Expanded bed behavior was modeled by using the Richardson-Zaki correlation between the superficial velocity of the feed stream and the void fraction of the bed. A polydisperse material, Chelating excellose® (70-210 Μm in diameter, 1.21 g/cm3 in density), which has Ni2+ ions for the selective binding of histidine-tagged proteins, was used as the resin. A method to modify the Stokes expression to express the terminal settling velocity of the resins by introducing two empirical parameters, the effective diameter of the resins and an exponent for p -ρ)/Μ term, was developed. Combined use of the Richardson-Zaki correlation and the modified Stokes expression was successful in modeling the bed expansion by incorporating physical properties of feed streams and the resins.  相似文献

In an effort to improve the properties of cyclodextrin glucanotransferase (CGTase) as an antistaling enzyme, error-prone PCR was used to introduce random mutations into a CGTase cloned from alkalophilic Bacillus sp. I-5 (CGTase I-5). A mutant CGTase[3-18] with the three mutations M234T, F259I and V591A was selected by agar plate assay. Sequence alignment of various CGTases indicated that M234 and F259 are located in the vicinity of the catalytic sites of the enzyme and V591 in the starch binding domain E. The cyclization activity of CGTase[3-18] was dramatically decreased by 10-fold, while the hydrolyzing activity was increased by up to 15-fold. These mutations near subsite +1 (M234T) and at subsite +2 (F259I) are likely to alter the enzyme activity in a concerted manner, promoting hydrolysis of substrate while retarding cyclization. The addition of CGTase[3-18] reduced the retrogradation rate of bread by as much as did the commercial antistaling enzyme Novamyl during 7-day storage at 4 degrees C. No cyclodextrin (CD) was detected in bread treated with CGTase[3-18], whereas 21 mg of CD per 10 g of bread was produced in bread treated with wild-type CGTase.  相似文献

Diblock copolymers with different poly(ε‐caprolactone) (PCL) block lengths were synthesized by ring‐opening polymerization of ε‐caprolactone in the presence of monomethoxy poly(ethylene glycol) (mPEG‐OH, MW 2000) as initiator. The self‐aggregation behaviors and microscopic characteristics of the diblock copolymer self‐aggregates, prepared by the diafiltration method, were investigated by using 1H NMR, dynamic light scattering (DLS), and fluorescence spectroscopy. The PEG–PCL block copolymers formed the self‐aggregate in an aqueous environment by intra‐ and/or intermolecular association between hydrophobic PCL chains. The critical aggregation concentrations of the block copolymer self‐aggregate became lower with increasing hydrophobic PCL block length. On the other hand, reverse trends of mean hydrodynamic diameters were measured by DLS owing to the increasing bulkiness of the hydrophobic chains and hydrophobic interaction between the PCL microdomains. The partition equilibrium constants (Kv) of pyrene, measured by fluorescence spectroscopy, revealed that the inner core hydrophobicity of the nanoparticles increased with increasing PCL chain length. The aggregation number of PCL chain per one hydrophobic microdomain, investigated by the fluorescence quenching method using cetylpyridinium chloride as a quencher, revealed that 4–20 block copolymer chains were needed to form a hydrophobic microdomain, depending on PCL block length. © 2006 Wiley Periodicals, Inc. With the rapid development of visual digital media, the demand for better quality of service has increased the pressure on broadcasters to automate their error detection and restoration activities for preserving their archives. Digital dropout is one of the defects that affect archived visual materials and tends to occur in block by block basis (size of 8 × 8). It is well established that human visual system (HVS) is highly adapted to the statistics of its visual natural environment. Consequently, in this paper, we have formulated digital dropout detection as a classification problem which predicts block label based on statistical features. These statistical features are indicative of perceptual quality relevant to human visual perception, and allow pristine images to be distinguished from distorted ones. Here, the idea is to extract discriminant block statistical features based on discrete cosine transform (DCT) coefficients and determine an optimal neighborhood sampling strategy to enhance the discrimination ability of block representation. Since this spatial frame based approach is free from any motion computation dependency, it works perfectly in the presence of fast moving objects. Experiments are performed on video archives to evaluate the efficacy of the proposed technique.  相似文献

Abstract— A flexible fluorescent lamp that utilizes the same plasma discharge mode as in PDPs has been manufactured. The structure of the flexible lamp is simple and easy to manufacture. All‐plastic materials including plastic substrates, barrier ribs (spacers), and sealants for low‐temperature manufacturing processing have been adopted except for the phosphor and MgO thin film. The MgO thin films were coated on the plastic substrates as a protection layer against the plasma discharge. The adhesion and biaxial texture of MgO thin film deposited on the plastic substrates, poly‐ethyle‐nenaphthalate (PEN) and polycarbonate (PC), at low temperature (100–180°C) has been characterized. The MgO film on PEN shows good adhesion under a repeated bending test. The manufactured flexible lamp consists of two plastic substrates of about 3 in. on the diagonal, barrier rib (spacer), and external ITO electrodes. The Ne‐Xe (5%) gas mixture at 100–200 Torr was used for the discharge gas. A maximum surface luminance of about 100 cd/m2 was achieved for a 1 ‐kHz AC pulse.  相似文献

This study explodes the relationships between the height adjustment of bed sectors and subjective comfort depending on three types of sleeping positions. An adjustable bed system that regulates the heights of 8 adjustment sectors of the bed was developed. To evaluate the comfort of 9 body areas, subjective ratings were requested from 64 subjects ranging in age from 25 to 50. The subjects included 29 males and 35 females. For an objective evaluation, the patterns of pressure distribution were investigated according to the three sleeping positions. During two separate states (before bed adjustment and after bed adjustment), measurements of the adjusted bed height and pressure distribution, and subjective ratings according to three sleeping positions were performed. Based on the results of the adjusted heights of the 8 sectors in the bed, the subjects preferred the W‐shaped sleeping posture in both the supine and side positions. However, the U‐shaped sleeping posture was preferred in the prone position. With respect to these sleeping positions, the heights of sectors significantly correlated with the subjective ratings presented by the subjects, as well as the ratios of pressure. © 2009 Wiley Periodicals, Inc.  相似文献

The maintenance of relevant backgrounds under various scene changes is very crucial to detect foregrounds robustly. We propose a background maintenance method for dynamic scenes including global intensity level changes caused by changes of illumination conditions and camera settings. If the global level of the intensity changes abruptly, the conventional background models cannot discriminate true foreground pixels from the background. The proposed method adaptively modifies the background model by estimating the level changes. Because there are changes caused by moving objects as well as global intensity level changes, we estimate the dominant level change over the whole image regions by mean shift. Then, the problem caused by saturated pixels are handled by an additional scheme. In the experiments for dynamic scenes, our proposed method outperforms previous methods by adaptive background maintenance and handling of saturated pixels.  相似文献

Diamond films 60 and 170 µm in thickness were grown by PACVD (plasma-assisted chemical vapor deposition) under similar conditions. The thermal diffusivity of these freestanding films was measured between 100 and 300 K using AC calorimetry. Radiation heat loss from the surface was estimated by analyzing both the amplitude and the phase shift of a lock-in amplifier signal. Thermal conductivity was calculated using the specific heat data of natural diamond. At room temperature, the thermal conductivity of the 60 and 170 m films is 9 and 16 W-cm–1. K–1 respectively, which is 40–70% that of natural diamond, The temperature dependence of thermal conductivity of the CVD diamond films is similar to that of natural diamond, Phonon scattering processes are considered using the Debye model, The microsize of the grain boundary has a significant effect on the mean free path of phonons at low temperatures. The grain in CVD diamond film is grown as a columnar structure, Thus, the thicker film has the larger mean grain size and the higher thermal conductivity. Scanning electron microscopy (SEM) and Raman spectroscopy were used to study the microstructure of the CVD diamond films. This article reviews state-of-the-art microfluidic biosensors of nucleic acids and proteins for point-of-care (POC) diagnostics. Microfluidics is capable of analyzing small sample volumes (10−9–10−18 l) and minimizing costly reagent consumption as well as automating sample preparation and reducing processing time. The merger of microfluidics and advanced biosensor technologies offers new promises for POC diagnostics, including high-throughput analysis, portability and disposability. However, this merger also imposes technological challenges on biosensors, such as high sensitivity and selectivity requirements with sample volumes orders of magnitude smaller than those of conventional practices, false response errors due to non-specific adsorption, and integrability with other necessary modules. There have been many prior review articles on microfluidic-based biosensors, and this review focuses on the recent progress in last 5 years. Herein, we review general technologies of DNA and protein biosensors. Then, recent advances on the coupling of the biosensors to microfluidics are highlighted. Finally, we discuss the key challenges and potential solutions for transforming microfluidic biosensors into POC diagnostic applications.  相似文献

In recent years, Radio Frequency Identification (RFID) industries have taken a great interest in utilizing the benefits of RFID for supply chain management, inventory control and various other applications. This paper proposed an adaptive load balancing technique for RFID middleware systems to meet the demands of scalability and heterogeneity. First, we explored five basic load balancing policies, namely, information policy, job selection policy, transfer policy, initiation policy and location policy. Eighteen load balancing schemes were then proposed for RFID middleware systems that were combinations of various types of the five basic load balancing policies. Our empirical study suggested that these load balancing strategies performed differently under different workload statuses. Finally, an adaptive load balancing strategy was proposed. The load balancing schemes and the proposed adaptive load balancing strategy have been implemented in the RFID Middleware Load Management System (RM‐LMS).
