Acacia angustissima has potential in agroforestry systems for forage production in tropical environments. However, feeding trials indicate that the leaves are potentially toxic. Signs of toxicity in sheep are similar to those presented by sheep fed flatpea (Lathyrus sylvestris) hay which contains a high concentration of the non‐protein amino acid 2,4‐diaminobutanoic acid (DABA). Cation exchange chromatography, high‐performance liquid chromatography (HPLC) and NMR were used to isolate and identify the non‐protein amino acid in A angustissima leaves. A quantitative HPLC method was used to determine the content of the non‐protein amino acid in leaves from 20 accessions. The non‐protein amino acid was identified as 4‐N‐acetyl‐2,4‐diaminobutanoic acid (ADAB), previously reported to occur in seeds. The average and range in ADAB content were 16.8 and 10.6–25.4 mg g?1 respectively. This content of ADAB is higher than the levels of DABA in flatpea hay that are toxic to sheep. © 2001 Society of Chemical Industry 相似文献

The flocculation of the sodium, potassium and lithium forms of a kaolinite material have been investigated and discussed in relation to the thickness of the adsorbed layer of water molecules on the clay surfaces. Two methods of assessing flocculation in concentrated suspensions were used, namely the rates of fall of the flocculated suspensions and the rates of filtration through the settled flocs. The flocculating agent was neutral polyacrylamide. Nitrogen and water vapour adsorption isotherms are reported on the various samples, together with heats of immersion in water.The extent of flocculation decreased in the order of Li clay > K clay > Na clay; the amount of adsorbed water decreased in the order Na clay > K clay > Li clay; and the heat of wetting in the order Na clay > Li clay ? K clay. The anomalous behaviour of the Li clay is associated with the strong hydration of the Li ion on the clay surface. 相似文献

Giant unilamellar vesicles (GUVs) are considered to be the gold standard for assembling artificial cells from the bottom up. In this study, we investigated the behavior of such biomimetic vesicles as they were subjected to mechanical compression. A microfluidic device is presented that comprises a trap to capture GUVs and a microstamp that is deflected downwards to mechanically compress the trapped vesicle. After characterization of the device, we show that single-phase GUVs can be controllably compressed to a high degree of deformation (D=0.40) depending on the pressure applied to the microstamp. A permeation assay was implemented to show that vesicle bursting is prevented by water efflux. Next, we mechanically compressed GUVs with co-existing liquid-ordered and liquid-disordered membrane phases. Upon compression, we observed that the normally stable lipid domains reorganized themselves across the surface and fused into larger domains. This phenomenon, observed here in a model membrane system, not only gives us insights into how the multicomponent membranes of artificial cells behave, but might also have interesting consequences for the role of lipid rafts in biological cells that are subjected to compressive forces in a natural environment. 相似文献

Constrained binding peptides (peptide aptamers) may serve as tools to explore protein conformations and disrupt protein-protein interactions. The quality of the protein scaffold, by which the binding peptide is constrained and presented, is of crucial importance. SQT (Stefin A Quadruple Mutant-Tracy) is our most recent development in the Stefin A-derived scaffold series. Stefin A naturally uses three surfaces to interact with its targets. SQT tolerates peptide insertions at all three positions. Peptide aptamers in the SQT scaffold can be expressed in bacterial, yeast and human cells, and displayed as a fusion to truncated pIII on phage. Peptides that bind to CDK2 can show improved binding in protein microarrays when presented by the SQT scaffold. Yeast two-hybrid libraries have been screened for binders to the POZ domain of BCL-6 and to a peptide derived from PBP2', specific to methicillin-resistant Staphylococcus aureus. Presentation of the Noxa BH3 helix by SQT allows specific interaction with Mcl-1 in human cells. Together, our results show that Stefin A-derived scaffolds, including SQT, can be used for a variety of applications in cellular and molecular biology. Freeze‐dried cellulose nanocrystals (CNCs) were dispersed in the thermoplastic polyurethane [Pellethane 2363‐55D (P55D)] by a solvent casting method to fabricate CNC‐reinforced nanocomposites. This study demonstrated that the addition of small amounts (1–5 wt %) of CNCs to P55D increased the thermal degradation temperature while maintaining a similar stiffness, strength, and elongation of the neat P55D. CNC additions to P55D did not alter the glass‐transition temperature, but the onset decomposition temperature was shifted from 286 to 327°C when 1 wt % CNCs was dispersed in the matrix. The higher onset decomposition temperature was attributed to the formation of hydrogen bonds between the hydroxyl groups on the CNC surface and urethane groups in the hard block of P55D. The ultimate tensile strength and strain to failure (εf) of the nanocomposites were minimally affected by additions up to 5 wt % CNCs, whereas the elastic modulus was increased by about 70%. The observation that εf was unchanged with the addition of up to 5 wt % CNCs suggested that the flow/sliding of the hard blocks and chains were not hindered by the presence of the CNCs during plastic deformation. The ramifications of this study was that CNC additions resulted in wider processing temperatures of P55D for various biomedical devices while maintaining a similar stiffness, strength, and elongation. © 2015 Wiley Periodicals, Inc. There exist several estimators of the memory parameter in long- memory time series models with the spectrum specified only locally near zero frequency. In this paper we give an asymptotic lower bound for the minimax risk of any estimator of the memory parameter as a function of the degree of local smoothness of the spectral density at zero. The lower bound allows one to evaluate and compare different estimators by their asymptotic behaviour, and to claim the rate optimality for any estimator attaining the bound. A log-periodogram regression estimator, analysed by Robinson (Log-periodogram regression of time series with long range dependence. Ann. Fragile histidine triad protein (Fhit) is a diadenosine triphosphate
(ApppA) hydrolase encoded at the human chromosome 3 fragile site which is
frequently disrupted in tumors. Reintroduction of FHIT coding sequences to
cancer cell lines with FHIT deletions suppressed the ability of these cell
lines to form tumors in nude mice even when the reintroduced FHIT gene had
been mutated to allow ApppA binding but not hydrolysis. Because this
suggested that the tumor suppressor activity of Fhit protein depends on
substrate-dependent signaling rather than ApppA catabolism, we prepared two
crystalline forms of Fhit protein that are expected to model its
biologically active, substrate-bound state. Wild-type and the His96Asn
forms of Fhit were overexpressed in Escherichia coli, purified to
homogeneity and crystallized in the presence and absence of ApppA and an
ApppA analog. Single crystals obtained by vapor diffusion against ammonium
sulfate diffracted X-rays to beyond 2.75 A resolution. High quality native
synchrotron X-ray data were collected for an orthorhombic and a hexagonal
crystal form.

An equation relating the measured irradiance and the output power of a fluorescent lamp was derived by Keitz. The equation forms the basis for a new protocol that has been proposed for quantifying the total flux from an ultraviolet lamp. There has been confusion in the literature regarding the spatial distribution of flux from lamp emitters, which has led to emission models that are similar to the Keitz model but are incorrect.
