Soft textured Atlantic salmon is a sporadic and occasionally very severe problem for the farming and processing industries. The firm and soft fillets examined in this work differed in their gelatinase activities, cross-reactivity with anti-ubiquitin and anti-cathepsin L antibodies, as well as in the in-gel α-chymotryptic peptide maps of electrophoretically isolated myosin heavy chain (MHC) bands. The immunodetections of actin, α-actinin, MHC, and the MALDI TOF MS peptide mass fingerprinting of electrophoretically isolated MHCs only showed minor differences between samples. Other analyses revealed merely individual differences. These results seem to indicate a higher level of gelatinase activation, ubiquitination and cathepsin L cross-reacting material in softer muscle. These results would be consistent with a myopathy, but also with what could be expected in the skeletal muscle of healthy salmonid fish during a normal period of hyperplastic growth.  相似文献

In this work, an innovative method for gas conditioning in biomass gasification is analyzed. The objective is to remove tar by selectively reforming the unwanted hydrocarbons in the product gas with a chemical looping reformer (CLR), while minimizing the carbon formation during the process. Toluene, in a concentration of 600-2000 ppmv, was chosen as a tar model compound. Experiments were performed in a TGA apparatus and a fixed bed reactor. Four oxygen carriers (60% NiO/MgAl2O4 (Ni60), 40% NiO/NiAl2O4 (Ni40), 40% Mn3O4/Mg-ZrO2 (Mn40) and FeTiO3 (Fe)) were tested under alternating reducing/oxidizing cycles. Several variables affecting the reducing cycle were analyzed: temperature, time for the reduction step and H2O/C7H8 molar ratio. Ni40 and Mn40 presented interesting characteristics for CLR of biomass tar. Both showed stable reactivity to C7H8 after a few cycles. Ni40 showed a high tendency to carbon deposition compared to Mn40, specially at high temperatures. Carbon deposition could be controlled by decreasing the temperature and the time for the reduction step. The addition of water also reduced the amount of carbon deposited, which was completely avoided working with a H2O/C7H8 molar ratio of 26.4.  相似文献

In this paper we examine cross-subsidisation among combined heat and power producers in Denmark. Information on stand-alone costs for heat generation allows us to empirically compare the Faulhaber tests, tests with an upper bound on stand-alone costs (the Palmer tests) and the fully distributed cost test (FDC). All tests indicate a substantial amount of cross-subsidisation from heat generation to power generation. It is shown that the FDC test is closer to that of the Faulhaber tests in its results than the Palmer tests. Thus as the Faulhaber tests are considered in the literature to be the theoretically correct tests, the FDC test is shown to be the best approximation for tests of cross-subsidisation for this specific sector.  相似文献

Functionalized synthetic oligonucleotides are finding growing applications in research, clinical studies, and therapy. However, it is not easy to prepare them in a biocompatible and highly efficient manner. We report a new strategy to synthesize oligonucleotides with promising nucleic acid targeting and detection properties. We focus in particular on the pH sensitivity of these new probes and their high target specificity. For the first time, human copper(I)‐binding chaperon Cox17 was applied to effectively catalyze click labeling of oligonucleotides. This was performed under ultramild conditions with fluorophore, peptide, and carbohydrate azide derivatives. In thermal denaturation studies, the modified probes showed specific binding to complementary DNA and RNA targets. Finally, we demonstrated the pH sensitivity of the new rhodamine‐based fluorescent probes in vitro and rationalize our results by electronic structure calculations.  相似文献

De novo design and chemical synthesis of proteins and of other artificial structures that mimic them is a central strategy for understanding protein folding and for accessing proteins with new functions. We have previously described carbohydrates that act as templates for the assembly of artificial proteins, so‐called carboproteins. The hypothesis is that the template preorganizes the secondary structure elements and directs the formation of a tertiary structure, thus achieving structural economy in the combination of peptide, linker, and template. We speculate that the structural information from the template could facilitate protein folding. Here we report the design and synthesis of three‐helix‐bundle carboproteins on deoxyhexopyranosides. The carboproteins were analyzed by CD, analytical ultracentrifugation (AUC), small‐angle X‐ray scattering (SAXS), and NMR spectroscopy, and this revealed the formation of the first compact and folded monomeric carboprotein, distinctly different from a molten globule. En route to this carboprotein we observed a clear effect originating from the template on protein folding.  相似文献
