Electron transfer in proteins is essential in crucial biological processes. Although the fundamental aspects of biological electron transfer are well characterized, currently there are no experimental tools to determine the atomic‐scale electronic pathways in redox proteins, and thus to fully understand their outstanding efficiency and environmental adaptability. This knowledge is also required to design and optimize biomolecular electronic devices. In order to measure the local conductance of an electrode surface immersed in an electrolyte, this study builds upon the current–potential spectroscopic capacity of electrochemical scanning tunneling microscopy, by adding an alternating current modulation technique. With this setup, spatially resolved, differential electrochemical conductance images under bipotentiostatic control are recorded. Differential electrochemical conductance imaging allows visualizing the reversible oxidation of an iron electrode in borate buffer and individual azurin proteins immobilized on atomically flat gold surfaces. In particular, this method reveals submolecular regions with high conductance within the protein. The direct observation of nanoscale conduction pathways in redox proteins and complexes enables important advances in biochemistry and bionanotechnology.  相似文献

Stem cells secrete paracrine factors including extracellular vesicles (EVs) which can mediate cellular communication and support the regeneration of injured tissues. Reduced oxygen (hypoxia) as a key regulator in development and regeneration may influence cellular communication via EVs. We asked whether hypoxic conditioning during human induced pluripotent stem cell (iPSC) culture effects their EV quantity, quality or EV-based angiogenic potential. We produced iPSC-EVs from large-scale culture-conditioned media at 1%, 5% and 18% air oxygen using tangential flow filtration (TFF), with or without subsequent concentration by ultracentrifugation (TUCF). EVs were quantified by tunable resistive pulse sensing (TRPS), characterized according to MISEV2018 guidelines, and analyzed for angiogenic potential. We observed superior EV recovery by TFF compared to TUCF. We confirmed hypoxia efficacy by HIF-1α stabilization and pimonidazole hypoxyprobe. EV quantity did not differ significantly at different oxygen conditions. Significantly elevated angiogenic potential was observed for iPSC-EVs derived from 1% oxygen culture by TFF or TUCF as compared to EVs obtained at higher oxygen or the corresponding EV-depleted soluble factor fractions. Data thus demonstrate that cell-culture oxygen conditions and mode of EV preparation affect iPSC-EV function. We conclude that selecting appropriate protocols will further improve production of particularly potent iPSC-EV-based therapeutics.  相似文献

The authors report their experience of the application of microsurgical techniques in total and subtotal thyroidectomy operations from January 1984-January 1997. Four hundred and eighty-six patients with thyroid diseases were operated upon with both traditional and microsurgical techniques. The comparison of results shows that microsurgical skills are effective in reducing the dangers and important complications in both laryngeal nerves and parathyroids.  相似文献

Ontologies are shared models of a domain that encode a view which is common to a set of different parties. Contexts are local models that encode a party’s subjective view of a domain. In this paper, we show how ontologies can be contextualized, thus acquiring certain useful properties that a pure shared approach cannot provide. We say that an ontology is contextualized or, also, that it is a contextual ontology, when its contents are kept local, and therefore not shared with other ontologies, and mapped with the contents of other ontologies via explicit (context) mappings. The result is Context OWL (C-OWL), a language whose syntax and semantics have been obtained by extending the OWL syntax and semantics to allow for the representation of contextual ontologies.  相似文献

The implementation of a versatile VLSI chip certainly represents an important step to improve the research on Cellular Neural Networks. In this paper a VLSI realization of the multi-chip oriented, the 6 × 6 Digitally Programmable Cellular Neural Network (6 × 6 DPCNN) chip, will be presented. This chip covers most of the available one-neighbourhood templates for image processing applications. Moreover, it can be easily interconnected to others to carry out very large CNN arrays. The designs and some measured results of a single chip and a multi-chip board (the 720 DPCNN System) will be shown.  相似文献

The reaction mechanism and the rate-determining step (RDS) of the isomerisation of n-alkanes (C4–C6) over partially reduced MoO3 catalysts were studied through the effects of the addition of an alkene isomerisation catalyst (i.e. CoAlPO-11). When an acidic CoAlPO-11 sample was mechanically mixed with the MoO3, a decrease of the induction period and an increase of the steady-state conversion of n-butane to isobutane were observed. These data support previous assumptions that a bifunctional mechanism occurred over the partially reduced MoO3 (a complex nanoscale mixture of oxide-based phases) during n-butane isomerisation and that the RDS was the skeletal isomerisation of the linear butene intermediates. The only promotional effect of CoAlPO-11 on the activity of partially reduced MoO3 for C5–C6 alkane hydroisomerisation was a reduction of the induction period, as the RDS at steady-state conditions appeared to be dehydrogenation of the alkane in this case. However, lower yields of branched isomers were observed in this case, the reason of which is yet unclear.  相似文献

The aim of this work is to study from an experimental point of view the oxidative steam reforming of methanol by investigating the behaviour of a dense Pd/Ag membrane reactor (MR) in terms of methanol conversion as well as hydrogen production. The main parameters considered are the operating temperature and the O2/CH3OH feed ratio. This is a pioneer work in the application of MR to this kind of reaction, whose goal should be to produce a CO-free hydrogen stream suitable for hydrogen fuel cell applications. The experimental results show that the MR gives methanol conversions higher than traditional reactors (TRs) at each temperature investigated, confirming the good potential of the membrane reactor device for this interesting reaction system.  相似文献
