Several studies have demonstrated that three‐dimensional (3‐D) culture systems influence human embryonic stem cell (hESC) phenotypes and fate choices. However, the effect that these microenvironmental changes have on signaling pathways governing hESC behaviors is not well understood. Here, a 3‐D microwell array was used to investigate differences in activation of developmental pathways between 2‐D and 3‐D cultures of both undifferentiated hESCs and hESCs undergoing initial differentiation in embryoid bodies (EBs). An increased induction into mesoderm and endoderm and differences in expression of genes from multiple signaling pathways that regulate development, including Wnt/β‐catenin, TGF‐β superfamily, Notch, and FGF during EB‐mediated differentiation were observed in 3‐D microwells as compared with the 2‐D substrates. In undifferentiated hESCs, differences in epithelial‐mesenchymal transition phenotypes and the TGFβ/BMP pathway between cultures in 3‐D and 2‐D were also observed. Theory and research examining length of acquaintance and consensus among personality judgments have predominantly examined each dimension of personality separately. In L. J. Cronbach's (1955) terminology, this trait-centered approach combines consensus on elevation, differential elevation, and differential accuracy in personality judgments. The current article extends D. A. Kenny's (1991, 1994) weighted average model (WAM)--a theoretical model of the factors that influence agreement among personality judgments--to separate out two of Cronbach's components of consensus: stereotype accuracy and differential accuracy. Consistent with the predictions based on the WAM, as length of acquaintance increased, self-other agreement and consensus differential accuracy increased, stereotype accuracy decreased, and trait-level or raw profile correlations generally remained unchanged. Discussion focuses on the conditions under which a relationship between length of acquaintance and consensus and self-other agreement among personality evaluations emerges and how impressions change over time. Dielectrophoretic manipulation of nanoscale materials is typically performed in nonionic, highly insulating solvents. However, biomolecular recognition processes, such as DNA hybridization and protein binding, typically operate in highly conducting, aqueous saline solutions. Here, we report investigations of the manipulation and real-time detection of individual nanowires bridging microelectrode gaps in saline solutions. Measurements of the electrode impedance versus frequency show a crossover in behavior at a critical frequency that is dependent on the ionic strength. We demonstrate that by operating above this critical frequency, it is possible to use dielectrophoresis to manipulate nanowires across electrode gaps in saline solutions. By using electrical ground planes and nulling schemes to reduce the background currents, we further demonstrate the ability to electrically detect bridging and unbridging events of individual nanowires in saline solutions. The ability to both manipulate and detect bridging events with electrical signals provides a pathway toward automated assembly of nanoscale devices that incorporate biomolecular recognition elements.  相似文献

The plastic yield behavior of strip annealed 70/30 brass sheet has been investigated using several experimental techniques. Proportional path, stress-strain relations were measured in two strain states using a recently devised plane-strain test and a standard sheet tensile test. Based on these data, 70/30 brass exhibits a dramatic departure from Hill's plasticity models. Particularly notable is the lower work-hardening rate in plane strain. A second series of tests was carried out by deforming first in plane-strain tension and subsequently in uniaxial tension. The relative orientation of the principal strain directions in the two strain paths strongly affected the transient yielding behavior, but the original work-hardening pattern and plastic anisotropy were approached after an additional effective strain of ∼0.04. These observations are consistent with a two yield-surface model;i.e., one an underlying, proportional path yield surface and one an instantaneous, transient yield surface.  相似文献

Multivitamin/multimineral complexes are the most common dietary supplements. Unlike minerals in foods that are incorporated in bioorganic structures, minerals in dietary supplements are typically in an inorganic form. These minerals can catalyze the generation of free radicals, thereby oxidizing antioxidants during digestion. Here we examine the ability of a matrix consisting of an amino acid and non-digestible oligosaccharide (AAOS) to blunt metal-catalyzed oxidations. Monitoring of ascorbate radical generated by copper shows that ascorbate is oxidized more slowly with the AAOS matrix than with copper sulfate. Measurement of the rate of oxidation of ascorbic acid and Trolox® by catalytic metals confirmed the ability of AAOS to slow these oxidations. Similar results were observed with iron-catalyzed formation of hydroxyl radicals. When compared to traditional forms of minerals used in supplements, we conclude that the oxidative loss of antioxidants in solution at physiological pH is much slower when AAOS is present.  相似文献

We propose a model of the shape, motion and appearance of a scene, seen through a sequence of images, that captures occlusions, scene deformations, unconstrained viewpoint variations and changes in its radiance. This model is based on a collection of overlapping layers that can move and deform, each supporting an intensity function that can change over time. We discuss the generality and limitations of this model in relation to existing ones such as traditional optical flow or motion segmentation, layers, deformable templates and deformotion. We then illustrate how this model can be used for inference of shape, motion, deformation and appearance of the scene from a collection of images. The layering structure allows for automatic inpainting of partially occluded regions. We illustrate the model on synthetic and real sequences where existing schemes fail, and show how suitable choices of constants in the model yield existing schemes, from optical flow to motion segmentation and inpainting.  相似文献

The optimization of electrospinning SU‐8 2100 negative photoresist was performed to create carbon micro/nanofibers and beads that can be patterned after electrospinning by using UV radiation. The fiber diameters had a range of 300 nm to 1 μm, based upon the selected electrospinning parameters. Low concentrations of the SU‐8 2100 resulted in beads while specific higher concentrations produced well‐defined fibers. Fibers and beads were converted to carbon through pyrolysis and retained their three dimensional structure. By utilizing the photosensitive properties of the SU‐8 negative photoresist, the electrospun fibers were patterned by UV photolithography. Serine hydrolases have diverse intracellular substrates, biological functions, and structural plasticity, and are thus important for biocatalyst design. Amongst serine hydrolases, the recently described ybfF enzyme family are promising novel biocatalysts with an unusual bifurcated substrate‐binding cleft and the ability to recognize commercially relevant substrates. We characterized in detail the substrate selectivity of a novel ybfF enzyme from Vibrio cholerae (Vc‐ybfF) by using a 21‐member library of fluorogenic ester substrates. We assigned the roles of the two substrate‐binding clefts in controlling the substrate selectivity and folded stability of Vc‐ybfF by comprehensive substitution analysis. The overall substrate preference of Vc‐ybfF was for short polar chains, but it retained significant activity with a range of cyclic and extended esters. This broad substrate specificity combined with the substitutional analysis demonstrates that the larger binding cleft controls the substrate specificity of Vc‐ybfF. Key selectivity residues (Tyr116, Arg120, Tyr209) are also located at the larger binding pocket and control the substrate specificity profile. In the structure of ybfF the narrower binding cleft contains water molecules prepositioned for hydrolysis, but based on substitution this cleft showed only minimal contribution to catalysis. Instead, the residues surrounding the narrow binding cleft and at the entrance to the binding pocket contributed significantly to the folded stability of Vc‐ybfF. The relative contributions of each cleft of the binding pocket to the catalytic activity and folded stability of Vc‐ybfF provide a valuable map for designing future biocatalysts based on the ybfF scaffold.  相似文献

The Ceramic On-Demand Extrusion (CODE) process is a novel additive manufacturing method for fabricating dense (~99% of theoretical density) ceramic components from aqueous, high solids loading pastes (>50?vol%). In this study, 3?mol% Y2O3 stabilized zirconia (3YSZ) specimens were fabricated using the CODE process. The specimens were then dried in a humidity-controlled environmental chamber and afterwards sintered under atmospheric conditions. Mechanical properties of the sintered specimens were examined using ASTM standard test techniques, including density, Young’s modulus, flexural strength, Weibull modulus, fracture toughness, and Vickers hardness. The microstructure was analyzed and grain size measured using scanning electron microscopy. The results were compared with those from Direct Inkjet Printing, Selective Laser Sintering, Lithography-based Ceramic Manufacturing (LCM), and other extrusion-based processes, and indicated that zirconia specimens produced by CODE exhibit superior mechanical properties among the additive manufacturing processes. Several sample components were produced to demonstrate CODE’s capability for fabricating geometrically complex ceramic components.
