The DNA band patterns generated by polymerase chain reaction (PCR) using the du2 primer and template DNAs from various strains of Escherichia coli and non-E. coli bacteria were compared. Among three to five prominent bands produced, the three bands at about 1.8, 2.7, and 5.0 kb were detected in all of the E. coli O157 strains tested. Some nonpathogenic E. coli and all pathogenic E. coli except E. coli O157 showed bands at 1.8 and 5.0 kb. It seems that the band at 2.7 kb is specific to E. coli O157. Sequence analysis of the 2.7-kb PCR product revealed the presence of a DNA sequence specific to E. coli O157:H- and E. coli O157:H7. Since the DNA sequence from base 15 to base 1,008 of the PCR product seems to be specific to E. coli O157, a PCR assay was carried out with various bacterial genomic DNAs and O157-FHC1 and O157-FHC2 primers that amplified the region between base 23 and base 994 of the 2.7-kb PCR product. A single band at 970 bp was clearly detected in all of the strains of E. coli O157:H- and E. coli O157:H7 tested. However, no band was amplified from template DNAs from other bacteria, including both nonpathogenic and pathogenic E. coli except E. coli O157. All raw meats inoculated with E. coli O157:H7 at 3 x 10(0) to 3.5 x 10(2) CFU/25 g were positive both for our PCR assay after cultivation in mEC-N broth at 42 degrees C for 18 h and for the conventional cultural method.  相似文献

Previous serolgoical studies strongly suggested Akabane virus to be the etiologic agent of epizootic abortion and congenital arthrogryposis-hydranencephaly in cattle, and this view was further corroborated in this study by the isolation of the virus from an aborted fetus in an epizootic of the disease and from a fetus extracted froma cow which was suggested by serologic tests to have a recent infection with the virus. The latter fetus had histological changes of encephalomyelitis and polymyositis, and specific antigens of Akabane virus was shown by the immunofluorescent technique in brain tissues as well as skeletal muscular tissues. The virus was recovered from various fetal tissues and fluids, and in relatively large amounts from brain, spinal cord, cerebral fluid, skeletal muscles and fetal placenta. The intracranial inoculation of suckling mice, 1-2 days of age, was the most sensitive system for Akabane virus isolation and HmLu-1, a continuous cell line from hamster lung, seemed almost as sensitive as suckling mice.  相似文献

We have developed a simple technique to grow ribbon-like single crystalline films of Bi- 2212 on Ag substrates. The process is performed under ambient pressure only by heating a Bi-2212 pellet on the edge of the substrates. 1 The ribbon-like films thus grown showed sharp superconducting transitions with an onset temperature of 80K. The formation of the ribbons was studied by observing their growth with a high- temperature microscope. In situ observation with the microscope revealed that most of the ribbon crystals grew during the 10 minutes of the plateau at 905° C. In particular, the early part of the plateau was the dominant stage of growth.  相似文献

We tested for four kinds of allergic substances (egg, milk, wheat and peanuts) in 52 imported processed foods using immunochromatographic test kits (ITK). ELISA was also employed to confirm the effectiveness of the ITK. Among 92 data from 23 samples, allergic substances were detected in 9 cases with one kind of ITK, but not with the ELISA test. Among 116 data from 29 samples, 6 were negative with one kind of ITK and but positive with the other ITK. These results suggested that these 4 kinds of allergic substances in imported foods can be detected by using a double-check method with two kinds of ITK.  相似文献

Anomalous heat effect by interaction of hydrogen isotope gas and metal nanocomposites supported by zirconia or by silica has been examined. Observed absorption and heat evolution at RT were not too large to be explained by some chemical processes. At elevated temperatures of 200–300 °C, most samples with binary metal nanocomposites produced excess power of 3–24 W lasting for up to several weeks. The excess power was observed not only in the D-Pd·Ni system but also in the HPd·Ni system and HCu·Ni system, while single-element nanoparticle samples produced no excess power. The Pd/Ni ratio is one of the keys to increase the excess power. The maximum phase-averaged excess heat energy exceeded 270 keV/D, and the integrated excess heat energy reached 100 MJ/mol-M or 90 MJ/mol-H. It is impossible to attribute the excess heat energy to any chemical reaction; it is possibly due to radiation-free nuclear process.  相似文献

Tetraphenylporphyrin (TPP) is a symmetrically substituted synthetic porphyrin whose properties can be readily modified, providing it with significant advantages over naturally occurring porphyrins. Herein, we report the first example of a stable complex between a native biomolecule, the haemoprotein HasA, and TPP as well as its derivatives. The X-ray crystal structures of nine different HasA-TPP complexes were solved at high resolutions. HasA capturing TPP derivatives was also demonstrated to inhibit growth of the opportunistic pathogen Pseudomonas aeruginosa. Mutant variants of HasA binding FeTPP were shown to possess a different mode of coordination, permitting the cyclopropanation of styrene.  相似文献
