The evolution of free amino acids and amino acid-derived volatile compounds during the ripening of Iberian ham from pigs reared in a Montanera system (outdoor-based, with acorn and pasture available) and a Pienso system (indoor-based, with a high oleic acid concentrate) was studied. Ripening time influenced significantly all the free amino acids detected (p < 0.05) except for threonine and tyrosine. The total free amino acid content increased significantly from day 120 to day 230 (drying stage) and then the concentration remained almost steady. This marked increase in the free amino acid content matched an increase in the amino acid-derived volatile compounds. The volatile compounds also increased after day 230. Conversely, rearing system had a weak effect on the free amino acid content and on the amino acid-derived volatile compounds. Only glutamic acid was significantly influenced (p = 0.027), and a slight effect on proline and aspartic acid was found (p = 0.051 and p = 0.084, respectively), concentrations being larger in Montanera hams than in Pienso ones. With regard to the amino acid derived volatile compounds, only a significant influence of rearing system on acetaldehyde and on the coelution of 2,6-dimethylpyrazine+dihidro-2(3H)furanone was found. The small differences caused by rearing system confirm the great importance of concentrate formulation.  相似文献

The susceptibility of 440 Listeria monocytogenes strains isolated from food (n=401) and clinical cases (n=39) between 1995 and 2005 was determined by standard agar dilution and E-test methods. Antimicrobial drugs currently used in veterinary and human therapy were tested, and they included penicillin G, ampicillin, cephalothin, gentamicin, chloramphenicol, tetracycline, doxycycline, trimethoprim, erythromycin, and clindamycin. The sensitivity of strains was established using Clinical and Laboratory Standards Institute (formerly NCCLS) breakpoints and MIC50 (the MIC for 50% of the strains) to MIC90 values. In general, isolates were susceptible to the majority of the antimicrobials tested, including beta-lactamics and aminoglycosides, which are normally used in the treatment of listeriosis. Resistance to tetracycline and doxycycline was found in five strains isolated from fresh trout belonging to the same fish farm. Molecular analysis by restriction endonuclease analysis showed a similar profile, suggesting the persistence of a strain well adapted to the presence of tetracycline in the environment of a fish farm, which is frequently used in aquaculture in order to prevent infections of fish.  相似文献

NADH oxidase from Lactobacillus brevis was kinetically characterized in two different buffers: Tris-HCl and glycine-sodium pyrophosphate (pH 9.0). Reaction kinetics was described using the Michaelis-Menten model with product (NAD(+)) inhibition. It was found that this type of inhibition is uncompetitive. Experiments in the continuously operated enzyme membrane reactor revealed a strong enzyme deactivation at two different residence times: 12 and 60 min. A stronger deactivation was observed at the lower residence time in the glycine-sodium pyrophosphate buffer. Enzyme deactivation was assumed to be of the first order. The developed mathematical model for the continuously operated enzyme membrane reactor described these experiments very well. The mathematical model simulations revealed that a high enzyme concentration (up to 30 g cm(-3)) is necessary to obtain and maintain the stationary NADH conversion near 100% for a longer period of time.  相似文献

The aim of this study was to find out whether very low doses of nephrotoxic and hepatotoxic mycotoxins ochratoxin A (OTA) and fumonisin B1 (FB1) induce oxidative stress in rat kidney and liver and whether their effect is synergistic. Rats were treated orally with OTA (5 ng/kg b.w. and 50 microg/kg b.w.) and FB1 (200 ng/kg b.w. and 50 microg/kg b.w.), or their combinations. Malondialdehyde (MDA) and protein carbonyls (PCs) concentration in kidney was affected with lower dose of OTA than in liver (p<0.05). FB1 did not affect MDA and PCs concentrations in the liver, while in the kidney both FB1 doses increased MDA concentration (p<0.05). The combination of the lower doses of OTA+FB1 increased the MDA and PCs concentration both in the liver and the kidney, compared to controls and animals treated with respective doses of mycotoxins (p<0.05). The combinations of mycotoxins reduced the catalase activity only in the kidney when compared to controls (p<0.05). In contrast to the increased kidney concentrations of MDA and PCs even with very low doses of OTA and FB1, the activity of catalase and SOD does not change. Combinations of OTA+FB1 affected almost all parameters, which indicates their potential to produce oxidative damage.  相似文献

The objective of this study was to determine whether a short training program, using real foods, would decreased their portion-size estimation errors after training. 90 student volunteers (20.18+/-0.44 y old) of the University of the Basque Country (Spain) were trained in observational techniques and tested in food-weight estimation during and after a 3-hour training period. The program included 57 commonly consumed foods that represent a variety of forms (125 different shapes). Estimates of food weight were compared with actual weights. Effectiveness of training was determined by examining change in the absolute percentage error for all observers and over all foods over time. Data were analyzed using SPSS vs. 13.0. The portion-size errors decreased after training for most of the foods. Additionally, the accuracy of their estimates clearly varies by food group and forms. Amorphous was the food type estimated least accurately both before and after training. Our findings suggest that future dietitians can be trained to estimate quantities by direct observation across a wide range of foods. However this training may have been too brief for participants to fully assimilate the application.  相似文献

We examined the in vivo contribution of insulin, T090137 (T09), agonist of liver X receptor (LXR), fenofibrate, agonist of peroxisome proliferator activated receptor (PPAR-α) and sterol regulatory element binding protein-1c (SREBP-1c) on the unsaturated fatty acid synthesis controlled by Δ6 and Δ5 desaturases, compared with the effects on stearoylcoenzyme A desaturase-1. When possible they were checked at three levels: messenger RNA (mRNA), desaturase protein and enzymatic activity. In control rats, only fenofibrate increased the insulinemia that was maintained by the simultaneous administration of T09, but this increase has no specific effect on desaturase activity. T09 enhanced SREBP-1 in control animals and the mRNAs and activity of the three desaturases in control and type-1 diabetic rats, demonstrating a LXR/SREBP-1-mediated activation independent of insulin. However, simultaneous administration of insulin and T09 to diabetic rats led to a several-fold increase of the mRNAs of the desaturases, suggesting a strong synergic effect between insulin and LXR/retinoic X receptor (RXR). Moreover, this demonstrates the existence of an interaction between unsaturated fatty acids and cholesterol metabolism performed by the insulin/SREBP-1c system and LXR/RXR. PPAR-α also increased the expression and activity of the three desaturases independently of the insulinemia since it was equivalently evoked in streptozotocin diabetic rats. Besides, PPAR-α increased the palmitoylcoenzyme A elongase, evidencing a dual regulation in the fatty acid biosynthesis at the level of desaturases and elongases. The simultaneous administration of fenofibrate and T09 did not show additive effects on the mRNA expression and activity of the desaturases. Therefore, the results indicate a necessary sophisticated interaction of all these factors to produce the physiological effects.  相似文献

Although a safe procedure, hemodialysis (HD) can cause numerous complications. The objective of this study was to evaluate the incidence of complications during dialysis, interdialytic weight gain, and the predialysis and postdialysis blood pressure in HD patients with and without variable sodium. Patients were observed during 12 HD sessions and those presenting with recurrent hypotension were selected for a step-wise model of variable sodium profiling. A total of 53 patients were evaluated; the mean-SD age was 53.7+/-16.3 years and 22 (41.5%) were male. Of these, 18 (34.0%) were selected to receive variable sodium profiling: the mean (SD) age was 59.9+/-12.6 years, and 10 (55.6%) were female. A significant decline in the occurrence of cramps (p<0.027), in the mean interdialytic weight gain (p<0.009), and a tendency to reduce the number of hypotensive episodes were detected in patients using variable sodium profiling. On the other hand, predialysis systolic blood pressure presented a significant increase (p<0.048). Using variable sodium, there was a statistically significant reduction in cramps and in the mean interdialytic weight gain. There was a significant increase in predialysis systolic pressure. Regarding hypotension episodes, only a tendency toward a reduction in the frequency of hypotension episodes could be detected.  相似文献

An expression for the friction factor of rough turbulent meandering flows is suggested: flow cross section is rectangular, the plan shape of the meandering channel is sine-generated. This expression gives the value of the friction factor as a function of position (which is determined by, among other factors, the local channel curvature) and channel sinuosity. The validity of the present expression is tested against results of laboratory measurements. The measurements were carried out in two meandering channels, one typifying “small” sinuosity, the other “large” sinuosity. It is found that the vertically averaged flows in these two channels exhibit two distinctly different (“in-” and “out-going”) flow patterns, depending on whether the channel sinuosity is “large” or “small.” These two radically different flow pictures cannot be supplied by the vertically averaged equations of motion if they are solved for a constant friction factor. The present consideration of the friction factor as a function of position and channel sinuosity is found to yield the computed vertically averaged flows that are in agreement with the flow pictures measured for both large and small values of sinuosity.  相似文献

 A penetrometry study was made of the effect of the addition of gelatine on the texture characteristics of yoghurts and acid-heat-induced milk gels. Detailed analysis of the various penetration profiles provided valuable information about the nature of the samples studied. Gelatine was compatible with the milk systems over a wide range of concentrations and it prevented syneresis. In yoghurts prepared with and without the addition of skim-milk powder, the addition of increasing concentrations of gelatine up to 1.5% gradually modified the mechanical characteristics of the final gels. In acid-heat-induced milk gels, whose mechanical characteristics depend on the final pH value, the addition of increasing quantities of gelatine up to 6.67% practically eliminated this dependence, and the effect of pH was only significant in the gels with 1.5% gelatine, which were structurally weaker.
