Presynaptic N-type calcium channels interact with syntaxin and synaptosome-associated protein of 25 kDa (SNAP-25) through a binding site in the intracellular loop connecting domains II and III of the alpha1 subunit. This binding region was loaded into embryonic spinal neurons of Xenopus by early blastomere injection. After culturing, synaptic transmission of peptide-loaded and control cells was compared by measuring postsynaptic responses under different external Ca2+ concentrations. The relative transmitter release of injected neurons was reduced by approximately 25% at physiological Ca2+ concentration, whereas injection of the corresponding region of the L-type Ca2+ channel had virtually no effect. When applied to a theoretical model, these results imply that 70% of the formerly linked vesicles have been uncoupled after action of the peptide. Our data suggest that severing the physical interaction between presynaptic calcium channels and synaptic proteins will not prevent synaptic transmission at this synapse but will make it less efficient by shifting its Ca2+ dependence to higher values. 相似文献

The acoustic emission (AE) behavior during fatigue propagation in aluminum 6061 and aluminum 6061 matrix composites containing
5, 10, and 20 wt pct SiC particle reinforcement was investigated under tension-tension fatigue loading. The purpose of this
investigation was to monitor fatigue crack propagation by the AE technique and to identify the source(s) of AE. Most of the
AEs detected were observed at the top of the load cycles. The cumulative number of AE events was found to correspond closely
to the fatigue crack growth and to increase with increasing SiC content. Fractographic studies revealed an increasing number
of fractured particles and to a lesser extent decohered particles on the fatigue fracture surface as the crack propagation
rate(e.g., †K) or the SiC content was increased.

Hourly images registered by the Meteosat satellite are used to derive the global irradiance at ground level. Keeping almost the full spatial information of the original images, maps of irradiance characteristics for Africa are derived with a resolution of approx. 10 × 10 km2. Monthly irradiance conditions are characterized by means and standard deviations of daily sums and the irradiance of specific daytime hours. The data also allow for the extraction of irradiance time series for specific sites. In addition, the analysis of the spatial structure of the satellite-derived irradiance fields is used to identify regions in which the radiation characteristics call for special attention. 相似文献

This study focused on analyzing active pathways and the metabolic flux distribution in human neuronal AGE1.HN cells that is a desirable basis for a rational design and optimization of producing cell lines and production processes for biopharmaceuticals. 13C-labeling experiments and 13C metabolic flux analysis were conducted using glucose, glutamine, alanine and lactate tracers in parallel experiments. Connections between cytosolic and mitochondrial metabolite pools were verified, e.g., flux from TCA cycle metabolite 13C to glycolytic metabolites. It was also found that lactate and alanine are produced from the same pyruvate pool and that consumed alanine is mainly directly metabolized and secreted as lactate. Activity of the pentose phosphate pathway was low being around 2.3% of the glucose uptake flux. This might be compensated in AGE1.HN by high mitochondrial malic enzyme flux producing NADPH. Mitochondrial pyruvate transport was almost zero. Instead pyruvate carbons were channeled via oxaloacetate into the TCA cycle which was mainly fed via α-ketoglutarate and oxaloacetate during the investigated phase. The data indicate that further optimization of this cell line should focus on the improved substrate usage which can be accomplished by an improved connectivity between glycolytic and mitochondrial pyruvate pools or by better control of the substrate uptake. 相似文献

This article outlines a Bayesian bootstrap method for case based imprecision estimates in Bayes classification. We argue that this approach is an important complement to methods such as k-fold cross validation that are based on overall error rates. It is shown how case based imprecision estimates may be used to improve Bayes classifiers under asymmetrical loss functions. In addition, other approaches to making use of case based imprecision estimates are discussed and illustrated on two real world data sets. Contrary to the common assumption, Bayesian bootstrap simulations indicate that the uncertainty associated with the output of a Bayes classifier is often far from normally distributed. 相似文献

A prototype inspection station based on the United States Department of Agriculture (USDA) inspection standards was developed for potato grading. The station consisted of an imaging chamber, conveyor, camera, sorting unit, and personal computer for image acquisition, analysis, and equipment control. A sample of 9.1kg (201b) of pregraded potatoes were evaluated in three separate experimental runs to assess the system performance. The system correctly classified 80%, 77%, and 88% of the moving potatoes in the three runs at 3 potatoes/min, and 98%, 97%, and 97%, in three runs of stationary potatoes. Among the molecular, cellular, and systemic events that have been proposed to modulate the function of the hippocampus and the entorhinal cortex (EC), one of the most frequently cited possibilities is the activation of the serotonergic system. Neurons in the hippocampus and in the EC receive a strong serotonergic projection from the raphe nuclei and express serotonin (5-HT) receptors at high density. Here we review the various effects of 5-HT on intrinsic and synaptic properties of neurons in the hippocampus and the EC. Although similar membrane-potential changes following 5-HT application have been reported for neurons of the entorhinal cortex and the hippocampus, the effects of serotonin on synaptic transmission are contrary in both areas. Serotonin mainly depresses fast and slow inhibition of the principal output cells of the hippocampus, whereas it selectively suppresses the excitation in the entorhinal cortex. On the basis of these data, we discuss the possible role of serotonin under physiological and pathophysiological circumstances. 相似文献

This study focuses on the fortuitous discovery of an atypical atherosclerotic lesion in four of 49 male adult cynomolgus monkeys (macacus fascicularis) which were maintained for a long time at a high level of hypercholesterolemia, and in seven of 19 female cynomolgus monkeys examined from the second to the 24th week of hypercholesterolemic diet: this lesion was in formation or already mature during this period of diet. This atypical lesion was formed by a collagen and elastic network surrounding synthetic smooth muscle cells without fibrofatty or fibrous plaques. Lipids were occasionally seen in the inner intima. The lesion appeared early (from the third week of diet). Once established, its morphology did not change. It became more extensive, but was not complicated by lipid overload in spite of prolonged, permanent hypercholesterolemia. This response to hypercholesterolemia is interesting because the activity of the smooth muscle cells differs from that observed in the classic lesion: they intervene earlier, their replication is very marked and rapid, their elastin secretion is greater and remains constant over time, and their phagocytic properties are reduced.
