higher than the melting point of the compound. Tablets made from the fast-dried granulation exhibit equivalent physical characteristics to tablets made from granulations dried at a single, lower temperature. 相似文献

This paper presents a fully integrated PVDF-on-silicon pyroelectric sensor array. The pyroelectric sensor has two main features: a subpixel low noise charge amplifier and a self-absorbing layered structure. The integrated low noise charge amplifier is implemented in a standard CMOS process technology. It is located directly under the sensing structure, maximizing the pixel fill factor. The self-absorbing pyroelectric sensor is a three-layer stack, consisting of a conductive polymer as an absorber layer and front electrode, a thin PVDF film as the pyroelectric material, and a rear metal layer acting as a reflector layer and rear electrode. The manufacture of the pyroelectric sensor array requires five maskless post-CMOS processing steps and is compatible with any n-well, double metal, double polysilicon, CMOS process. The array has an average pixel voltage sensitivity of 2200 V/W at 100 Hz, an NEP of 2.4/spl times/10/sup -11/ W//spl radic/Hz at 100 Hz, and a specific detectivity of 4.4/spl times/10/sup 8/ cm /spl radic/Hz/W at 100 Hz. 相似文献

Dicopper(II) tetrakis(3,5-diisopropylsalicylate), [Cu(II)(2)(3,5-DIPS)(4)], is effective in increasing survival of lethally irradiated mice when it is administered after irradiation. The possibility that this radiorecovery activity might also facilitate recovery from radiation-induced impaired increase in body mass and locomotion was examined. Cu(II)(2)(3,5-DIPS)(4) was used to treat LD 50/30 gamma irradiated female C57BL/6 mice after irradiation. A dose of 0, 5, 10, or 20 mumol Cu(II)(2) (3, 5-DIPS)(4) /kilogram of body mass was administered subcutaneously 3 hrs after LD 50/30 irradiation and change in body mass and locomotor activity measured daily throughout the 30 day post-irradiation period. Treatment with 5, 10, or 20 mumol Cu(II)(2) (3,5-DIPS)(4) /kg of body mass increased survival, which was statistically significant for the 10 mumol /kg of body mass-treated group compared to the vehicle-treated group (P<0.05), significantly (P<0.05) increased recovery of locomotion from days 13 to 15 post-irradiation onward for all treated groups compared to vehicle-treated mice, and increased recovery of body mass gain from day 14 onward for the 20 mumol /kg of body mass-treated group (P<0.001) and day 21, although not statistically significant, for the 10 mumol /kg of body mass-treated group. There were no statistically significant differences between the increase in survival, recovered increase in body mass, and recovered increase in locomotion for mice treated with 10 mumol or 20 mumol Cu(II)(2)(3,5-DIPS)(4) /kg on day 30 post-irradiation. It is concluded that Cu(II)(2)(3,5-DIPS)(4) in addition to increasing survival of irradiated mice increases the rate of recovery of radiation-induced decrease in body mass and locomotion. 相似文献

Partitioning in situ total spectral absorption by use of moored spectral absorption-attenuation meters 总被引:1,自引:0,他引:1
High-temporal-resolution spectral absorption data were acquired by use of one bottom-mounted (~68-m) and three moored spectral absorption and attenuation meters (ac-9 meters at 14, 37, and 52 m) on the Middle Atlantic Bight continental shelf during the fall 1996 period of the Coastal Mixing and Optics experiment. We employed a previously published spectral absorption model with the data to partition total absorption into absorption by water, phytoplankton, detritus, and gelbstoff (dissolved matter). We validated the model by comparing its results against coincident in vivo absorption coefficients derived from discrete bottle samples. Correlations between modeled and in vivo spectra range from 0.873 to 0.998. We optimized these correlations to determine the model parameters. These parameters could not be determined solely from the moored ac-9 results. Therefore a separate set of absorption measurements (from discrete bottle samples) was necessary to permit values for the model parameters to be determined. Model results allow us to separate particulate and dissolved components of absorption and to examine the temporal variability and the vertical distributions and concentrations of each component, given the total absorption in the water column. 相似文献
