Checkpoints maintain the order of cell-cycle events. At G2/M, a checkpoint blocks mitosis in response to damaged or unreplicated DNA. There are significant differences in the checkpoint responses to damaged DNA and unreplicated DNA, although many of the same genes are involved in both responses. To identify new genes that function specifically in the DNA replication checkpoint pathway, we searched for high-copy suppressors of overproducer of Cdc25p (OPcdc25(+)), which lacks a DNA replication checkpoint. Two classes of suppressors were isolated. One class includes a new gene encoding a putative DEAD box helicase, suppressor of uncontrolled mitosis (sum3(+)). This gene negatively regulates the cell-cycle response to stress when overexpressed and restores the checkpoint response by a mechanism that is independent of Cdc2p tyrosine phosphorylation. The second class includes chk1(+) and the two Schizosaccharomyces pombe 14-3-3 genes, rad24(+) and rad25(+), which appear to suppress the checkpoint defect by inhibiting Cdc25p. We show that rad24Delta mutants are defective in the checkpoint response to the DNA replication inhibitor hydroxyurea at 37 degrees and that cds1Delta rad24Delta mutants, like cds1Delta chk1Delta mutants, are entirely checkpoint deficient at 29 degrees. These results suggest that chk1(+) and rad24(+) may function redundantly with cds1(+) in the checkpoint response to unreplicated DNA.  相似文献

Extracellular and whole-cell patch clamp recordings were used to study the excitatory responses elicited by purine nucleotides in pontine slices of the rat brain containing the locus coeruleus (LC). The P2 purinoceptor agonists, alphabeta-methyleneadenosine 5'-triphosphate (alphabetameATP) and adenosine 5'-O-(2-thiodiphosphate) (ADPalphabetaS), and a novel purinoceptor agonist, alphabeta-methyleneadenosine 5'-diphosphate (alphabetameADP), elicited concentration-dependent increases in the spontaneous firing rate over the concentration range (1-300 microM). On vagus nerve or dorsal root preparations alphabetameADP (100 microM) had no agonist activity. In the presence of both alphabetameATP (300 microM), ADPbetaS (300 microM) elicited a further and significant increase in the firing rate of the LC neurones, whilst neither alphabetameATP nor alphabetameADP (300 microM) elicited a further response. The P2 purinoceptor antagonists, suramin (100 microM) and pyridoxalphosphate-6-azophenyl-2',4'-disulphonic acid (PPADS; 30 microM), markedly attenuated responses to all three agonists. Whole-cell recording of membrane current showed that, at - 60 mV, alphabetameATP and alphabetameADP (both 100 microM) elicited inward currents of a similar magnitude, whilst the inward currents elicited by a lower concentration of ADPbetaS (30 microM) were larger and faded in the presence of this agonist. In the presence of tetrodotoxin and a combination of other neurotransmission blockers, both alphabetameATP and alphabetameADP still produced inward currents. Based on the known selectivity of the agonists used in this study, there appear to be two distinct P2 purinoceptor types present on neurones in the LC, which correspond to the P2X and P2Y types. The responses elicited by alphabetameADP appear to be mediated through a putative P2X purinoceptor, although further work is required to determine which P2X receptor subtype(s) are involved.  相似文献

We have perforated a series of experiments to study cavitation in superfluid helium into which electrons are injected by field-emission from a sharp tip. The injected electrons force open small cavities in the liquid (“electron bubble”). These objects explode at a critical negative pressure P c, and in previous experiments we have studied the cavitation that resugts from these explosions. In the present experiments we have detected cavitation events that occur before a negative pressure as large as P c is reached. We suggest that these events may arise from a process in which two neutral helium dimers interact and an electron is injected into the liquid through Penning ionization.  相似文献

Coupled active antenna oscillator arrays are used for power-combining at microwave and millimeter-wave frequencies. It is known that the relative phase determined by the element separation distance ultimately determines the array operational mode and, hence, far-field radiation characteristics. Separately, it is known that coupled oscillator modal stability is achieved by coupling oscillators through lumped capacitive elements. In this paper, an arrangement whereby lumped capacitive elements (placed across the oscillator loads) and radiative coupling are employed concurrently is investigated. The arrangement takes the form of a ring of coupled oscillators used to excite a 2×2 antenna array. The effect that these couplings have on array behavior are evaluated using time-domain analysis and analytically derived equations. Experimental results for far-field radiation patterns are discussed in relation to coupled oscillator dynamical behavior. These suggest that the theoretical predictions are valid, offering a robust design tool for studies of larger power-combining arrays  相似文献

Small-conductance, ATP-sensitive K(+)-channels (KATP) localized in apical membranes of both thick ascending limb of the loop of Henle and cortical collecting duct cells may be involved in Na+ reabsorption and K+ secretion in the mammalian kidney. Possible pharmacologic tools to evaluate such an hypothesis may be the antidiabetic sulfonylureas which block K(+)-channels in pancreatic beta-cells. In saline-loaded conscious rats, glyburide (GLY) dose-dependently increased urinary Na+ excretion with little change in urinary K+ excretion after i.p. administration (10-100 mg/kg). In renal clearance studies, GLY at 25 mg/kg i.v. increased Na+ excretion 350% during the first hour post-treatment without affecting K+ excretion, glomerular filtration rate, mean arterial pressure or heart rate. GLY at 50 mg/kg was no more natriuretic than the 25 mg/kg dose, whereas 12.5 mg/kg of GLY increased Na+ excretion 200%. The change in Na+ excretion produced by 25 mg/kg of GLY in streptozotocin-induced diabetic rats was significantly greater than the change after drug vehicle in these animals. It is unlikely that the natriuresis produced by GLY is secondary to changes in plasma insulin and/or glucose because the doses used were far above GLY's insulin-releasing action (i.e., all natriuretic doses would have produced maximal insulin release) and GLY was natriuretic in streptozotocin-induced diabetic rats. It is possible that GLY interferes with reabsorption of Na+ by blocking KATP and thereby interrupting K+ recycling and Na(+)-2Cl(-)-K+ cotransport in the loop of Henle.  相似文献

Herein is a review of clear cell neoplasms of selected sites in the urinary tract and male reproductive system, including the kidney, the urinary bladder, testis, epididymis, and prostate. Clear cell cytoplasmic alteration in neoplasms at these sites is a relatively common light microscopic finding. Examples of such neoplasms with clear cell change include the clear cell type of renal cell carcinoma, clear cell adenocarcinoma of urethra and bladder, the classic type of seminoma, papillary cystadenoma of the epididymis, and well-differentiated adenocarcinoma of the prostate. Of importance, numerous non-neoplastic benign entities may also manifest cleared cytoplasm and therefore are presented in the differential in this review. Indeed, knowledge of the neoplastic and non-neoplastic entities displaying clear cell change at each anatomic site should enable the surgical pathologist to approach the differential diagnosis of these conditions in a more logical and rigorous fashion.  相似文献

Describes how the 2nd author's (see record 1975-02991-001) structural analysis of social behavior (SASB) offers a new approach to the old problem of how to measure the complex and elusive concepts that are critical to family theory and therapy. SASB includes a theoretical model and an assessment methodology that can be applied to disturbed family relationships and interaction patterns. It also provides operational definitions and measures of distressed family processes that have been linked to major psychopathology, such as double binds and pseudomutuality. The conceptual framework of SASB is compatible with family systems and psychodynamic theories, and it applies to a range of healthy and distressed family functioning. SASB's promise for family assessment is illustrated, and an overview of the SASB model and methods is presented. A comprehensive library of magnetic bubble logic components has been established, with emphasis on standardization in terms of dimensions and I/O to facilitate VLSI chip composition. These components were designed in a current-access perforated-sheet configuration and fabricated on magnetic garnet wafers supporting 2-µm bubbles. The operating margin for each component was studied individually on 8-µm period devices at 1 MHZ by using a high-speed magneto-optical sampling-camera system. The bubble-to-bubble interaction force was found to be very reliable in producing successful logic operation with a 12-percent bias-field margin for the XOR/AND gate equal to 80 percent of the bubble-propagation margin and about 60 percent of the free-bubble bias-field margin. The experimental results show 10.8, 10.7, 10.2, 10, 10.6, 8.5, 12, 8.5, 15, and 9.5-percent bias-field margins for the SWITCH, AND/OR, COMPARATOR, SEARCH, LATCH, BIT-PAIR SEPARATOR, MERGER, CROSSOVER, ANNIHILATOR, and SPLITTER gates, respectively. An 8.5-percent overlapped bias margin has been obtained for all of the logic components. Transient pictures taken with a small incremented delay revealed that the bubble motion in perforated-sheet logic devices is uniform both in the velocity excursion and shape distortion. The behavior of various components at different values of the bias field and drive current is all qualitatively interpretable on the basis of experimental observations. The feasibility of these logic components is successfully demonstrated.  相似文献

We report studies of the negative pressure required to lead to the nucleation of bubbles in superfluid helium-4 in the temperature range down to 0.8 K. At the lowest temperatures studied, the tensile strength is found to be 3 bar. The results are compared with nucleation theory and the possible importance of nucleation on vortices is discussed.  相似文献
