A novel chitinase gene of tobacco was isolated and characterized by DNA sequence analysis of a genomic clone and a cDNA clone. Comparative sequence analysis of both clones showed an identity of 94%. The proteins encoded by these sequences do not correspond to any of the previously characterized plant chitinases of classes I-IV and are designated as class V chitinases. Comparison of the chitinase class V peptide sequence with sequences in the Swiss Protein databank revealed significant sequence similarity with bacterial exo-chitinases from Bacillus circulans, Serratia marcescens and Streptomyces plicatus. It was demonstrated that class V chitinase gene expression is induced after treatment of tobacco with different forms of stress, like TMV-infection, ethylene treatment, wounding or ultraviolet irradiation. Two related chitinase class V proteins of 41 and 43 kDa were purified from Samsun NN tobacco leaves inoculated with tobacco mosaic virus. The proteins were purified by Chelating Superose chromatography and gel filtration. In vitro assays demonstrated that class V chitinases have endo-chitinase activity and exhibit antifungal activity toward Trichoderma viride and Alternaria radicina. In addition, it was shown that class V chitinase acts synergistically with tobacco class I beta-1,3-glucanase against Fusarium solani germlings.  相似文献

The stability and applicability of a previously developed inverse procedure for the noninvasive determination of the activation sequence of the human heart has been evaluated. In particular, the possibility of using a standard geometrical configuration representing the heart and the inhomogeneous volume conductor in this procedure has been tested. Results show that in order to obtain reliable inverse solutions, true "tailored" geometry should be used.  相似文献

The effects of time of boning and storage period on creatine kinase (CK) activity, transmission value, drip losses and water-holding capacity (WHC) measured by various methods was investigated. At 40 min post-mortem 30 pig carcasses with pH values > 6·2 in the loin were selected. The right loin of each carcass was hot boned and vacuum packaged immediately. The left loin was cold boned and packaged after overnight chilling at 2 ± 2°C. After 1, 5 and 12 days of storage at 0 ± 1°C, 10 hot- and cold-boned loins were unpacked and sampled. Time of boning did not affect drip losses. At 1 day post-mortem the CK activity was higher in hot- than in cold-boned pork. Sarcomere lengths were not affected by time of boning. The WHC was investigated by two controlled methods. A filter-paper press method was not sensitive enough to assess differences in WHC of hot- vs cold-boned pork. The results of a gravimetric method depended on the sample location. Results of the gravimetric test decreased with increasing storage periods. The results suggest that under the experimental conditions the chilling rates of hot-boned vs carcass-attached muscles were similar.  相似文献

A model for surface acoustic wave (SAW) transversal filters with special attention to the system and circuit designer's point of view is summarized. In the ideal situation the SAW transversal filter is driven with a voltage and the short-circuit current is sensed, which results in a minimization of the triple transit echo distortion. The aperture (width) of the SAW device is the only parameter that is not determined by the frequency dependence of the transfer and therefore it can be used to optimize the SAW device in relation to the electronic circuitry. Noise and dynamic range calculations on an amplifier-filter-amplifier configuration, are performed. It is shown that for a low noise floor at the input of the SAW device, the aperture of this device should be chosen large. However, the dynamic range of the amplifier-filter-amplifier configuration can be maximized by choosing a small aperture.  相似文献

The thermal conductivity, , is one of the few transport coefficients which shows a relatively small change at the solid-liquid phase transition, and hence it is a property that can be used in comparing dynamic properties of both ordered and disordered systems. Although the discontinuity in can be accounted for largely by the difference in density, , of solid and liquid at the phase transition, its volume dependence is examined more closely. The thermal diffusivity, which is known to dominate the dynamic structure factor of liquid argon, has been determined around the phase transition also; the sound velocity has been considered in addition. In an attempt to elaborate good manufacturing practices, including the collection, processing and storage of porcine gullets, their bacterial condition immediately after collection (100 samples), as well as that of deep frozen gullets just before incorporation into meat products (40 samples), was assessed. Fresh gullets were found to be contaminated to a high degree: poured plate colony count at 30°C (PPCC) approximately 10(6) to 10(7) and Enterobacteriaceae approximately 10(3) to 10(4) cfu g(-1). Deep frozen gullets showed even higher counts: PPCC approximately 10(7) to 10(8) and Enterobacteriaceae approximately 10(4) to 10(5) cfu g(-1). Hygiene during collection was visually assessed in six abattoirs and found to be satisfactory in two, moderate in three and poor in one. The effects of processing, by cleaning or removal of the mucus membrane, on bacterial condition, pH, colour and odour were assessed before and during storage at 4°C and 20°C. Both cleaning and removal of the mucus membrane resulted in up to approximately a tenfold reduction of colony counts. After 7 days' storage at 4°C these were significantly lower than those of unprocessed gullets (P < 0·01). Processed gullets stored at 4°C were no longer fit for consumption after 4 days' storage. It being impossible to achieve a marked improvement in the bacteriological condition of gullets, the incorporation of these products into sausages should be discouraged and their use in petfoods only allowed under reasonable conditions of hygiene and chilling.  相似文献

Summary The purpose of this study was to find experimental conditions for the complete solubility of collagen-free muscle proteins (CFMP) using acetone powder of Guelders ring sausage. Preliminary experiments were carried out to choose the best procedure for preparing the acetone dry powder. Two different methods of acetone extraction of minced sausage were compared. The acetone dry mass (ADM) method using continuous extraction in a Soxhlet [2] apparatus gave better results than the acetone powder (ACP) method, which used a blender [1]. The ADM method was used for further investigations. ADM was extracted with two types of sodium dodecyl sulphate (SDS), containing solvents A and B. Solvent A contains a Tris-boric acid buffer (pH 8.2) with 1.5% (m/v) SDS and 0.05% (m/v) dithioerythreitol [3]. Solvent B is a borate-chloric acid buffer (pH 9.0) with 2.0% (m/v) SDS and 1.0% (m/v) mercapto-ethanol [2]. Both solvents showed a linear relationship between the quantities of CFMP in ADM and the dissolved CFMP. The linear relationships were found between quantities of 10.0 and 30.0 mg (solution A) and of 5.0 and 30.0 mg ADM (solution B) per ml solvent. The solubility of CFMP was better in solvent B than in solution A. Completely dissolved CFMP from ADM was only obtained in the case of 5.0 mg ADM in 1.0 ml solution B. These conditions will be used in liquid chromatography experiments, the results of which will be reported later.
Quantitative Aspekte zweier Verfahren für das Auflösen kollagenfreier Muskelproteine aus acetontrockenen Pulvern der Gelderschen Rauchwurst
Zusammenfassung Der Zweck dieser Untersuchungen ist, die experimentellen Bedingungen für die vollstän-dige Löslichkeit des kollagenfreien Muskelproteins (CFMP) aus dem Acetonpulver der Gelderschen Rauchwurst zu finden. Durch Vorversuche wurde die beste Arbeitsweise für das Zubereiten des Acetonpulvers gewählt. Zwei verschiedene Extraktionsverfahren mit zerkleinertem Wurstmaterial wurden miteinander verglichen. Die Methode mit der Acetontrockenmasse (ADM) mittels kontinuierlicher Extraktion [2] führte zu besseren Ergebnissen als die Methode mit Acetonpulver (ACP), wozu ein Mischgerät [1] verwendet wurde. Die ADM-Methode wurde für weitere Untersuchungen angewendet. ADM wurde mit zwei verschiedenen Extraktionslösungen von Natrium-Dodecylsulfat (SDS) (A und B) extrahiert. Lösung A enthalt einen Tris-Borsäure Puffer (pH 8,2) mit SDS (1,5%) und Dithioerithritol (0,05%) [3]. Die Lösung B enthält einen Borat-Salzsäure Puffer (pH 9,0) mit SDS (2,0%) und Mercapto-Ethanol (1,0%) [2]. Beide Extraktionslösungen zeigen ein lineares Verhalten zwischen den Mengen vom CFMP in ADM und in aufgelöstem CFMP. Diese Linearität wurde von 10,030,0 mg ADM (Lösungsmittel A) und von 5,030,0 mg ADM (Lösungsmittel B) gefunden. Die Lös-lichkeit in Lösung B ist gegeniiber Lösung A besser. Ein vollständig gelöstes CFMP aus ADM wurde nur bei der Extraktion von 5,0 mg ADM in 1,0 ml der Losung B erhalten. Diese Bedingung soll in unseren künftigen flüssigchromatographischen Experimenten verwendet werden.

Organofunctional silanes recently have emerged as outstanding, environmentally friendly corrosion protectors for metal substrates, compared with conventional chromate treatments. A simple immersion technique is typically used to coat the metal surface with silane films. However, the thickness and uniformity of the films are uncontrolled in this process. This paper proposes a new deposition technique for the silane films on the metal surface, i.e., by electrodeposition. Hydrolyzed silanes are water-soluble, ionized molecules, so they can be deposited on metals by electrodeposition. Various combinations of silane mixtures were tested at different voltages, pH values, bath concentrations, and exposure times on panels of alloy aluminum and mirror-polished ferro-plate. The surface structure was characterized by scanning electron microscopy (SEM) and ellipsometry. The resistance of the film to corrosion was investigated by direct current (DC) polarization and electrochemical impedance spectroscopy (EIS) techniques. Electrodeposition results in a more organized and uniform film with fewer pores, compared with immersed or dipped films.
