Synthetic diacylglycerol‐lactones (DAG‐lactones) are effective modulators of critical cellular signaling pathways, downstream of the lipophilic second messenger diacylglycerol, that activate a host of protein kinase C (PKC) isozymes and other nonkinase proteins that share similar C1 membrane‐targeting domains with PKC. A fundamental determinant of the biological activity of these amphiphilic molecules is the nature of their interactions with cellular membranes. This study examines the biological properties of charged DAG‐lactones exhibiting different alkyl groups attached to the heterocyclic nitrogen of an α‐pyridylalkylidene chain, and particularly the relationship between membrane interactions of the substituted DAG‐lactones and their respective biological activities. Our results suggest that bilayer interface localization of the N‐alkyl chain in the R2 position of the DAG‐lactones inhibits translocation of PKC isoenzymes onto the cellular membrane. However, the orientation of a branched alkyl chain at the bilayer surface facilitates PKC binding and translocation. This investigation emphasizes that bilayer localization of the aromatic side residues of positively charged DAG‐lactone derivatives play a central role in determining biological activity, and that this factor contributes to the diversity of biological actions of these synthetic biomimetic ligands.  相似文献

Amyloid‐β peptides (Aβ) and the protein human serum albumin (HSA) interact in vivo. They are both localised in the blood plasma and in the cerebrospinal fluid. Among other functions, HSA is involved in the transport of the essential metal copper. Complexes between Aβ and copper ions have been proposed to be an aberrant interaction implicated in the development of Alzheimer's disease, where Cu is involved in Aβ aggregation and production of reactive oxygen species (ROS). In the present work, we studied copper‐exchange reaction between Aβ and HSA or the tetrapeptide DAHK (N‐terminal Cu‐binding domain of HSA) and the consequence of this exchange on Aβ‐induced ROS production and cell toxicity. The following results were obtained: 1) HSA and DAHK removed CuII from Aβ rapidly and stoichiometrically, 2) HSA and DAHK were able to decrease Cu‐induced aggregation of Aβ, 3) HSA and DAHK suppressed the catalytic HO. production in vitro and ROS production in neuroblastoma cells generated by Cu–Aβ and ascorbate, 4) HSA and DAHK were able to rescue these cells from the toxicity of Cu–Aβ with ascorbate, 5) DAHK was more potent in ROS suppression and restoration of neuroblastoma cell viability than HSA, in correlation with an easier reduction of CuII–HSA than Cu–DAHK by ascorbate, in vitro. Our data suggest that HSA is able to decrease aberrant CuII–Aβ interaction. The repercussion of the competition between HSA and Aβ to bind Cu in the blood and brain and its relation to Alzheimer's disease are discussed.  相似文献

BACKGROUND: This research provides new insights into the biosorption of zinc on a waste product from the orange juice industry. Optimal operating conditions maximizing percentage zinc removal were determined in batch and fixed‐bed systems. Biomass was characterized by FTIR spectroscopy and by major cation content in order to better understand the biosorpion mechanism. Zn‐loaded orange waste was proposed to be used as an alternative fuel in cement kilns. RESULTS: Sorption capacity was strongly affected by biosorbent dose and solution pH, and was not strongly sensitive to particle size under the experimental conditions studied. Equilibrium data were successfully described by a Langmuir model and sorption kinetic data were adequately modelled with the pseudo‐second‐order and Elovich rate equation. The biomass was found to possess high sorption capacity (qmax = 0.664 mmol g?1) and biosorption equilibrium was established in less than 3 h. Experimental breakthrough curves were adequately fitted to the Thomas model and the dose–response model, obtaining sorption capacities in continuous assays higher than those found in batch mode. Characterization of the biomass suggested the possible contribution of carboxyl and hydroxyl groups of biomass in Zn2+ biosorption and it also highlighted the important role of light metal ions in a possible ion‐exchange mechanism. The study of partial oxidation of methane (POM) over bimetallic nickel–lanthanide oxides was undertaken. Binary intermetallic compounds of the type LnNi (Ln = Pr, Gd, Lu) were used as bimetallic nickel–lanthanide oxides precursors and the products (NiO·Pr2NiO4, 2NiO·Gd2O3 and 2NiO·Lu2O3) were characterized by means of X-ray diffraction, X-ray photoelectron spectroscopy, Raman and temperature programmed reduction. The catalytic activity increases when gadolinium or lutetium replaces praseodymium and the selectivity of the bimetallic nickel–lanthanide oxides is clearly different from that of single metal oxides and/or mechanical mixtures.The existence of an unusual synergism effect between the two metal oxide phases (NiO and Ln2O3) that lead to higher conversions of methane and higher selectivities to hydrogen and carbon monoxide correlate also the catalysts stability to deactivation. The activity and selectivity of the gadolinium and lutetium compounds is, under the same conditions, equivalent to that of a platinum commercial catalyst, 5 wt% Pt/Al2O3, which stresses the good catalytic performance of this new type of compounds for the production of H2 and CO (H2/CO = 2).  相似文献

The objective of this article is to analyse how green supply chain management (GSCM) practices are being adopted by some high-tech companies located in Brazil. The research was conducted using the case study approach, focusing on eight companies that are representative of this sector. The main results are: (a) the most adopted GSCM practices in the studied high-tech companies located in Brazil are internal environmental management, investment recovery and reverse logistics and (b) Brazilian environmental legislation and international policies are very important in driving the adoption of GSCM practices. The internationalisation of companies was also found to be a variable that interferes with the adoption of GSCM practices. This is one of the first studies that examine the relationship between GSCM and the internationalisation of companies located in Brazil.  相似文献

The yolk sac (YS) is the main source of embryonic nutrition during the period when the placenta has not yet formed. It is also responsible for hematopoiesis because the blood cells develop from it as part of the primitive embryonic circulation. The objective of this study was to characterize the transitional area between the YS and primitive gut using the techniques of light microscopy, transmission electron microscopy, and immunohistochemistry to detect populations of pluripotent cells by labeling with Oct4 antibody. In all investigated embryos, serial sections were made to permit the identification of this small, restricted area. We identified the YS connection with the primitive intestine and found that it is composed of many blood islands, which correspond to the vessels covered by vitelline and mesenchymal cells. We identified large numbers of hemangioblasts inside the vessels. The mesenchymal layer was thin and composed of elongated cells, and the vitelline endodermal membrane was composed of large, mono‐ or binucleated cells. The epithelium of the primitive intestine comprised stratified columnar cells and undifferentiated mesenchymal cells. The transitional area between the YS and the primitive intestine was very thin and composed of cells with irregular shapes, which formed a delicate lumen containing hemangioblasts. In the mesenchyme of the transitional area, there were a considerable number of small vessels containing hemangioblasts. Using Oct4 as a primary antibody, we identified positive cells in the metanephros, primordial gonad, and hepatic parenchyma as well as in YS cells, suggesting that these regions contain populations of pluripotent cells. The application of resin‐based materials is an alternative of treatment for eroded lesions. Nevertheless, there are no studies about the penetration of these materials into eroded lesion, which might affect its adhesion. Therefore, this study evaluated the penetration of four resin‐based materials, with and without enamel etching. By using an in vitro protocol, types of treatment were studied at five levels (AdheSE®, Tetric N‐Bond®, Single Bond 2®, Helioseal Clear®, Icon®) and types of enamel etching in two levels (with and without). Materials were stained with 0.02 mg/mL ethanolic solution of tetramethylrhodamine isothiocyanate. Bovine enamel samples (4 × 4 mm) were immersed in 0.01 M HCl, pH 2.3, for 30 seconds to produce initial eroded lesions. Afterward, the materials were applied on half of sample enamel surface following the manufacturer's instructions. On the other half of sample, the materials were applied without etching the enamel. Materials penetration into the enamel was assessed by Confocal Laser Scanning Microscopy on reflection and fluorescence modes. The penetration depth (PD) was measured using ImageJ software. Data were analyzed by two‐way ANOVA and Tukey test (P < 0.05). Regardless of the material, etched enamel resulted in higher PD than non‐etched (P < 0.05). Icon® showed the highest PD in enamel followed by Helioseal Clear® (P < 0.05), with significant difference between them (P < 0.05) and no difference was found among AdheSE®, Tetric N‐Bond®, and Single Bond 2® (P > 0.05). It can be concluded that prior enamel etching increased the materials penetration into eroded enamel and the Icon®—infiltrant presented highest penetration. Microsc. Res. Thermodynamic analysis of three binary Ti-based alloys: Ti–Al, Ti–V, and Al–V, as well as ternary alloy Ti–Al–V, is shown in this paper. Thermodynamic analysis involved thermodynamic determination of activities, coefficient of activities, partial and integral values for enthalpies and Gibbs energies of mixing and excess energies at four different temperatures: 2000, 2073, 2200 and 2273 K, as well as calculated phase diagrams for the investigated binary and ternary systems. The objective of this study was to determine whether a short training program, using real foods, would decreased their portion-size estimation errors after training. 90 student volunteers (20.18+/-0.44 y old) of the University of the Basque Country (Spain) were trained in observational techniques and tested in food-weight estimation during and after a 3-hour training period. The program included 57 commonly consumed foods that represent a variety of forms (125 different shapes). Estimates of food weight were compared with actual weights. Effectiveness of training was determined by examining change in the absolute percentage error for all observers and over all foods over time. Data were analyzed using SPSS vs. 13.0. The portion-size errors decreased after training for most of the foods. Additionally, the accuracy of their estimates clearly varies by food group and forms. Amorphous was the food type estimated least accurately both before and after training. Our findings suggest that future dietitians can be trained to estimate quantities by direct observation across a wide range of foods. However this training may have been too brief for participants to fully assimilate the application.  相似文献
