Application of artificial nucleases (ANs) in genome editing is still hindered by their cytotoxicity related to off‐target cleavages. This problem can be targeted by regulation of the nuclease domain. Here, we provide an experimental survey of computationally designed integrated zinc finger nucleases, constructed by linking the inactivated catalytic centre and the allosteric activator sequence of the colicin E7 nuclease domain to the two opposite termini of a zinc finger array. DNA specificity and metal binding were confirmed by electrophoretic mobility shift assays, synchrotron radiation circular dichroism spectroscopy, and nano‐electrospray ionisation mass spectrometry. In situ intramolecular activation of the nuclease domain was observed, resulting in specific cleavage of DNA with moderate activity. This study represents a new approach to AN design through integrated nucleases consisting of three (regulator, DNA‐binding, and nuclease) units, rather than simple chimera. The optimisation of such ANs could lead to safe gene editing enzymes. 相似文献

A versatile solid‐phase approach based on peptide chemistry was used to construct four classes of structurally diverse polyamines with modified backbones: linear, partially constrained, branched, and cyclic. Their effects on DNA duplex stability and structure were examined. The polyamines showed distinct activities, thus highlighting the importance of polyamine backbone structure. Interestingly, the rank order of polyamine ability for DNA compaction was different to that for their effects on circular dichroism and melting temperature, thus indicating that these polyamines have distinct effects on secondary and higher‐order structures of DNA. 相似文献

Methanosarcina species pyrrolysyl‐tRNA synthetase (PylRS) attaches Pyl to its cognate amber suppressor tRNA. The introduction of two mutations (Y384F and Y306A) into PylRS was previously shown to generate a mutant, designated LysZ‐RS, that was able to attach N‐benzyloxycarbonyl‐L ‐lysine (LysZ) to its cognate tRNA. Despite the potential of LysZ derivatives, further LysZ‐RS engineering has not been performed; consequently, we aimed to generate LysZ‐RS mutants with improved LysZ incorporation activity through in vitro directed evolution. Using a liposome‐based in vitro compartmentalization (IVC) approach, we screened a randomly mutagenized gene library of LysZ‐RS and obtained a mutant that showed increased LysZ incorporation activity both in vitro and in vivo. The ease and high flexibility of liposome‐based IVC should enable the evolution of not only LysZ‐RS that can attach various LysZ derivatives but also of other enzymes involved in protein translation. 相似文献

Thermal decomposition of titanium(IV) tetra-tert-butoxide (TTB) in inert organic solvents at 573 K yielded microcrystalline
anatase (titanium(IV) oxide, TiO2) powders with a crystallite size of ca. 9 nm and a surface area of <100 m2 g-1. Primary and secondary alkoxides of titanium(IV), however, were not decomposed under similar conditions, indicating that
the thermal stability of C-O bonds in the alkoxides was a decisive factor for their decomposition. The TiO2 prepared from TTB by this manner was thermally stable upon calcination in air and retained high surface area of ca. 100 m2 g-1 even after calcination at 823 K. The as-prepared TiO2 powders, without calcination, exhibited much higher rate of carbon dioxide formation than any other active photocatalysts
such as Degussa P-25 and Ishihara ST-01 in the photocatalytic mineralization of acetic acid in aerated aqueous solutions.
The higher activity of the present TiO2 photocatalysts is attributed to both high crystallinity and large surface of the present product. The calcination of the
as-prepared TiO2 in air reduced the photocatalytic activity, but it was still higher than the other commercially available TiO2's.

Relationship between degree of polymerization and optical and thermal properties of fluorene in polycarbonate polymers 下载免费PDF全文
The influences of average degree of polymerization (Dp) and terminal group on thermal and optical properties of high refractive indexed transparent polymers were investigated. In this study, 9,9‐bis[4–(2‐hydroxyethoxy) phenyl] fluorene (BPEF) homo polymer was selected because it has been used as a representative monomer in high refractive index polymers as well as its unique property. BPEF has stable amorphous phase and reacts like a polymer. Its unique reaction allows continuous investigation from monomer to polymer. For hydroxyl‐terminated polymer, the refractive index (nd) decreased with increasing Dp. On the other hand, for a phenolic‐terminated group, nd increased with increasing Dp, and both converged to same value in high Dp region. As for glass transition temperatures (Tg), both terminal group series were increased as Dp increased. Though Tg of hydroxyl‐terminated polymer was higher than that of phenolic‐terminated polymer in the low Dp region, both converged to the same value and the inverse number of Tg had linear correlation against the weight percentage of carbonyl groups (CO), which was calculated by Dp. © 2017 Wiley Periodicals, Inc. Indoor air pollution is caused predominantly by emission of primary pollutants from building materials or finishing materials. Recently, there has been increasing interest in the new secondary pollutants caused by chemical reaction of the primary pollutants. In this study, the pollutants emitted from poly(vinyl chloride) (PVC) flooring material were measured, and 2-ethyl-1-hexanol (2E1H) and volatile organic compounds (VOCs) emitted from composite building materials made of PVC flooring material used as a self-leveling flooring were measured to identify the mechanism of emission of 2E1H. The moisture sorption isotherm was measured with a Magnetic Suspension Balance to examine the moisture content of the self-leveling flooring material and its effect on emission of 2E1H. 2E1H was emitted from the PVC flooring material through hydrolysis or oxidation of di-2-ethylhexyl phthalate (DEHP). The composite building material including the self-leveling flooring material emitted more 2E1H through hydrolysis over time. It was determined that liquid prevails in the self-leveling flooring material when the water content is higher than 4% and vapor prevails when the water content is lower than 4%. The prevailing liquid in the flooring material contributes to the increased emission of 2E1H and the prevailing vapor in the material did not cause increase in emission of 2E1H. It is considered that control of water in the self-leveling flooring material or concrete slab is very important in order to inhibit emission of 2E1H through hydrolysis of DEHP on the floor. 相似文献

Polycarbonates were synthesized by transesterification polymerization of isosorbide, bisphenol A, and diphenyl carbonate in the presence of n‐BuSn(?O)OH as the catalyst. The polymers were prepared by varying the ratio of the two monomers. The polymers exhibited good thermal stability, rigidity, and strength. The storage moduli for the polymers were 2.6 to 3.2 GPa at 25°C and increased with increasing isosorbide content. The dynamic mechanical analysis indicated that the storage moduli (E′) of the polymers are constant to higher temperatures than that of bisphenol A polycarbonate. © 2012 Wiley Periodicals, Inc. J. Silicon nanowire (SiNW) arrays were prepared on silicon substrates by metal-assisted chemical etching and peeled from the substrates, and their optical properties were measured. The absorption coefficient of the SiNW arrays was higher than that for the bulk silicon over the entire region. The absorption coefficient of a SiNW array composed of 10-μm-long nanowires was much higher than the theoretical absorptance of a 10-μm-thick flat Si wafer, suggesting that SiNW arrays exhibit strong optical confinement. To reveal the reason for this strong optical confinement demonstrated by SiNW arrays, angular distribution functions of their transmittance were experimentally determined. The results suggest that Mie-related scattering plays a significant role in the strong optical confinement of SiNW arrays. 相似文献

Metabolic syndrome is associated with an increased risk of colorectal cancer. This study investigated the impact of hypertension, a component of metabolic syndrome, on azoxymethane (AOM)-induced colorectal carcinogenesis using SHRSP/Izm (SHRSP) non-diabetic/hypertensive rats and SHRSP.Z-Leprfa/IzmDmcr (SHRSP-ZF) diabetic/hypertensive rats. Male 6-week-old SHRSP, SHRSP-ZF, and control non-diabetic/normotensive Wister Kyoto/Izm (WKY) rats were given 2 weekly intraperitoneal injections of AOM (20 mg/kg body weight). Two weeks after the last injection of AOM, the SHRSP and SHRSP-ZF rats became hypertensive compared to the control WKY rats. Serum levels of angiotensin-II, the active product of the renin-angiotensin system, were elevated in both SHRSP and SHRSP-ZF rats, but only the SHRSP-ZF rats developed insulin resistance, dyslipidemia, and hyperleptinemia and exhibited an increase in adipose tissue. The development of AOM-induced colonic preneoplastic lesions and aberrant crypts foci, was significantly accelerated in both SHRSP and SHRSP-ZF hypertensive rats, compared to WKY normotensive rats. Furthermore, induction of oxidative stress and exacerbation of inflammation were observed in the colonic mucosa and systemically in SHRSP and SHRSP-ZF rats. Our findings suggest that hypertension plays a role in the early stage of colorectal carcinogenesis by inducing oxidative stress and chronic inflammation, which might be associated with activation of the renin-angiotensin system. 相似文献

To achieve a high-efficiency silicon nanowire (SiNW) solar cell, surface passivation technique is very important because a SiNW array has a large surface area. We successfully prepared by atomic layer deposition (ALD) high-quality aluminum oxide (Al2O3) film for passivation on the whole surface of the SiNW arrays. The minority carrier lifetime of the Al2O3-depositedSiNW arrays with bulk silicon substrate was improved to 27 μs at the optimum annealing condition. To remove the effect of bulk silicon, the effective diffusion length of minority carriers in the SiNW array was estimated by simple equations and a device simulator. As a result, it was revealed that the effective diffusion length in the SiNW arrays increased from 3.25 to 13.5 μm by depositing Al2O3 and post-annealing at 400°C. This improvement of the diffusion length is very important for application to solar cells, and Al2O3 deposited by ALD is a promising passivation material for a structure with high aspect ratio such as SiNW arrays. 相似文献
