3D printing offers great potential for developing complex flexure mechanisms. Recently, thickness-correction factors (TCFs) were introduced to correct the thickness and stiffness deviations of powder-based metal 3D printed flexure hinges during design and analysis. However, the reasons for the different TCFs obtained in each study are not clear, resulting in a limited value of these TCFs for future design and fabrication. Herein, the influence of the porous layer of 3D printed flexure hinges on the hinge thickness is investigated. Samples of parallelogram flexure mechanisms (PFMs) were 3D printed using selective laser melting (SLM) and 316L stainless steel powder. A 3D manufacturing error analysis was completed for each PFM sample via 3D scanning, surface roughness measurement and morphological observation. The thickness of the porous layer of the flexure hinge was independent of the designed hinge thickness and remained close to the average powder particle diameter. The effective hinge thickness could be estimated by subtracting twice the value of the porous layer thickness from the designed value. Guidelines based on finite element analysis and stiffness experiments are proposed. The limitations of the presented method for evaluating the effective hinge thickness of flexure hinges 3D printed via SLM are also discussed.
Feature engineering is one of the most complex aspects of system design in machine learning. Fortunately, kernel methods provide the designer with formidable tools to tackle such complexity. Among others, tree kernels (TKs) have been successfully applied for representing structured data in diverse domains, ranging from bioinformatics and data mining to natural language processing. One drawback of such methods is that learning with them typically requires a large number of kernel computations (quadratic in the number of training examples) between training examples. However, in practice substructures often repeat in the data which makes it possible to avoid a large number of redundant kernel evaluations. In this paper, we propose the use of Directed Acyclic Graphs (DAGs) to compactly represent trees in the training algorithm of Support Vector Machines. In particular, we use DAGs for each iteration of the cutting plane algorithm (CPA) to encode the model composed by a set of trees. This enables DAG kernels to efficiently evaluate TKs between the current model and a given training tree. Consequently, the amount of total computation is reduced by avoiding redundant evaluations over shared substructures. We provide theory and algorithms to formally characterize the above idea, which we tested on several datasets. The empirical results confirm the benefits of the approach in terms of significant speedups over previous state-of-the-art methods. In addition, we propose an alternative sampling strategy within the CPA to address the class-imbalance problem, which coupled with fast learning methods provides a viable TK learning framework for a large class of real-world applications. 相似文献
Summary The effects of cooking and sterilization at several temperatures on the free amino acids (FAA) content in albacore (Thunnus alalunga) muscle were studied during the processing of canned tuna. FAAs were derivatized witho-phtalaldehyde, separated on a C18 column by HPLC and detected by both fluorescence and ultra-violet detectors. After cooking the loss of FAAs was not significant. However, in the final product sterilized at 115 °C and 110 °C (throughout the whole process) there were significant losses with regard to the start material, but not at 118 °C (all temperatures leading to the same lethal F-value). The influence of the thermal process time at 115 °C was evaluated for 60 and 100 min. Significant losses were found between both canned products (25%) and between the raw fish and the final product (12% and 34%, process time 60 and 100 min, respectively). The determination of the content of FAA present in canned albacore may be a useful indication of the severity of the thermal processing.

Zusammenfassung Die Wirkung des Kochens und Sterilisierens bei verschiedenen Temperaturen auf den Gehalt an freien Aminosäuren (FAA) im Thunfischmuskel (Thunnus alalunga) wurde während der Herstellung von Thunfischkonserven untersucht. Die freien Aminosäuren wurden mito-Phtalaldehyd derivatisiert, auf einer C-18-Kolonne mit HPLC abgetrennt und durch Fluorescenz und UV-Detektoren nachgewiesen. Nach dem Kochen war der Verlust an FAA nicht signifikant, jedoch in dem bei 110° und 115 °C sterilisierten Endprodukt ergaben sich signifikante Verluste bezogen auf das Ausgangsmaterial, aber nicht auf das bei 118 °C erhitzte; alle Erhitzungstemperaturen führen zu demselben letalen F-Wert. Der Zeiteinfluß der Erhitzung bei 115 °C wurde bei 60 und 100 min bewertet. Signifikante Verluste sind bei beiden Dosenkonserven aufgetreten (25%) und zwischen diesen und dem Rohfisch ( 12% und 34%) bei einer Erhitzungszeit von 60 und 100 min). Die Bestimmung des FAA-Gehalts in Thunfischkonserven kann für den Nachweis der Einwandfreiheit des thermischen Prozesses sehr nützlich sein.
Methamphetamine is, worldwide, one of the most consumed drugs of abuse. One important side effect is neurodegeneration leading to a decrease in life expectancy. The aim of this paper was to check whether the drug affects one of the receptors involved in neurodegeneration/neuroprotection events, namely the adenosine A2A receptor (A2AR). First, we noticed that methamphetamine does not affect A2A functionality if the receptor is expressed in a heterologous system. However, A2AR becomes sensitive to the drug upon complexes formation with the cannabinoid CB1 receptor (CB1R) and the sigma 1 receptor (σ1R). Signaling via both adenosine A2AR and cannabinoid CB1R was affected by methamphetamine in cells co-expressing the two receptors. In striatal primary cultures, the A2AR–CB1R heteromer complex was detected and methamphetamine not only altered its expression but completely blocked the A2AR- and the CB1R-mediated activation of the mitogen activated protein kinase (MAPK) pathway. In conclusion, methamphetamine, with the participation of σ1R, alters the expression and function of two interacting receptors, A2AR, which is a therapeutic target for neuroprotection, and CB1R, which is the most abundant G protein-coupled receptor (GPCR) in the brain. 相似文献

The control of the fungal contamination on crops is considered a priority by the sanitary authorities of an increasing number of countries, and this is also due to the fact that the geographic areas interested in mycotoxin outbreaks are widening. Among the different pre- and post-harvest strategies that may be applied to prevent fungal and/or aflatoxin contamination, fungicides still play a prominent role; however, despite of countless efforts, to date the problem of food and feed contamination remains unsolved, since the essential factors that affect aflatoxins production are various and hardly to handle as a whole. In this scenario, the exploitation of bioactive natural sources to obtain new agents presenting novel mechanisms of action may represent a successful strategy to minimize, at the same time, aflatoxin contamination and the use of toxic pesticides. The Aflatox® Project was aimed at the development of new-generation inhibitors of aflatoxigenic Aspergillus spp. proliferation and toxin production, through the modification of naturally occurring molecules: a panel of 177 compounds, belonging to the thiosemicarbazones class, have been synthesized and screened for their antifungal and anti-aflatoxigenic potential. The most effective compounds, selected as the best candidates as aflatoxin containment agents, were also evaluated in terms of cytotoxicity, genotoxicity and epi-genotoxicity to exclude potential harmful effect on the human health, the plants on which fungi grow and the whole ecosystem. 相似文献
