Development of an analytical method with appropriate combination of extraction and quantification approaches for undigested phytate in ruminant feces and digesta will advance knowledge of phytate degradation in ruminants and help to reduce phosphorus excretion. Established quantification methods give satisfactory results for feedstuffs and nonruminant manure but recovery of phytate is incomplete for ruminant feces and digesta because of their complex sample matrix and low ratio of phytate to inorganic P. The objective was to develop a robust, accurate, sensitive, and inexpensive method to extract and quantify phytate in feeds, ruminant feces, and digesta. Diets varying in phytate content were fed to dairy heifers, dry cows, and lactating cows to generate digesta and fecal samples of varying composition to challenge extraction and quantification methods. Samples were extracted with 0.5 M HCl or 0.25 M NaOH + 0.05 M EDTA. Acid extracts were mixed with 20% NaCl, alkaline extracts were acidified to final pH <2, and then both extracts were clarified with C18 cartridges and 0.2-μm filters. High-performance ion chromatography (HPIC) was used to quantify phytate. In feed samples, the measured phytate was comparable in alkaline and acid extracts (2,965 vs. 3,085 μg/g of DM). In digesta and fecal samples, alkaline extraction yielded greater estimates of phytate content than did acid extraction (40.7 vs. 33.6 and 202.9 vs. 144.4 μg/g of DM for digesta and fecal samples, respectively). Analysis of alkaline extracts by HPIC is usually not possible because of sample matrix interferences; acidification and C18-cartridge elution of alkaline extracts prevented this interference. Pure phytate added to dry samples before extraction was almost completely recovered (88 to 105%), indicating high extraction efficiency, no adverse effect of extract clean-up procedures, and accurate quantification of phytate. The proposed method is rapid, inexpensive, robust, and combines the extraction power of NaOH-EDTA with the precision and sensitivity of HPIC quantification, allowing accurate quantification of phytate in feeds, ruminant digesta, and fecal samples.  相似文献

This paper describes the design and laboratory testing of novel generation apparatus for supplying an isolated DC load from a self-excited induction generator operable at variable speed. The variable-speed generating apparatus consists of a self-excited induction machine, a controlled Graetz bridge rectifier, a voltage-boost power converter, and a control system. The induction generator supplies the rectifier. The voltage-boost power converter interfaces the variable output voltage of the rectifier to the fixed DC voltage required for the load. The rectifier is operated at levels of average DC current and voltage which control machine voltage to the rated AC voltage and which also draw the necessary power to supply the DC load. Performance is enhanced with respect to earlier apparatus in that both the DC voltage supplied to the load and the AC voltage on the machine are simultaneously controlled to fixed reference levels over broad operating ranges of load and speed  相似文献

The purpose of this study was to examine the effects of excessive fluid in the knee joint on proprioception. Twenty healthy subjects with no knee pathology participated in this study. Subjects were randomly assigned to control and experimental groups. Baseline measurements of subjects performing a tracking task were then recorded. Specifically, one knee joint was moved passively by a dynamometer through a range of motion from approximately 90 degrees flexion to 10 degrees from full extension at a joint angular velocity of 60 degrees/sec. Subjects were blindfolded and required to track the passively moving joint as accurately as possible for a period of 1.25 minutes with the opposite limb. An electrogoniometer was used to monitor the motion of the active tracking limb. Ninety mL of a solution of saline and dextrose were injected into the knee joint cavity of the experimental subjects, and they repeated the tracking task. After a 5-minute rest, the control group subjects also repeated the task. The results indicated that the injection of fluid did not change the subjects' error in tracking the passively moving limb (p > 0.05). No change in the error associated with tracking was observed for the control group (p > 0.05). It is suggested that the effects of long-term effusions and the nature of the inflammatory fluid might be more responsible for the loss of proprioception observed in some clinical conditions.  相似文献

CONTEXT: The incidence of infective endocarditis after surgical repair of congenital heart defects is unknown. OBJECTIVE: To determine the long-term incidence of endocarditis after repair of any of 12 congenital heart defects in childhood. DESIGN: Population-based registry started in 1982. SETTING: State of Oregon. PARTICIPANTS: All Oregon residents who underwent surgical repair for 1 of 12 major congenital defects at the age of 18 years or younger from 1958 to the present. MAIN OUTCOME MEASURE: Diagnosis of infective endocarditis confirmed by hospital or autopsy records. RESULTS: Follow-up data were obtained from 88% of this cohort of 3860 individuals through 1993. At 25 years after surgery, the cumulative incidence of infective endocarditis was 1.3% for tetralogy of Fallot, 2.7% for isolated ventricular septal defect, 3.5% for coarctation of the aorta, 13.3% for valvular aortic stenosis, and 2.8% for primum atrial septal defect. In the cohorts with shorter follow-up, at 20 years after surgery the cumulative incidence was 4.0% for dextrotransposition of the great arteries; at 10 years, the cumulative incidence was 1.1% for complete atrioventricular septal defect, 5.3% for pulmonary atresia with an intact ventricular septum, and 6.4% for pulmonary atresia with ventricular septal defect. No children with secundum atrial septal defect, patent ductus arteriosus, or pulmonic stenosis have had infective endocarditis after surgery. CONCLUSION: The continuing incidence of endocarditis after surgery for congenital heart defect, particularly valvular aortic stenosis, merits education about endocarditis prophylaxis for children and adults with repaired congenital heart defects.  相似文献

The development and evaluation of new drugs often rely on surrogacy. An intermediate outcome becomes a surrogate outcome if it fulfils certain criteria, it should be easier to measure compared with the clinical outcome, a statistical relationship should exist between the clinical outcome and the surrogate outcome, a relation should exist allowing prediction of the degree of clinical effect based on the measured effect on the surrogate outcome. Development and authorization of drugs today often rely on so-called surrogate outcomes. Is this use sound? The validity of such outcomes has been reviewed in different therapeutic areas: hypertension, venous thromboembolism, AIDS, osteoporosis, hepatitis C. Based on this review, a pragmatic strategy is proposed which allows for the validation and proper use of surrogate outcomes.  相似文献

Soils overfertilized with phosphorus (P) are widespread in the European Union and there is consensus among soil scientists to better explore their potential to release phosphate. In this work we report the principal physical and chemical properties of twelve overfertilized benchmark soils of contrasting agricultural areas in Italy, Germany, Great Britain, and Spain. The criterion used to consider them as overfertilized was that the available P amount, measured by the regional soil P test, was at least twice as large as the accepted critical level for an average crop. The soils could be usefully divided into three groups, calcareous, slightly acidic and acidic based upon their basic chemical properties and reactions and proportion of the major P fractions (NaOH- plus citrate-bicarbonate-, citrate-bicarbonate-dithionite-, and HCl-extractable P). Six extraction procedures commonly used to evaluate potentially plant-available P (Olsen, anion-exchange resin (resin-), anion plus cation-exchange resin (resin±), Ca acetatelactate (CAL), water, and Fe oxide-impregnated paper strips (strip) were compared. The extractable P values by each method were correlated but the amount of P extracted varied and differed in the order water-P< Olsen-P< CAL-P< strip-P< resin(±)-P相似文献

The interactions between three polynuclear aromatic hydrocarbons (PAHs), namely fluorene, naphthalene and anthracene, and three fractions of a soil, namely fulvic acid (FA), humic acid (HA) and humin/inorganic fractions, were studied experimentally. Prepared PAH + soil fraction binary mixtures, with PAH concentrations ranging over 0.8–7 mass%, were tested using a differential scanning calorimeter (DSC) over a temperature range of 20?390°C. The DSC thermograms for the PAH + HA mixtures showed distinctly different characteristics compared to mixtures with the other two soil fractions, where the endothermic peak for the vaporization of the PAH was absent. With the aid of vapour-liquid flash calculations, the lack of the boiling point peak is interpreted to be due to the liquid-phase miscibility of the PAHs and the HA fraction. Implications of the DSC results on the design and performance of the thermal desorption process are discussed.  相似文献

The state-by-state transient screening approach based on a pulse-response thin-zone TAP experiment is further developed whereby single-pulse kinetic tests are treated as small perturbations to catalyst compositions and analyzed using integral method of moments. Results on three primary kinetic characteristics, termed basic kinetic coefficients, are presented. These three coefficients were introduced as main observables from experimentally measured TAP-responses in a kinetic-model-free manner. Each was analytically determined from moments of responses with no assumption about the detailed kinetic model. In this paper, the inverse question of how well these coefficients represent the time evolution of the observed responses is addressed. Sets of three basic kinetic coefficients are calculated from model and experimental responses and these calculated values are used to generate 3-coefficient curves in a kinetic-model-free manner. The comparison of these 3-coefficient curves with original responses shows that three basic kinetic coefficients can be sufficient to describe the observed kinetics of exit flow time dependencies with no assumption regarding the detailed kinetic model.  相似文献
