The bone marrow (BM) microenvironment plays a crucial role in the development and progression of leukemia (AML). Intracellular reactive oxygen species (ROS) are involved in the regulation of the biology of leukemia-initiating cells, where the antioxidant enzyme GPx-3 could be involved as a determinant of cellular self-renewal. Little is known however about the role of the microenvironment in the control of the oxidative metabolism of AML cells. In the present study, a coculture model of BM mesenchymal stromal cells (MSCs) and AML cells (KG1a cell-line and primary BM blasts) was used to explore this metabolic pathway. MSC-contact, rather than culture with MSC-conditioned medium, decreases ROS levels and inhibits the Nrf-2 pathway through overexpression of GPx3 in AML cells. The decrease of ROS levels also inactivates p38MAPK and reduces the proliferation of AML cells. Conversely, contact with AML cells modifies MSCs in that they display an increased oxidative stress and Nrf-2 activation, together with a concomitant lowered expression of GPx-3. Altogether, these experiments suggest that a reciprocal control of oxidative metabolism is initiated by direct cell–cell contact between MSCs and AML cells. GPx-3 expression appears to play a crucial role in this cross-talk and could be involved in the regulation of leukemogenesis.  相似文献

In previous work, an atmospheric pressure plasma jet was applied to successfully improve the electrospinnability of poly-ε-caprolactone (PCL) which enabled the fabrication of beadless nanofibers. In this paper, we report the aging effect of the plasma treatment to evaluate the robustness of the developed process. For this purpose, plasma-treated PCL polymer solutions with different exposure time were stored for 1 and 8 days and the aged solutions were analyzed in terms of conductivity, viscosity, surface tension, pH, and polymer molecular weight. During storage, the surface tension and acidity of the plasma-treated solutions were maintained constant. However, the viscosity was found to be significantly lower after 8 days which was attributed to PCL degradation. Electrospinning of all stored PCL solutions resulted in the generation of beadless PCL nanofibers. The plasma treatment effects were thus found to be highly stable over time and capable of producing high-quality PCL nanofibers up to 8 days. © 2020 Wiley Periodicals, Inc. J. Appl. Polym. In this article, an extension of the spatial filter method to study Fabry‐Perot antennas with homogeneous or inhomogeneous partially reflecting surface (PRS) of finite size is proposed. This tool was subsequently validated through the study of different Fabry‐Perot antennas, with homogeneous PRS and inhomogeneous GRadient‐INdex (GRIN) PRS that present very high side lobe level (SLL). Since they are due to structure truncation, a new inhomogeneous PRS to reduce the SLL is designed with the new analytical tool. The new inhomogeneous PRS for Fabry‐Perot antenna is characterized by simulations and measurements. Compared to the homogeneous PRS antenna, the proposed PRS allows a SLL reduction of 5 dB without decreasing the 14 dBi directivity.  相似文献

Anomaly detection in streaming data is an important problem in numerous application domains. Most existing model‐based approaches to stream learning are based on decision trees due to their fast construction speed. This paper introduces streaming autoencoder (SA), a fast and novel anomaly detection algorithm based on ensembles of neural networks for evolving data streams. It is a one‐class learner, which only requires data from the positive class for training and is accurate even when anomalous training data are rare. It features an ensemble of threaded autoencoders with continuous learning capacity. Furthermore, the SA uses a 2‐step detection mechanism to ensure that real anomalies are detected with low false‐positive rates. The method is highly efficient because it processes data streams in parallel with multithreads and alternating buffers. Our analysis shows that SA has a linear runtime and requires constant memory space. Empirical comparisons to the state‐of‐the‐art methods on multiple benchmark data sets demonstrate that the proposed method detects anomalies efficiently with fewer false alarms.  相似文献

Printed image multiplexing based on the design of metasurfaces has attracted much interest in the past decade. Optical switching between different images displayed directly on the metasurface is performed by altering the parameters of the incident light such as polarization, wavelength, or incidence angle. When using white light, only two-image multiplexing is implemented with polarization switching. Such metasurfaces are made of nanostructures perfectly controlled individually, which provide high-resolution pixels but small images and involve long fabrication processes. Here, it is demonstrated that laser processing of nanocomposites offers a versatile low-cost, high-speed method with large area processing capabilities for controlling the statistical properties of random metasurfaces, allowing up to three-image multiplexing under white light illumination. By independently controlling absorption and interference effects, colors in reflection and transmission can be varied independently yielding two-image multiplexing under white light. Using anisotropy of plasmonic nanoparticles, a third image can be multiplexed and revealed through polarization changes. The design strategy, the fundamental properties, and the versatility of implementation of these laser-empowered random metasurfaces are discussed. The technique, applied on flexible substrate, can find applications in information encryption or functional switchable optical devices, and offers many advantages for visual security and anticounterfeiting.  相似文献

In order to remove xylene vapors from an air stream, an upflow laboratory scale biofilter was operated for a period of 2 months. The experimental study consisted of two different phases: in the first phase, the biofilter was operated at various gas flow rates and the xylene inlet concentration was maintained at 1.39 g m−3. In the second phase, various inlet concentrations of the contaminant were tested at a constant gas flow rate of 0.4 m3 h−1 corresponding to an empty bed residence time of 150 s. The biofilter response to steep and abrupt variations in the xylene inlet concentration and gas flow rate was examined. The results obtained revealed that the removal efficiency of the biofilter regained its high values (above 96%) in less than 24 h following the change to low concentrations and gas flow rate. Temperature measurements showed that the biofilter temperature strongly depends on the intensity of the microbial activity in the filter bed. The experimental mass ratio of carbon dioxide produced to the xylene removed was equal to 2.72 indicating that the contaminant was eliminated exclusively by aerobic biodegradation. These findings suggest that a follow up of the amount of carbon dioxide produced in the filter bed can be very helpful in monitoring the performance of the biofilter. For relatively small inlet loads of xylene, the contributions of the different sections of the biofilter to the removal efficiency of the contaminant and the carbon dioxide production were unevenly balanced but became more uniformly distributed for relatively high inlet loads.  相似文献
