The Glanville fritillary butterfly Melitaea cinxia feeds upon two host plant species in Å land, Finland, Plantago lanceolataand Veronica spicata, both of which produce iridoid glycosides. Iridoids are known to deter feeding or decrease the growth rate of many generalist insect herbivores, but they often act as oviposition cues to specialist butterflies and are feeding stimulants to their larvae. In this study, two iridoid glycosides (aucubin and catalpol) were analyzed by micellar electrokinetic capillary chromatography. We measured the spatial and temporal variation of iridoid glycosides in natural populations of the host plants of M. cinxia. We also analyzed the aucubin and catalpol content in plants in relation to their use by ovipositing females, and in relation to the incidence of parasitism of M. cinxia larvae in natural populations. The mean concentrations of aucubin and catalpol were higher in P. lanceolata than in V. spicata, and catalpol concentrations were higher than aucubin concentrations in both host species. Plantago lanceolata individuals that were used for oviposition by M. cinxia had higher aucubin concentrations than random plants and neighboring plants. Additionally, oviposition and random plants had higher catalpol concentrations than neighboring plants, indicating that ovipositing females select for high iridoid glycoside plants or that oviposition induces iridoid glycoside production in P. lanceolata. Parasitism by the specialist parasitoid wasp Cotesia melitaearum occurred most frequently in larval groups that were feeding on plants with low concentrations of catalpol, irrespective of year, population, and host plant species. Lignin‐based chemicals, starch, and urea were used as modifiers for phenol–formaldehyde resol resins. The effects of the addition stage of the modifiers used in the synthesis of the resins and the type of modification reagent on the structures of the resins and their molar masses and reactivities were investigated. The modifications with corn starch and lignin promoted condensation; this was verified by increased molar masses and high ratios of methylene bridges to the sum of free ortho and para aromatic groups with respect to the corresponding reference resin without a modification reagent. The later the modifier was added to the resin condensation mixture, the more methylene bridges were formed with respect to the amounts of free ortho and para aromatic groups. In addition, when urea or wheat starch was added in the later condensation stage, the final condensation also reached high stages. The modifications with lignosulfonate and starch, as well as the early addition of urea, enhanced p–p′ bridge structures. The lowest condensation stage and, therefore, the highest reactivity were found when wheat starch was added with the starting reagents. Enzymes are effective biological catalysts that accelerate almost all metabolic reactions in living organisms. Synthetic modulators of enzymes are useful tools for the study of enzymatic reactions and can provide starting points for the design of new drugs. Here, we report on the discovery of a class of biologically active compounds that covalently modifies lysine residues in human liver pyruvate kinase (PKL), leading to allosteric activation of the enzyme (EC50=0.29 μM). Surprisingly, the allosteric activation control point resides on the lysine residue K282 present in the catalytic site of PKL. These findings were confirmed by structural data, MS/MS experiments, and molecular modelling studies. Altogether, our study provides a molecular basis for the activation mechanism and establishes a framework for further development of human liver pyruvate kinase covalent activators. 相似文献

The ability to identify inhibitors of protein–protein interactions represents a major challenge in modern drug discovery and in the development of tools for chemical biology. In recent years, fragment‐based approaches have emerged as a new methodology in drug discovery; however, few examples of small molecules that are active against chemotherapeutic targets have been published. Herein, we describe the fragment‐based approach of targeting the interaction between the tumour suppressor BRCA2 and the recombination enzyme RAD51; it makes use of a screening pipeline of biophysical techniques that we expect to be more generally applicable to similar targets. Disruption of this interaction in vivo is hypothesised to give rise to cellular hypersensitivity to radiation and genotoxic drugs. We have used protein engineering to create a monomeric form of RAD51 by humanising a thermostable archaeal orthologue, RadA, and used this protein for fragment screening. The initial fragment hits were thoroughly validated biophysically by isothermal titration calorimetry (ITC) and NMR techniques and observed by X‐ray crystallography to bind in a shallow surface pocket that is occupied in the native complex by the side chain of a phenylalanine from the conserved FxxA interaction motif found in BRCA2. This represents the first report of fragments or any small molecule binding at this protein–protein interaction site. 相似文献

The first part of this article reports the analytical form of the Electrical Low Pressure Impactor (ELPI) kernel functions. In this latter part, the numerical quality of ELPI response matrices is studied and an example of an inversion algorithm is given. The ELPI assemblies with and without an electrical filter stage and with smooth or sintered impaction plates are studied and compared with basic impactor kernels and the kernels for the calculation of the aerosol mass distribution. It is shown that the ELPI assembly with the electrical filter stage and smooth impaction plates should be the best choice for the inversion of data if no bounce occurs. The comparison to a mass impactor shows that the devices are on par in data inversion. The inversion ELPI data is studied with a Bayesian algorithm assuming a bimodal lognormal size distribution of the aerosol. The algorithm includes a novel procedure for obtaining an initial guess of the distribution parameters. To our knowledge, it is also the first algorithm to use ELPI current readings as its input. Simulations and diesel emission measurements show that the proposed algorithm is a useful tool in the study of ELPI data. 相似文献

Nanometric ceria powders with fluorite-type structure were obtained by applying self-propagating room temperature method. The obtained powders were subsequently thermally treated (calcined) at different temperatures for different times. Powder properties such as specific surface area, crystallite size, particle size and lattice parameter have been studied. Roentgen diffraction analysis (XRD), BET and Raman scattering measurements were used to characterize the as-obtained (uncalcined) powder as well as powders calcined at different temperatures.It was found that the average diameter of the as-obtained crystallites is in the range of 3–5 nm whereas the specific surface area is about 70 m2/g. The subsequent, 15 min long, calcination of as-obtained powder at different temperatures gradually increased crystallite size up to ~60 nm and reduced specific surface down to 6 m2/g. Raman spectra of synthesized CeO2?y depicts a strong red shift of active triply degenerate F2 g mode as well as additional peak at 600 cm?1. The frequency of F2 g mode increased while its line width decreased with an increase in calcination temperature. Such a behavior is considered to be the result of particle size increase and agglomeration during the calcination. After the heat treatment at 800 °C crystallite size reached value larger than 50 nm. Second order Raman mode, which originates from intrinsic oxygen vacancies, disappeared after calcination. 相似文献

Brain is one of the richest organs in lipid content. Phospholipids (glycerophospholipids and sphingolipids) are important building blocks of cell membranes, which provide an optimal environment for protein interactions, trafficking and function. Because of that, alterations in their cellular levels could lead to different pathogenic processes in the brain, such as in Alzheimer’s disease (AD), the most common type of dementia among older populations. There is increasing evidence that phospholipid changes occur during pathogenic processes in AD. It is known that lipids are tightly connected with metabolism of the Amyloid Precursor Protein (APP), which produces Amyloid-beta peptide (Aβ), the main component of senile plaques, which represent the main pathological hallmark of AD. However, the mechanism(s) of the lipid-effect on Aβ metabolism and AD pathogenesis is still not completely understood. This review summarizes the current knowledge on phospholipid changes occurring during normal aging and discusses phospholipid changes in the human brain associated with different stages of AD, as well changes in the cerebrospinal fluid and blood/plasma, which are interesting potential biomarkers for AD diagnosis and disease monitoring. At the end, we have discussed future perspectives of phospholipid changes as potential biomarkers and as targets for development of novel treatment strategies against AD. 相似文献

Manganese is a commonly found substance in groundwater in Finland. As a powerful oxidant, ozone can be used for the oxidizing of manganese even without raising the pH. The SFS (Finnish Standards Association) standard has set the accepted limit for soluble manganese to 0.45 μm. However, some research papers have used the limit of 30 kD (kilodalton) for soluble manganese. This research concentrates on the size fractions of manganese in four samples of untreated groundwater and in four samples of ozonized groundwater when treated with 0.45 μm, 0.20 μm, 100 kD, 30 kD, and 10 kD filters. In all tests, nearly all manganese contained in raw water penetrated all filters. There were slight variations in the flocculation of manganese in ozonized groundwater; nevertheless, hardly any reduction in manganese levels took place beyond 100 kD. After ozonation, there were two water samples which surpassed the manganese limit of 50 μgl?1 set for domestic water when the filtration was 0.20 μm and another two samples when the filtration was 100 kD. 相似文献

Vitamin A is an essential fat-soluble vitamin that occurs in various chemical forms. It is essential for several physiological processes. Either hyper- or hypovitaminosis can be harmful. One of the most important vitamin A functions is its involvement in visual phototransduction, where it serves as the crucial part of photopigment, the first molecule in the process of transforming photons of light into electrical signals. In this process, large quantities of vitamin A in the form of 11-cis-retinal are being isomerized to all-trans-retinal and then quickly recycled back to 11-cis-retinal. Complex machinery of transporters and enzymes is involved in this process (i.e., the visual cycle). Any fault in the machinery may not only reduce the efficiency of visual detection but also cause the accumulation of toxic chemicals in the retina. This review provides a comprehensive overview of diseases that are directly or indirectly connected with vitamin A pathways in the retina. It includes the pathophysiological background and clinical presentation of each disease and summarizes the already existing therapeutic and prospective interventions. 相似文献

The paper deals with the tray-to-tray method for determining the required number of trays in columns with intensive entrainment of liquid when the operating line of the “dry working regime” is not straight and with a small concentration of diffusing component in gas. Presented calculations show that the number of transfer units for gas and liquid should be calculated using the AIChE method accompanied with longitudinal mixing according to Gilbert properly transformed for case of diffusion through stagnant film. The suggested method can be applied for TEG dehydration absorbers-columns for dehydration of natural gas using triethylene glycol with system factor SF = 0.5. 相似文献
