Heavy metals polluting the 100-year-old waste heap in Bolesław (Poland) are acting as a natural selection factor and may contribute to adaptations of organisms living in this area, including Trifolium repens and its root nodule microsymbionts—rhizobia. Exopolysaccharides (EPS), exuded extracellularly and associated with bacterial cell walls, possess variable structures depending on environmental conditions; they can bind metals and are involved in biofilm formation. In order to examine the effects of long-term exposure to metal pollution on EPS structure and biofilm formation of rhizobia, Rhizobium leguminosarum bv. trifolii strains originating from the waste heap area and a non-polluted reference site were investigated for the characteristics of the sugar fraction of their EPS using gas chromatography mass-spectrometry and also for biofilm formation and structural characteristics using confocal laser scanning microscopy under control conditions as well as when exposed to toxic concentrations of zinc, lead, and cadmium. Significant differences in EPS structure, biofilm thickness, and ratio of living/dead bacteria in the biofilm were found between strains originating from the waste heap and from the reference site, both without exposure to metals and under metal exposure. Cell transplantation has been studied extensively as a therapeutic strategy for neurological disorders. However, to date, its effectiveness remains unsatisfactory due to low precision and efficacy of cell delivery; poor survival of transplanted cells; and inadequate monitoring of their fate in vivo. Fortunately, different bio-scaffolds have been proposed as cell carriers to improve the accuracy of cell delivery, survival, differentiation, and controlled release of embedded stem cells. The goal of our study was to establish hydrogel scaffolds suitable for stem cell delivery that also allow non-invasive magnetic resonance imaging (MRI). We focused on alginate-based hydrogels due to their natural origin, biocompatibility, resemblance to the extracellular matrix, and easy manipulation of gelation processes. We optimized the properties of alginate-based hydrogels, turning them into suitable carriers for transplanted cells. Human adipose-derived stem cells embedded in these hydrogels survived for at least 14 days in vitro. Alginate-based hydrogels were also modified successfully to allow their injectability via a needle. Finally, supplementing alginate hydrogels with Mn ions or Mn nanoparticles allowed for their visualization in vivo using manganese-enhanced MRI. Although the outcome has improved over the past decades, due to improved supportive care, a better understanding of risk factors, and intensified chemotherapy, pediatric acute myeloid leukemia remains a life-threatening disease, and overall survival (OS) remains near 70%. According to French-American-British (FAB) classification, AML is divided into eight subtypes (M0–M7), and each is characterized by a different pathogenesis and response to treatment. However, the curability of AML is due to the intensification of standard chemotherapy, more precise risk classification, improvements in supportive care, and the use of minimal residual disease to monitor response to therapy. The treatment of childhood AML continues to be based primarily on intensive, conventional chemotherapy. Therefore, it is essential to identify new, more precise molecules that are targeted to the specific abnormalities of each leukemia subtype. Here, we review abnormalities that are potential therapeutic targets for the treatment of AML in the pediatric population. 相似文献

A photopolymer holographic grating is used to produce the two sheared images in an electronic speckle pattern shearing interferometer. A ground glass screen following the grating eliminates unwanted diffraction orders and removes the requirement for the CCD camera to resolve the diffraction grating's pitch. The sheared images on the ground glass are further imaged onto the CCD camera. The fringe pattern contrast was estimated to be above 90%. A validation of the system was done by comparing the theoretical phase difference distribution with the experimental data from the three-point bending test. 相似文献

Results from the investigation of the diffusion processes in a dry acrylamide-based photopolymer system are presented. The investigation is carried out in the context of experimental research on optimization of the high-spatial-frequency response of the photopolymer. Tracing the transmission holographic grating dynamics at short times of exposure is utilized to measure diffusion coefficients. The results reveal that two different diffusion processes contribute with opposite sign to the refractive-index modulation responsible for the diffraction grating buildup. Monomer diffusion from dark to bright fringe areas increases the refractive-index modulation. It is characterized with diffusion constant D0 = 1.6 x 10(-7) cm2/s. A second diffusion process takes place during the recording. It decreases the refractive-index modulation and we ascribe it to diffusion of short-chain polymer molecules or radicals from bright to dark fringe areas. The estimated diffusion coefficient for this process is D0 = 6.35 x 10(-10) cm2/s. The presence of the second process could be responsible for the poor high-spatial-frequency response of the investigated photopolymer system. Comparison with the diffusion in photopolymer systems known for their good response at high spatial frequencies shows that both investigated diffusion processes occur in a much faster time scale. 相似文献

The paper presents a general method to evaluate a masonry structure with a complex geometry and unknown material properties. This multidisciplinary approach integrates close range digital photogrammetry, ground probing radar (GPR) and finite elements analysis (FEM) to analyse a masonry bridge. The resulting information from photogrammetry and radar is applied to define a 3D model, which is then used to simulate the behaviour of the bridge using FEM. Two different approaches to model the internal composition of the bridge with finite elements are presented. Sensitivity analysis is applied to study the reasons of the structural damage. 相似文献

This paper presents an analysis of colour change in concrete under the influence of heat. The colour change observed in concrete is primarily a result of the gradual dehydration of the cement paste, but also of transformations occurring within the aggregate. The colour change may be used to reveal the exposure temperature of concrete from which the corresponding fire damage of concrete can be estimated. The paper presents the results of tests carried out on ordinary and high performance concretes (OC and HPC) prepared with natural river-bed aggregates. In addition, mortars and cement pastes prepared with the same components were observed to change colour upon heating. The colour change was investigated using Scion Image v. 4.0.3, an image analysis software package (Scion Corporation ©, USA). In the proposed method the digital image is split into three RGB colour components: red, green and blue, which are then presented as a histogram using counts of pixel intensity. The histogram results show colour distributions in unheated cementitious material and in material heated to temperatures ranging from 100°C to 1000°C. The concrete colour changes as a result of heating are linked to the physical and chemical transformations taking place in the heated material. 相似文献

A new catalyst based on biosilica doped with palladium(II) chloride nanoparticles was prepared and tested for efficient degradation of methyl orange (MO) in water solution under UV light excitation. The obtained photocatalyst was characterized by X-ray diffraction, TEM and N2 adsorption/desorption isotherms. The photocatalytic degradation process was studied as a function of pH of the solution, temperature, UV irradiation time, and MO initial concentration. The possibilities of recycling and durability of the prepared photocatalysts were also tested. Products of photocatalytic degradation were identified by liquid chromatography–mass spectrometry analyses. The photocatalyst exhibited excellent photodegradation activity toward MO degradation under UV light irradiation. Rapid photocatalytic degradation was found to take place within one minute with an efficiency of 85% reaching over 98% after 75 min. The proposed mechanism of photodegradation is based on the assumption that both HO• and O2•− radicals, as strongly oxidizing species that can participate in the dye degradation reaction, are generated by the attacks of photons emitted from diatom biosilica (photonic scattering effect) under the influence of UV light excitation. The degradation efficiency significantly increases as the intensity of photons emitted from biosilica is enhanced by palladium(II) chloride nanoparticles immobilized on biosilica (synergetic photonic scattering effect). 相似文献

This paper is an attempt to determine the actual boron adsorption and retention in wood and its isolated components: cellulose and lignin. The rate of boron retention in cellulose, lignin and wood powder of Scots pine Pinus silvestris L. was investigated using denaturing properties of protein contained in reaction solutions. Both animal and plant origin protein was used in this study. In order to determine the reactivity of the analyzed formulation the reactions of cellulose, lignin and wood powder with protein borates were carried out. The aim of the analyses was also to compare methods of protein denaturation: thermal and chemical with the use of tannic acid solution. The obtained products were subjected to water extraction at 20 °C. Boron content in the solid and water extracts was analyzed using atomic absorption spectrometry (AAS). The results showed that the method of protein denaturation played the crucial role in the rate of boron retention. The type of protein used in the reaction solution had also significant effect on the boron retention. 相似文献
