Light composition modulates plant growth and defenses, thus influencing plant–pathogen interactions. We investigated the effects of different light-emitting diode (LED) red (R) (665 nm) and blue (B) (470 nm) light combinations on Actinidia chinensis performance by evaluating biometric parameters, chlorophyll a fluorescence, gas exchange and photosynthesis-related gene expression. Moreover, the influence of light on the infection by Pseudomonas syringae pv. actinidiae (Psa), the etiological agent of bacterial canker of kiwifruit, was investigated. Our study shows that 50%R–50%B (50R) and 25%R–75%B (25R) lead to the highest PSII efficiency and photosynthetic rate, but are the least effective in controlling the endophytic colonization of the host by Psa. Monochromatic red light severely reduced ΦPSII, ETR, Pn, TSS and photosynthesis-related genes expression, and both monochromatic lights lead to a reduction of DW and pigments content. Monochromatic blue light was the only treatment significantly reducing disease symptoms but did not reduce bacterial endophytic population. Our results suggest that monochromatic blue light reduces infection primarily by modulating Psa virulence more than host plant defenses.  相似文献

ABCB11 is responsible for biliary bile acid secretion at the canalicular membrane of hepatocytes. Variations in the ABCB11 gene cause a spectrum of rare liver diseases. The most severe form is progressive familial intrahepatic cholestasis type 2 (PFIC2). Current medical treatments have limited efficacy. Here, we report the in vitro study of Abcb11 missense variants identified in PFIC2 patients and their functional rescue using cystic fibrosis transmembrane conductance regulator potentiators. Three ABCB11 disease-causing variations identified in PFIC2 patients (i.e., A257V, T463I and G562D) were reproduced in a plasmid encoding an Abcb11-green fluorescent protein. After transfection, the expression and localization of the variants were studied in HepG2 cells. Taurocholate transport activity and the effect of potentiators were studied in Madin–Darby canine kidney (MDCK) clones coexpressing Abcb11 and the sodium taurocholate cotransporting polypeptide (Ntcp/Slc10A1). As predicted using three-dimensional structure analysis, the three variants were expressed at the canalicular membrane but showed a defective function. Ivacaftor, GLP1837, SBC040 and SBC219 potentiators increased the bile acid transport of A257V and T463I and to a lesser extent, of G562D Abcb11 missense variants. In addition, a synergic effect was observed when ivacaftor was combined with SBC040 or SBC219. Such potentiators could represent new pharmacological approaches for improving the condition of patients with ABCB11 deficiency due to missense variations affecting the function of the transporter.  相似文献

IspH, also called LytB, a protein involved in the biosynthesis of isoprenoids through the methylerythritol phosphate pathway, is an attractive target for the development of new antimicrobial drugs. Here, we report crystal structures of Escherichia coli IspH in complex with the two most potent inhibitors: (E)-4-mercapto-3-methylbut-2-en-1-yl diphosphate (TMBPP) and (E)-4-amino-3-methylbut-2-en-1-yl diphosphate (AMBPP) at 1.95 and 1.7 Å resolution, respectively. The structure of the E. coli IspH:TMBPP complex exhibited two conformers of the inhibitor. This unexpected feature was exploited to design and evolve new antimicrobial candidates in silico.  相似文献

Histone chaperones regulate the flow and dynamics of histone variants and ensure their assembly into nucleosomal structures, thereby contributing to the repertoire of histone variants in specialized cells or tissues. To date, not much is known on the distribution of histone variants and their modifications in the dry seed embryo. Here, we bring evidence that genes encoding the replacement histone variant H3.3 are expressed in Arabidopsis dry seeds and that embryo chromatin is characterized by a low H3.1/H3.3 ratio. Loss of HISTONE REGULATOR A (HIRA), a histone chaperone responsible for H3.3 deposition, reduces cellular H3 levels and increases chromatin accessibility in dry seeds. These molecular differences are accompanied by increased seed dormancy in hira-1 mutant seeds. The loss of HIRA negatively affects seed germination even in the absence of HISTONE MONOUBIQUITINATION 1 or TRANSCRIPTION ELONGATION FACTOR II S, known to be required for seed dormancy. Finally, hira-1 mutant seeds show lower germination efficiency when aged under controlled deterioration conditions or when facing unfavorable environmental conditions such as high salinity. Altogether, our results reveal a dependency of dry seed chromatin organization on the replication-independent histone deposition pathway and show that HIRA contributes to modulating seed dormancy and vigor.  相似文献

Sheet molding compound (SMC) parts are fiber‐reinforced unsaturated polyester (UP) composites molded at 140–170 °C under a pressure of 60–100 bar. For economic and ecological reasons, the aim of this research project is to develop new SMC formulations to modify the molding conditions. For this, SMC formulations were modified and optimized to decrease the molding temperature to 100 °C. The strategy was to change the catalytic system (peroxides) in order to obtain highly reactive formulations at 100 °C. First, the temperatures of initiation of the reaction were determined by rheological and DSC measurements for each peroxide. Second, the UP resin crosslinking kinetics were measured for the various peroxides during an isothermal curing at 100 °C. The results obtained with the three experimental methods are compared and discussed. Finally, the laboratory analyses were validated by SMC molding trials. © 2019 Society of Chemical Industry  相似文献

This article presents two high‐temperature thermoplastic powders which were sintered by spark plasma sintering in order to get homogeneous mechanical properties. Dense polyimide (PI) and polyetheretherketone (PEEK) specimens were obtained at temperatures as low as 320°C for PI and 200°C for PEEK, respectively. Relative densities higher than 99% were reached for both materials. In order to characterize their properties, in situ measurements with compression and hardness tests were carried out on sintered samples. A relatively simple method is described for routine determination of hydroxyl values for a wide range of hydroxyl containing compounds including soybean oil polyols, polyols derived from soy meal, polyether polyols and ethylene glycol as well as their blends. This method is based on reacting the hydroxyl compound with hexamethyldisilazane (HMDS) and determining the FTIR peak area of the silylated product at 1,251 cm?1. The method is simple, accurate and reproducible. It is not limited to a specific family of polyol compounds. It does not require any special equipment, hazardous chemicals and can be carried out by non‐technical staff as a rapid and convenient method for quantitative determination of hydroxyl values. An excellent linear correlation was obtained between this spectroscopic method and conventional titration methods for different polyols over a wide range of hydroxyl values. Furthermore, unlike the titration methods the current method is not affected by the presence of acid, base or small amounts of water in the test sample.  相似文献
