Liposomes are used as effective nanodelivery devices to improve the physicochemical stability and biological efficacy of the encapsulated peptides and proteins. In this study, nanoliposome composite of lipoid S75-entrapped angiotensin I-converting enzyme (ACE)-inhibitory biopeptides was prepared by conventional (BLS75-CM) and direct heating (BLS75-DHM) methods. The nanoliposomes (BLS75-CM and BLS75-DHM) were stored at 4°C for 8 weeks and evaluated for physicochemical stability in terms of particle size, polydispersity index (pdi), zeta potential, and encapsulation efficiency (EE). These were also studied for residual ACE-inhibitory efficacy following their digestion under simulated gastrointestinal tract condition. The BLS75-CM was found to maintain higher physicochemical stability in terms of particle size, pdi, and zeta potential compared to BLS75-DHM. However, the BLS75-DHM indicated higher EE and efficacy with greater residual ACE-inhibitory activity of 47.37% compared to 44.18% and 36.84% that were obtained for the digested BLS75-CM and digested biopeptides without encapsulation, respectively. In vitro release study showed a cumulative biopeptides release of 66.41% and 69.00% from BLS75-CM and BLS75-DHM, respectively. The results of transmission electron microscopy showed spherical appearance of the nanoliposome capsules while Fourier Transform Infrared spectroscopy indicated the presence of ionic complexation and hydrogen bonds between the biopeptides and their phospholipid matrix.  相似文献

The efficiency of various techniques pulsed electric field (PEF), ultrasound (US), high‐pressure microfluidisation (HPMF), hydrochloric acid (HCl) and ionic liquids (ILs) for cell wall disruption in astaxanthin extraction from Haematococcus pluvialis was compared. The results indicated that ILs, HCl and HPMF treatment were shown the most efficient for cell disruption with more than 80% astaxanthin recovery. While the cell wall integrity of H. pluvialis cyst cell was less affected by US and PEF treatment. It was found that imidazolium‐based ILs showed the greater potential for cell disruption than pyridinium‐based and ammonium‐based ILs. Among all the ILs examined, 1‐butyl‐3‐methylimidazolium chloride ([Bmim] Cl) exhibited efficient cell disruption and capability of astaxanthin recovery at mild condition (pretreatment with 40% IL aqueous solution at 40 °C, followed by extraction with methanol at 50 °C) without extensive energy consumption and special facility requirement. This study evaluates the synergistic antifungal effects between thyme essential oils and Lactobacillus plantarum cell‐free supernatant (LCFS) against Penicillium spp. and in situ antifungal activity in rice grains. Thyme essential oil and LCFS showed remarkable antifungal activities against Penicillium spp. with the minimum inhibitory concentration (MIC) of 40 and 80 µL/mL, respectively. The analysis of fractional inhibitory concentration (FIC) index showed the antifungal synergism between thyme essential oil and LCFS against Penicillium spp. with FIC index of 0.5. This synergism also resulted in fourfold reduction in their MICs when applied in combination. The antifungal modes of action were characterized by observing the changes in cell membrane permeability and degradation of fungal cell wall. The combination of thyme essential oil and LCFS (2 × MIC of each) showed remarkable in situ antifungal effect and completely inhibit the growth of Penicillium in rice seeds. The results suggested the possible applications of the observed synergism on actual crops.

Practical applications

Essential oils are used as preservative in food industry and high concentration of essential oil is associated with negative organoleptic characteristics. This study presented a novel approach for synergistic antifungal effects by using the combination of thyme essential oil and Lactobacillus plantarum cell‐free supernatant (LCFS) against Penicillium spp. and systematic evaluation of the antifungal effect by using fractional inhibitory concentration (FIC) index method. This approach will be a role model for future research on synergism and overcome the major drawbacks of using live bacteria and the negative effects arising from antimicrobial activities of essential oils.  相似文献

A presence of mycotoxins in feed is one of the most alarming issues in the poultry feed industry. Ochratoxins, produced by several Aspergillus and Penicillium species, are important mycotoxin regarding the health status of poultry birds. Ochratoxins are further classified into to several subtypes (A, B, C, etc) depending on their chemical structures, but ochratoxin A (OTA) is considered the most important and toxic. Bentonite clay, belonging to phyllosilicates and formed from weathering of volcanic ashes, has adsorbent ability for several mycotoxins. The present study was designed to study the effects of bentonite clay upon OTA-induced immunosuppression in broiler chicks. For this, 480 day-old broiler chicks were procured from a local hatchery and then different combinations of OTA (0.15, 0.3, or 1.0 mg/kg) and bentonite clay (5, 10, and 20 g/kg) were incorporated into their feed. At 13, 30, and 42 days of age, parameters such as antibody responses to sheep red blood cells, in situ lymphoproliferative responses to mitogen (PHA-P), and in situ phagocytic activity (i.e., via carbon clearance) were determined respectively. The results indicated there was a significant reduction of total antibody and immunoglobulin titres, lymphoproliferative responses, and phagocytic potential in OTA-treated birds, suggesting clear immunosuppression by OTA in birds in a dose-dependent manner. These results were also significantly lower in all combination groups (OTA with bentonite clay), suggesting few to no effects of feeding bentonite clay upon OTA- induced alterations in different immune parameters.  相似文献
