Melt electrospun fibers of poly(?-caprolactone) are accurately deposited using an automated stage as the collector. Matching the translation speed of the collector to the speed of the melt electrospinning jet establishes control over the location of fiber deposition. In this sense, melt electrospinning writing can be seen to bridge the gap between solution electrospinning and direct writing additive manufacturing processes.  相似文献

As a step toward prevention of bovine mastitis, a plasmid-mediated gene transfer technique was used to enable mammary cells to synthesize and secrete bovine lactoferricin and bovine tracheal antibacterial peptides. For this purpose, a series of mammary tissue-specific expression vectors, harboring the antibacterial peptide gene, the 5′-flanking regulation sequence of goat β-casein, and the bovine growth hormone polyadenylation signal sequence, were constructed using a eukaryotic expression vector pIRES1-neo. The mammary gland tissue-specific expression vector carrying the antimicrobial peptide genes dissolved in physiologic saline was injected directly into the lactating mammary glands of goats. The milk samples after injection were checked by Tricine-SDS-PAGE and bacterium inhibition zone assay. The results of these tests showed that the mammary gland tissue-specific expression vector driven by the goat β-casein gene promoter could efficiently direct the expression of antibacterial peptides in goat milk; the expression of antibacterial proteins lasted for 3 to 6 d. All of the milk samples collected from the mammary glands that had been injected with different vectors harboring the antibacterial peptide gene(s) exhibited bacteriostatic activity against different bacterial pathogens. These results demonstrated that the mammary gland tissue-specific expression vector could be used to introduce antibacterial peptide gene into the goat mammary gland, enabling secretion of a bioactive form of antibacterial peptide in the milk. This successful expression of antibacterial peptides in goat mammary glands provided a possible method to prevent mastitis in ruminants.  相似文献

The static fermentation of coconut water sucrose by Acetobacter xylinum was carried out at initial pH's of 3.0, 4.0, 5.0 or 6.0. Cellulose was produced at the surface, and its production was most favourable at pH's 4.0 and 5.0. These pH values also allowed for optimal bacterial growth. Oxygen concentration profiles were measured with microelectrodes at different cultivation stages, and steep profiles were obtained with penetration depths between 50 and 100 microm. A substrate penetration depth analysis confirmed the hypothesis that the first stage of the fermentation is entirely oxygen controlled. Diffusion calculations showed, however, that at a later stage sucrose becomes a limiting substrate also, which was confirmed by the decrease in cellulose production rate over time. The effective diffusion coefficient of oxygen in deactivated cellulose pellicles was measured with microelectrodes, and a value of 1.4 x 10(-9) m2/s was obtained under all investigated conditions. The oxygen flux was 5.9 x 10(-6) mol/m2.s, while a significantly higher value of 9.1 x 10(-6) mol/m2.s was obtained at pH 4.0.  相似文献

Addition of nanoparticulate zero-valent iron (nZVI) to oxygen-containing water results in oxidation of organic compounds. To assess the potential application of nZVI for oxidative transformation of organic contaminants, the conversion of benzoic acid (BA) to p-hydroxybenzoic acid (p-HBA) was used as a probe reaction. When nZVI was added to BA-containing water, an initial pulse of p-HBA was detected during the first 30 min, followed by the slow generation of additional p-HBA over periods of at least 24 h. The yield of p-HBA increased with increasing BA concentration, presumably due to the increasing 'ability of BA to compete with alternate oxidant sinks, such as ferrous iron. At pH 3, maximum yields of p-HBA during the initial phase of the reaction of up to 25% were observed. The initial rate of nZVI-mediated oxidation of BA exhibited a marked reduction at pH values above 3. Despite the decrease in oxidant production rate, p-HBA was observed during the initial reaction phase at pH values up to 8. Competition experiments with probe compounds expected to exhibit different affinities for the nZVI surface (phenol, aniline, o-hydroxybenzoic acid, and synthetic humic acids) indicated relative rates of reaction that were similar to those observed in competition experiments in which hydroxyl radicals were generated in solution. Examination of the oxidizing capacity of a range of Fe0 particles reveals a capacity in all cases to induce oxidative transformation of benzoic acid, but the high surface areas that can be achieved with nanosized particles renders such particles particularly effective oxidants.  相似文献

Slag is a ubiquitous byproduct of the iron-smelting industry and influences geochemistry and water quality in adjacent geologic units, ground and surface water. Despite extensive slag deposition along the Indiana shoreline of Lake Michigan, definitive evidence that slag has affected lakebed sediments has not been established. Concerns for the protection of water and ecosystem resources in the Great Lakes motivated this study to determine if strontium and boron isotopes could be used to identify and delineate slag-affected bed sediment in Lake Michigan. Sixty-five samples of bed sediment were acquired from the southern lobe of Lake Michigan and analyzed for 87Sr/86Sr and deltat11B. Samples immediately offshore from Indiana steel mills and slag-disposal sites contained higher median 87Sr/86Sr values (0.70881) than shoreline sediments collected elsewhere in the basin (0.70847) and uniquely decreased with increasing distance from the shoreline. The highest delta11B values occurred in sediments from the Indiana shoreline (+12.9 to 16.4/1000) but were also elevated in sediments collected offshore from three Lake Michigan cities (+11.7 to 12.7/1000). Contoured isotope data indicated that 82-154 km2 of bed sediment along the Indiana shoreline had elevated 87Sr/86Sr and delta11B values relative to shoreline sediments elsewhere in southern Lake Michigan.  相似文献

主要介绍了如何对省、市气象计量检定部门原来配置的湿度检定系统进行升级改造,使之成为数字化自动化检定系统。总体方案和设计思想是以计算机为核心,开发研制高精度数据采集器和控制电路,将检定过程中的物理量全部数字化,计算机从采集器的串行异步口读人数据,实现自动气象站湿度传感器室内检定过程全自动化,同时保留原系统的检定能力。检定数据传人局域网服务器,实现数据远程处理、打印和共享。软件基于WINDOWS平台,采用VB6.0编写。本系统已成功投入使用,提高了检定的准确性和检定的工作效率,减轻检定人员的劳动强度。检定实践证明,该系统运行稳定可靠,使用方便,操作简单。  相似文献
