Indoleamine 2,3‐dioxygenase 1 (IDO1) has emerged as a key target for cancer therapy, as IDO1 plays a critical role in the capacity of tumor cells to evade the immune system. The pyrrolopiperazinone alkaloid longamide B and its derivatives were identified as novel IDO1 inhibitors based on docking studies and small library synthesis. The thioamide derivative showed higher IDO1 inhibitory activity than longamide B, and displayed an activity similar to that of a representative IDO1 inhibitor, 1‐methyl‐tryptophan. These results suggest that the pyrrolopiperazinone scaffold of longamide B could be used in the development of IDO1 inhibitors.  相似文献

Bis(triethoxysilyl)-substituted oligothiophene with a moderate π-conjugation length was newly synthesized and polymerized to give a polysilsesquioxane network having oligothiophene units dispersed homogeneously without phase separation. The polymer was fixed on glass or ITO substrate by spin-coating and annealing. The resulting film exhibited a high mechanical strength due to the covalent bonding with the substrates, and was electrochemically stable even after 300 redox cycles in electrolyte solution. Chemical oxidation of the polymer films yielded electrically conductive and almost transparent films.  相似文献

After the recent discovery of bacterial riboswitches, synthetic riboswitches have been engineered by using natural and artificial RNA aptamers. In contrast to natural riboswitches, the majority of synthetic riboswitches in bacteria reported to date are ON switches that activate gene expression in response to the aptamer ligand. In this study, we adopted a mechanism‐guided approach to design libraries predisposed to contain OFF riboswitches that respond to thiamine pyrophosphate (TPP). The first library design exploited a pseudo‐Shine‐Dalgarno (SD) sequence located near the 3′‐end of the TPP aptamer, which would be less accessible to the ribosome when the aptamer is bound to TPP. In the second library, an SD sequence was strategically placed in the aptamer's P1 stem, which is stabilized upon ligand binding. OFF riboswitches were obtained by dual genetic selection of these libraries. The results underscore the importance of effective library design to achieve desired riboswitch functions.  相似文献

The distribution behavior of n-alcohols, ketones and nitroalkanes in aqueous liquid chromatography with a column packed with polyethylene glycol (PEG) gel, TSKgel Ether-250, was compared with that in aqueous two-phase systems (ATPSs) formed from PEG and Na2SO4 or (NH4)2SO4. The plots of the distribution data obtained for the PEG gel system against those for the ATPS reveal that the separation selectivities exerted by the PEG gel system and the PEG-based ATPS are approximately the same. Differential scanning calorimetry studies on aqueous PEG solutions suggest that PEG polymer forms a hydration structure of which the composition is 50% (w/w) PEG or the hydration number per ethylene oxide is 2.4 and the separation selectivity of the PEG-water systems can be attributed to partition of solute compounds into the hydrated PEG polymer structure.  相似文献

Surface‐initiated atom transfer radical polymerization (ATRP) from poly(ethylene terephthalate) (PET) film was studied. Poly(methyl methacrylate) (PMMA), poly (acrylamide) (PAAM), and their diblock copolymer (PMMA/PAAM) on the surface of PET film were successfully prepared by surface‐initiated ATRP. The structures and properties of the modified PET film were characterized by FT‐IR/ATR, X‐ray photoelectron spectroscopy (XPS), measurements of contact angles, and scanning electronic microscopy (SEM). It is necessary in factories to assess the severity of the surface defects of castings, as a slight surface defect will be taken as qualified when it brings no bad effect or it can be removed by the subsequent processing. In practical production, professional technicians visually inspect the surface defect severity according to their individual experience. Therefore, it is difficult for them to maintain the same standard and accuracy in the subjective, tedious and labor-intensive work. Recently, image processing techniques based on optical images have been applied to achieve better accuracy and high efficiency. Unfortunately, optical images cannot directly quantify surface depth, which works as a crucial factor in the practical assessment of surface defect severity. The surface roughness evaluation algorithm, which takes into account of both area and depth information of the assessed surface, was applied to directly characterize surface defect severity based on surface asperity rather than optical image. The results using standard casting pieces show that surface defect severity has no apparent dependence on surface roughness. However, the subsequent results show that the root-mean-squared-deviation (RMSD) of surface gradient of flow line defects positively correlates with the increase of defect severity. The other types of defect do not present such tendency. Thus, practical workpieces with flow line defects on the surface were used to verify the universality of this tendency. The results show that surface roughness of an unqualified workpiece is larger than that of a qualified workpiece after surface slope adjustment, but presents no obvious coincidence before the adjustment. In contrast, the RMSD of an unqualified workpiece, no matter before or after the adjustment, is larger than that of a qualified one.  相似文献

The photoreaction of metal-organic compounds prepared by the chemical modification of metal alkoxides was investigated for the patterning of ceramic films. The chemical stability and photoreactivity of these compounds were found to be greatly influenced by the kind of chemical additive applied during the syntheses of the precursor solutions. In this study, the photoreaction of the precursor thin films prepared by the addition of alkanolamines was achieved by tuning the wavelength of the incident UV light. The decomposition of the organic moieties of the percursors was clearly observed in IR spectra during UV irradiation. Furthermore, the spectrum of the titania precursor film in the visible and ultraviolet regions (UV-vis) changed in accordance with the irradiation time. The photolithography of the titanium precursor pattern was successfully achieved by means of these techniques.  相似文献

Two-dimensional images of the distribution showing the temperature coefficient of the dielectric constant for two types of two-phase composite ceramics composed of TiO2-Bi2Ti4O11 and BaTi4O9-BaPr2Ti4O12 were obtained using scanning photothermal dielectric microscopy. The images of the TiO2-Bi2Ti4O11 ceramic showed that the TiO2 and Bi2Ti4O11 grains had negative and positive temperature coefficients, respectively, and that the macroscopic averaged temperature coefficient of the ceramic was relatively low because of the cancellation of the opposite signs of the coefficients. On the other hand, the images of the BaTi4O9-BaPr2Ti4O12 ceramic showed that the temperature coefficient of both grains had the same sign (negative), although their absolute values were quite different.  相似文献

High-toughness and high-strength lead zirconate titanate (PZT) composites that contain fine silver particles were successfully fabricated at low sintering temperatures. Addition of silver to a PZT matrix did not result in unwanted reaction phases; however, some silver diffused toward the perovskite crystal structure. A small quantity of silver accelerated the sinterability of the PZT composites. The formation of oxygen vacancies due to the partial substitution of silver appeared to enhance the sinterability of the PZT. Fracture toughness depended on the size and degree of sphericity of the silver particles, and SEM observations on crack propagation suggested that the toughening mechanism in the PZT/Ag composites involves crack bridging resulting from the ductile behavior of silver particles. It is proposed that high fracture strength in PZT/1 to 5 vol% Ag composites results from the relaxation of transformation-induced internal stress by the silver particles.  相似文献

Fine-grained ZK60 magnesium alloy with the grain size of 1.4 μm was processed by equal-channel-angular extrusion. The material exhibited low temperature superplasticity. The normalized plot suggested that the present material had equilibrium grain boundaries at the superplastic temperature in contrast to typical materials processed by severe plastic deformation.  相似文献
