Breast cancer was one of the first malignancies to benefit from targeted therapy, i.e., treatments directed against specific markers. Inhibitors against HER2 are a significant example and they improved the life expectancy of a large cohort of patients. Research on new biomarkers, therefore, is always current and important. AXL, a member of the TYRO-3, AXL and MER (TAM) subfamily, is, today, considered a predictive and prognostic biomarker in many tumor contexts, primarily breast cancer. Its oncogenic implications make it an ideal target for the development of new pharmacological agents; moreover, its recent role as immune-modulator makes AXL particularly attractive to researchers involved in the study of interactions between cancer and the tumor microenvironment (TME). All these peculiarities characterize AXL as compared to other members of the TAM family. In this review, we will illustrate the biological role played by AXL in breast tumor cells, highlighting its molecular and biological features, its involvement in tumor progression and its implication as a target in ongoing clinical trials.  相似文献

Bone is an inorganic template containing organic material inside which can be converted into hydroxyapatite‐rich material by pyrolysis. Nowadays, there is a growing research interest in the use of hydroxyapatite, the chemical formula of which is Ca10(PO4)6(OH)2. In the present work, pork bone, an abundant biomass source and food waste, has been converted into structured porous hydroxyapatite by a three‐step process including precharring under mild conditions, chemical activation, and thermal activation. The investigated activating agents were NaOH, KOH, K2CO3, H2SO4, and H3PO4. A thorough investigation of the influence of different activating protocols on the chemical and textural properties of the produced material was carried out by nitrogen adsorption–desorption at 77 K, potentiometric titrations, Fourier transform infrared, and X‐ray diffraction techniques. Chemical activation with NaOH, K2CO3, and H2SO4 increased the specific surface area up to 53%. H3PO4 reduced both surface area and pore volume, and KOH showed little influence on the pore structure. The produced materials were evaluated by methylene blue adsorption tests and showed significant improvement as a result of chemical activation. As a main effect, acid treatment increased methylene blue adsorption kinetics, probably owing to an increase in micropororosity, whereas alkali activation enhanced the adsorption capacity of the resultant biochar.  相似文献

Rare-earth chromite ErCrO3 powder was synthesized from metal nitrate precursors using microwave synthesis. (Micro-) structural characterizations were performed using X-ray diffraction, Rietveld refinement, High Resolution Transmission Electron Microscopy and Electron Micro-Diffraction. Magnetization vs. temperature measurements revealed anti-ferromagnetism with TNeel ≈ 135 K. An anti-ferrimagnetic moment of ≈0.4 μB was determined from magnetization vs. applied field measurements. Temperature dependent impedance spectroscopy (IS) indicated 3 dielectric relaxation processes: electrode interface, grain boundary and bulk. The intrinsic bulk activation energy was found to be 0.27 eV and the dielectric permittivity ?r was ≈23 in excellent agreement with Clausius-Mossotti predictions and showed no perceptible temperature dependence. This and the low ?r value suggested that ErCrO3 is a dielectric rather than ferroelectric compound. IS measurements with applied dc bias revealed the signs of an unconventional type of Schottky barrier at the metallic Au electrode/ceramic ErCrO3 interface.  相似文献

Herein we present the design, synthesis, and biological evaluation of potent and highly selective β-secretase 2 (memapsin 1, beta-site amyloid precursor protein cleaving enzyme 2, or BACE 2) inhibitors. BACE2 has been recognized as an exciting new target for type 2 diabetes. The X-ray structure of BACE1 bound to inhibitor 2 a {N3-[(1S,2R)-1-benzyl-2-hydroxy-3-[[(1S,2S)-2-hydroxy-1-(isobutylcarbamoyl)propyl]amino]propyl]-5-[methyl(methylsulfonyl)amino]-N1-[(1R)-1-phenylpropyl]benzene-1,3-dicarboxamide} containing a hydroxyethylamine isostere was determined. Based on this structure, a computational docking study was performed which led to inhibitor 2 a -bound BACE2 models. These were used to optimize the potency and selectivity of inhibitors. A systematic structure–activity relationship study led to the identification of determinants of the inhibitors’ potency and selectivity toward the BACE2 enzyme. Inhibitors 2 d [N3-[(1S,2R)-1-benzyl-2-hydroxy-3-[[(1S,2S)-2-hydroxy-1-(isobutylcarbamoyl)pentyl]amino]propyl]-N1-methyl-N1-[(1R)-1-phenylpropyl]benzene-1,3-dicarboxamide; Ki=0.031 nm , selectivity over BACE1: ≈174 000-fold] and 3 l [N1-((2S,3R)-3-hydroxy-1-phenyl-4-((3-(trifluoromethyl)benzyl)amino)butan-2-yl)-N3,5-dimethyl-N3-((R)-1-phenylethyl)isophthalamide; Ki=1.6 nm , selectivity over BACE1: >500-fold] displayed outstanding potency and selectivity. Inhibitor 3 l is nonpeptide in nature and may pave the way to the development of a new class of potent and selective BACE2 inhibitors with clinical potential.  相似文献

Ceria catalysts were found active and selective to the oxidehydrogenation of ethane (ODE) with CO2 and the actual contribution for C2H4 formation from heterogeneous catalysis was 75–55% in the range 953–993 K. The presence of calcium ions in solid solution in the ceria crystalline network increased significatively the selectivity to ethene and the efficiency of CO2 as oxidant in the heterogeneous reaction.  相似文献

Colloidal Co nanoparticles with sizes in the 3–8 nm range were obtained by thermal decomposition of Co2(CO)8 in the presence of ligands and impregnated on SiO2 to prepare SiO2-supported Co nanocatalysts. The catalysts showed activity for the steam reforming of ethanol with higher values for smaller Co particles. H2 adsorption results and Fourier transform infrared spectroscopy of adsorbed CO suggested that the fraction of accessible Co sites also depended on the synthesis conditions. Precipitation of the Co nanoparticles with methanol instead of ethanol before impregnation had a positive effect on the density of accessible Co sites to catalysis; similar result was verified by increasing the thermal treatment temperature under H2 flow before the reaction. Based on the distribution of products with temperature of reaction, a mechanism for steam reforming of ethanol on SiO2-supported Co nanocatalysts is suggested.  相似文献

In this work, oil obtained from seeds of different red grape varieties, grown in the Autonomous Regions of Castilla‐La Mancha and Murcia (Spain), was characterized by determining physicochemical and sensory quality parameters, stability, and the composition in fatty acids and sterols. The physicochemical quality parameters (free acidity, peroxide index, K270 and wax) scored high (meaning low quality) compared with virgin olive oils, while the negative sensory attributes stood out over the positive ones. Therefore, the oil was not considered suitable for table use without undergoing a refining process. The samples showed high linoleic and low linolenic acid contents, while β‐sitosterol was the main sterol found. Drying grape seeds with hot air before extraction gave higher physicochemical quality, total phenolic content and stability, and lower wax content in comparison to air‐drying of seeds. The drying process affected the sterol composition but not the fatty acid composition.  相似文献

Osteoporosis is the most common bone disease characterized by reduced bone mass and increased bone fragility. Genetic contribution is one of the main causes of primary osteoporosis; therefore, both genders are affected by this skeletal disorder. Nonetheless, osteoporosis in men has received little attention, thus being underestimated and undertreated. The aim of this study was to identify novel genetic variants in a cohort of 128 males with idiopathic low bone mass using a next-generation sequencing (NGS) panel including genes whose mutations could result in reduced bone mineral density (BMD). Genetic analysis detected in eleven patients ten rare heterozygous variants within the LRP5 gene, which were categorized as VUS (variant of uncertain significance), likely pathogenic and benign variants according to American College of Medical Genetics and Genomics (ACMG) guidelines. Protein structural and Bayesian analysis performed on identified LRP5 variants pointed out p.R1036Q and p.R1135C as pathogenic, therefore suggesting the likely association of these two variants with the low bone mass phenotype. In conclusion, this study expands our understanding on the importance of a functional LRP5 protein in bone formation and highlights the necessity to sequence this gene in subjects with idiopathic low BMD.  相似文献

The effect of the addition of CeO2 or La2O3 on the surface properties and catalytic behaviors of Al2O3-supported Pd catalysts was studied in the steam reforming of methane. The FTIR spectroscopy of adsorbed CO and the Pd dispersion suggest the partial coverage of Pd0 by ceria or lanthana species. This could lead to the formation of an adduct MPd x O (M = Ce or La) at the surface of the metal crystallites. The addition of ceria or lanthana resulted in an increase of the turnover rate and specific rate for steam reforming of methane. One possible explanation if that the Pd0*Pdδ+O–M interfacial species (M = Ce or La) are oxidized by H2O or CO2, promoting the O* transfer to the metal surface. This could facilitate the removal of C* species from the metal surface, resulting in the increase of specific reaction rate and increase of the accessibility of CH4 to metal active sites.  相似文献

Post-translational modification of the DNA replication machinery by ubiquitin and SUMO plays key roles in the faithful duplication of the genetic information. Among other functions, ubiquitination and SUMOylation serve as signals for the extraction of factors from chromatin by the AAA ATPase VCP. In addition to the regulation of DNA replication initiation and elongation, we now know that ubiquitination mediates the disassembly of the replisome after DNA replication termination, a process that is essential to preserve genomic stability. Here, we review the recent evidence showing how active DNA replication restricts replisome ubiquitination to prevent the premature disassembly of the DNA replication machinery. Ubiquitination also mediates the removal of the replisome to allow DNA repair. Further, we discuss the interplay between ubiquitin-mediated replisome disassembly and the activation of CDK1 that is required to set up the transition from the S phase to mitosis. We propose the existence of a ubiquitin–CDK1 relay, where the disassembly of terminated replisomes increases CDK1 activity that, in turn, favors the ubiquitination and disassembly of more replisomes. This model has important implications for the mechanism of action of cancer therapies that induce the untimely activation of CDK1, thereby triggering premature replisome disassembly and DNA damage.  相似文献
