The microglial fatty-acid-binding protein 4-uncoupling protein 2 (FABP4-UCP2) axis is a key regulator of neuroinflammation in high-fat-diet (HFD)-fed animals, indicating a role for FABP4 in brain immune response. We hypothesized that the FABP4-UCP2 axis is involved in regulating diet-induced cognitive decline. We tested cognitive function in mice lacking microglial FABP4 (AKO mice). Fifteen-week-old male AKO and wild-type (WT) mice were maintained on 60% HFD or normal chow (NC) for 12 weeks. Body composition was measured using EchoMRI. Locomotor activity, working memory, and spatial memory were assessed using behavioral tests (open field, T-maze, and Barnes maze, respectively). Hippocampal microgliosis was assessed via immunohistochemical staining. An inflammatory cytokine panel was assayed using hippocampal tissue. Real-time RT-PCR was performed to measure microglial UCP2 mRNA expression. Our data support that loss of FABP4 prevents cognitive decline in vivo. HFD-fed WT mice exhibited impaired long- and short-term memory, in contrast with HFD-fed AKO mice. HFD-fed WT mice had an increase in hippocampal inflammatory cytokine expression (IFNγ, IL-1β, IL-5, IL-6, KC/GRO(CXCL1), IL-10, and TNFα) and microgliosis, and decreased microglial UCP2 expression. HFD-fed AKO mice had decreased hippocampal inflammatory cytokine expression and microgliosis and increased microglial UCP2 expression compared to HFD-fed WT mice. Collectively, our work supports the idea that the FABP4-UCP2 axis represents a potential therapeutic target in preventing diet-induced cognitive decline.  相似文献

Differential scanning calorimetry (DSC) and dynamic mechanical analysis (DMA) were used to identify the mechanisms that lead to differences in the mechanical behavior of formulations of polypropylene blended with maleated polypropylene (MAPP) copolymers. MAPP lowered the melting temperature of PP indicating that less stable crystals were formed possibly because of cocrystallization of PP and MAPP. Crystallization kinetics revealed that copolymers do not change the rate of crystal growth, but may retard nucleation leading to a more spherulitic morphology. The dynamic storage modulus slightly increased in the glassy region with the small addition amounts of MAPP, while mechanical dampening systematically decreased with MAPP addition. An analysis of the viscoelastic behavior did not reveal any real differences in molecular coupling around the β‐transition of PP with the addition of the MAPP copolymer. At low addition levels, MAPP does not appear to have a significant impact on the viscoelastic properties of the polymer blend. © 2008 Wiley Periodicals, Inc. J Appl Polym Sci, 2009  相似文献

A model is presented that suggests that the optimal strategy for an animal walking or crawling on a substrate while searching for the source of a chemical carried by a shifting wind or current may be to move upwind (or against the current). The requirements are that (1) the current direction fluctuates rapidly within a range that exceeds 30 ° on both sides of the mean direction, and (2) the searching animal can move accurately up- or downwind and, once within the influence of the chemical plume, can move efficiently to the source. Under these conditions, an upwind search is shorter (on the average) by 0.9–0.3 of the range of influence of the chemical stimulus. Thus, this strategy is relatively more important when the total search path is short, i.e., when sources are typically close by. The mean length of the downwind search path is given by L d =d +r[(1/E) + cos ]/2 and the upwind search path by L u =d +r[(2/E) – (/sin ) – cos ]/2 whered is the starting distance up- or downwind of the source,r is the range of stimulus influence,E is the efficiency with which the searcher moves to the source within the influence of the plume,a is the angle of the maximum extent of wind direction from the mean, and the average, , is taken over all starting positions across the wind direction that lead to search paths that intercept the area swept by the plume.  相似文献

The subcritical growth of fatigue cracks under (tension-tension) cyclic loading is demonstrated for ceramic materials, based on experiments using compact C(T) specimens of a MgO-partially-stabilized zirconia (PSZ), heat-treated to vary the fracture toughness K c from ∼3 to 16 MPa·m1/2 and tested in inert and moist environments. Analogous to behavior in metals, cyclic fatigue-crack rates (over the range 10−11 to 10−5 m/cycle) are found to be a function of the stress-intensity range, environment, fracture toughness, and load ratio, and to show evidence of fatigue crack closure. Unlike toughness behavior, growth rates are not dependent on through0-thickness constraint. Under variable-amplitude cyclic loading, crack-growth rates show transient accelerations following low-high block overloads and transient retardations following high-low block overloads or single tensile overloads, again analogous to behavior commonly observed in ductile metals. Cyclic crack-growth rates are observed at stress intensities as low as 50% of K c , and are typically some 7 orders of magnitude faster than corresponding stress-corrosion crack-growth rates under sustained-loading conditions. Possible mechanisms for cyclic crack advance in ceramic materials are examined, and the practical implications of such "ceramic fatigue" are briefly discussed.  相似文献

Binder evolution information generated using thermal analysis techniques is used along with microstructural information to define a thermal cycle for debinderizing injection-molded articles. In addition, the roles of binder chemistry, powder morphology, binder loading, article size, heating rate, and environmental conditions in determining a satisfactory thermal cycle are investigated. Major binder evolution events and types of defects generated are identified. An improved binder removal cycle is developed from this evaluation for organics elimination of a honeycomb structure.  相似文献

We report a very simple synthesis strategy for the formation of moderately porous ionosilica materials containing ammonium groups. We synthesized an ionosilica xerogel and an ionosilica aerogel, either via conventional or supercritical work‐up. The synthesis scheme of both precursor and ionosilica materials could easily be up‐scaled to a 100 g scale. All syntheses and work‐up procedures are sustainable as no additional agent and very low quantities of solvents were used, and no additional purifications steps were necessary. Although both materials show excellent anion exchange properties for chromate adsorption, noticeable differences were found regarding the thermodynamics of the exchange process. We attribute these differences to different surface chemistries of the materials, induced by the different work‐up procedures. Due to the easy availability of high quantities of material, ionosilicas can be implemented as functional ion exchange materials in larger scale processes, thus opening new fields of applications.  相似文献

Chick endogenous phospholipids were doubly labeled by an intravenous injection of [32P] phosphate and [1-14C] oleic acid, and the free fatty acid and phospholipid fraction of gall bladder bile and in contents of upper small intestine were analyzed 4 days later. There was evidence of hydrolysis of biliary phosphatidylcholine to lysophosphatidylcholine in the duodenum and jejunum, but this did not account for the pronounced increase in the14C radioactivity of the free fatty acids relative to the32P phospholipid radioactivity between bile and upper intestinal segments. It is suggested that phosphatidylcholine is largely absorbed in the duodenum of the chick while the remainder is progressively hydrolyzed and absorbed.  相似文献

The mechanical degradation of pulp cellulose fiber was studied at ambient temperature and at 77°K. ESR findings reveal that mechanical degradation occurs via free-radical routes. Three types of mechanoradicals contributing singlet, doublet, and triplet ESR signals are identified. The singlet signals are derived from the alkoxy radicals at C4 positions as a consequence of the cleavage of glucosidic bonds, the radical pairs generated at C1 positions contributing the doublet signals. Triplet signals are derived from the C2 and C3 positions due to the cleavage of C2 and C3 bonds. Of these radicals, alkoxy radicals are the most stable at ambient temperature. Carbon radicals are capable of interacting rapidly with oxygen molecules to produce peroxy radical intermediates, where alkoxy radicals are inert toward oxygen molecules. ESR study also reveals that cellulose mechanoradicals are capable of initiating vinyl polymerization. MMA propagating radicals are identified when the monomers are in contact with cellulose mechanoradicals. The ability of mechanoradicals to initiate graft copolymerization from cellulose fiber is discussed.  相似文献
