The aim of the present study was to assess selective atrophy of the temporal lobe and amygdala in the early stages of Alzheimer dementia (AD). Magnetic resonance imaging (MRI) measurements and the presence of highsignal lesions (HSL) were analysed in 31 patients with mild to moderate probable AD and 22 controls. In the AD group, MRI findings were compared with cognitive variables and specific features of memory functions. Alzheimer patients showed a significant reduction in volumetric measurement compared with controls in the total volume (P < 0.01), temporal lobe (P < 0.01) and amygdala (P < 0.05). The temporal lobe/brain volume ratio was also significantly reduced in AD subjects (P < 0.05). Atrophy of temporal structures was significantly related to the degree of episodic and semantic memory impairment according to a material-specific effect. No significant correlations between amygdala and cognitive variables were found. The results of our study confirm the usefulness of measures of temporal lobe atrophy assessed with MRI in the diagnosis of AD. In contrast, HSL are relatively common in AD patients (12/31 cases) and were not related to volumetric findings, severity of dementia or functional disability. 相似文献

BACKGROUND: To date, to our knowledge, there is no systematic presentation of treatment outcome in large series of patients clinically diagnosed as having corticobasal degeneration. OBJECTIVE: To evaluate the clinical presentation and treatment outcome of patients clinically diagnosed as having corticobasal degeneration. SUBJECTS: We gathered case patients seen in 8 major movement disorder clinics during the last 5 years who were diagnosed as having corticobasal ganglionic degeneration. METHODS: Using a chart review method, we recorded the clinical presentation, medications used, response to medications, and adverse effects. RESULTS: A total of 147 case patients were reviewed, 7 were autopsy proven. Parkinsonian features were present in all, other movement disorders in 89%, and higher cortical dysfunction in 93%. The most common parkinsonian sign was rigidity (92%), followed by bradykinesia (80%), gait disorder (80%), and tremor (55%). Other movement disorders were dystonia in 71% and myoclonus in 55%. Higher cortical dysfunction included dyspraxia (82%), alien limb (42%), cortical sensory loss (33%), and dementia (25%). Ninety-two percent of the case patients received dopaminergic drugs, which resulted in a beneficial effect for 24%. Parkinsonian signs were the elements improving the most and levodopa was the most effective drug. Benzodiazepines, primarily clonazepam, were administered to 47 case patients, which resulted in improvement of myoclonus in 23% and dystonia in 9%. The most frequent disabling adverse effects of drug trials in these case patients were somnolence (n = 24), gastrointestinal complaints (n = 23), confusion (n = 16), dizziness (n =12), hallucinations (n = 5), and dry mouth (n = 5). CONCLUSIONS: Pharmacological intervention was largely ineffective in the management of corticobasal degeneration, and new treatments are needed for ameliorating the symptoms of this syndrome. 相似文献

BACKGROUND: Osteochondroma most frequently arises sporadically and as a solitary lesion, but also may arise as multiple lesions characterizing the autosomal dominant disorder hereditary multiple exostoses (HME) and the contiguous gene syndromes Langer-Giedion and DEFECT-11 syndromes. HME is genetically heterogeneous with association of three loci including 8q24.1 (EXT1), 11p11-12 (EXT2), and 19p (EXT3). Constitutional chromosomal microdeletions of 8q24.1 and 11p11-12 are features of the Langer-Giedion and DEFECT-11 syndromes, respectively. Cytogenetic studies of osteochondroma are rare. METHODS: Cytogenetic analysis was performed on 34 osteochondroma specimens from 22 patients with sporadic lesions and 4 patients with HME utilizing standard methodologies. Fluorescence in situ hybridization with chromosome specific probes was performed on three cases to define structural rearrangements further. RESULTS: Clonal abnormalities were detected in ten cases. Notably, deletion of 11p11-13 was observed in one case (a sporadic tumor) and loss or rearrangement of 8q22-24.1 in eight cases (seven sporadic and one hereditary tumor). CONCLUSIONS: These findings: 1) confirm previous observations of 8q24.1 karyotypic anomalies in sporadic osteochondroma, 2) reveal the presence of somatic chromosomal anomalies in hereditary osteochondromata, 3) suggest that similar to hereditary lesions, sporadic osteochondromas also are genetically heterogeneic (involvement of both 8q24.1 and 11p11-12), and 4) support the hypothesis that loss or mutation of EXT1 and EXT2, two putative tumor suppressor genes, may be important in the pathogenesis of sporadic as well as hereditary osteochondromata. 相似文献

Polypyrrole (PPy) is polymerized by chemical oxidative polymerization in presence of anionic surfactant sodium bis (2-ethylhexyl) sulfosuccinate
(DEHS) as the dopant. The electrical conductivity was optimized in terms of oxidant to monomer molar ratio and polymerization yield
was measured for these reactions. We have used ammonium persulphate (APS) as the oxidant for polymerization in this series of experiments. The effect of concentration of oxidant on the electrical
conductivity is examined. Chemical synthesis of polypyrrole is supported by FTIR spectrum. The electrical conductivity of
doped and undoped polypyrrole has been measured in the temperature range of 10–300 K and is found to increase with rise in
temperature. Electrical conductivity of PPy was analyzed in the light of various charge transport models. Analysis of the electrical conductivity data reveals that in
the temperature range 60–300 K electrical transport is predominantly governed by power law behaviour given by Kivelson model.

The aim of this study was to develop a pH-sensitive chitosan/polyvinyl pyrrolidone (PVP) based controlled drug release system for clarithromycin. The hydrogels were synthesized by cross-linking chitosan and PVP blend with glutaraldehyde to form a semi-interpenetrating polymer network (semi-IPN). These semi-IPNs were studied for their content uniformity, swelling index (SI), mucoadhesion, wettability, in vitro release and their release kinetics. The hydrogels showed more than 97% content of clarithromycin. These hydrogels showed high swelling and mucoadhesion under acidic conditions. The swelling may be due to the protonation of a primary amino group on chitosan. In acidic condition, chitosan would be ionized, and adhesion could have occurred between the positively charged chitosan and the negatively charged mucus. In the alkaline condition, less swelling and mucoadhesion was noticed. In vitro release study revealed that formulation containing chitosan (2% w/v) and PVP (4% w/v) in the ratio of 21:4 showed complete drug release after 12 h. Release profile showed that all the formulations followed non-Fickian diffusion mechanism. The cross-linking and compatibility of clarithromycin in the formulation was studied by Fourier transform infrared (FTIR) spectroscopic analysis, differential scanning calorimetry (DSC) and powder X-ray diffraction (p-XRD) study, which confirmed proper formation of semi-IPN and stability of clarithromycin in the formulations. The surface morphology of semi-IPN was studied before and after dissolution in simulated gastric fluid (SGF, pH 1.2) which revealed pores formation in membrane after dissolution. The results of study suggest that semi-IPNs of chitosan/PVP are potent candidates for delivery of clarithromycin in acidic environment. 相似文献

We conducted an in vivo study with the mouse model of Vibrio vulnificus infection to evaluate the efficacies of therapy with minocycline or cefotaxime alone and in combination. V. vulnificus was introduced subcutaneously into the area over the right thigh. The inoculum size ranged from 1.0 x 10(3) to 1.2 x 10(8) CFU from experiment to experiment but was constant for all animals in the same experiment. Antibiotics were given intraperitoneally 2 h after the bacteria were inoculated. In experiments 1 to 4, the standard dose for humans was used to treat the infection, while in experiment 5, five times the standard dose for humans was used to treat the infection. In experiment 1, with a small inoculum of 5 x 10(3) CFU, all mice in the saline-treated control group and the cefotaxime-, minocycline-, and combined antibiotic-treated groups survived. In experiment 2, with a moderate inoculum of 1.2 x 10(5) CFU, all the mice in the three antibiotic-treated groups survived, while only two of nine mice in the control group survived. In experiment 3, with a large inoculum of 8.0 x 10(7) CFU, six of nine mice in the combined antibiotic-treated group survived, while only one of nine mice in the cefotaxime-treated group and none of the mice in the control and minocycline-treated groups survived. In experiment 4, with a large inoculum of 1.2 x 10(8) CFU, 8 of 20 mice in the combined antibiotic-treated group survived, while none of the 20 mice in the control group, the group treated with cefotaxime alone, and the group treated with minocycline alone survived. In experiment 5, in which mice were infected with a large inoculum of 6.6 x 10(7) CFU and treated with five times the standard human dose of antibiotics, 10 of 12 mice in the combined antibiotic-treated group survived, while only 4 of 12 mice in the minocycline-treated group, 1 of 12 mice in the cefotaxime-treated group, and none of the mice in the control group survived. In experiments 3 to 5, the difference in the survival rates between the combined antibiotic-treated and minocycline-treated groups was statistically significant (P < 0.05). These results indicate that combination therapy with cefotaxime and minocycline is distinctly more advantageous than therapy with the single antibiotic regimen for the treatment of severe experimental V. vulnificus infections. 相似文献

BACKGROUND & AIMS: Recent reports suggest that platelet-activating factor (PAF) plays a role in pancreatitis and pancreatitis-associated lung injury. In this study, the effects on these processes of termination of PAF action by recombinant PAF-acetylhydrolase (rPAF-AH) were investigated. METHODS: Rats were given rPAF-AH and then infused with a supramaximally stimulating dose of cerulein to induce mild pancreatitis. Opossums underwent biliopancreatic duct ligation to induce severe pancreatitis, and rPAF-AH administration was begun 2 days later. RESULTS: In mild, secretagogue-induced pancreatitis, rPAF-AH given before the cerulein reduced hyperamylasemia, acinar cell vacuolization, and pancreatic inflammation but did not alter pancreatic edema or pulmonary microvascular permeability. In severe, biliopancreatic duct ligation-induced pancreatitis, rPAF-AH delayed and reduced the extent of inflammation and acinar cell injury/necrosis and completely prevented lung injury even though the rPAF-AH administration was begun after the onset of pancreatitis. CONCLUSIONS: PAF plays an important role in the regulation of pancreatic injury but not pancreatic edema or increased pulmonary microvascular permeability in mild, secretagogue-induced pancreatitis. PAF plays a critical role in the regulation of progression of pancreatic injury and mediation of pancreatitis-associated lung injury in severe biliary pancreatitis. Amelioration of pancreatitis and prevention of pancreatitis-associated lung injury can be achieved with rPAF-AH even if treatment is begun after pancreatitis is established. 相似文献

Gas hydrates are inclusion compounds in which gas is trapped in ice. They are widely found in nature in permafrost and deep-sea sediments and hold huge potential as a methane gas resource. As a result, a National Methane Hydrate R&D program was launched in 2000 by the United States Department of Energy. One of the projects under the program, run by the National Energy Technology Laboratory (NETL/DOE DE-FC26-01NT41331), is to drill, core, and test one gas hydrate well in the Prudhoe Bay/Kuparuk River area of northern Alaska. In this project, cores are studied to gather field data on a hydrate reservoir in permafrost regions. Tests were required to develop coring techniques and handling procedures on frozen rocks prior to actual coring in Alaska. These tests were performed at a Houston research facility where frozen rocks were prepared using an apparatus fabricated to simulate hydrate reservoir in permafrost. The permafrost formation was simulated with a water-saturated sand pack, which was frozen using liquid nitrogen. Frozen rocks of up to 3.6 m (12 ft) high and 0.254 m (10 in.) diameter were prepared with the apparatus. The setup was designed and evaluated in several stages. First, a computer model was developed to understand the rock-freezing process. Then a prototype setup was constructed to have similar radial dimensions as the full-scale apparatus, but with a smaller height. Several experiments were performed to test the prototype and the full-scale apparatus. The apparatus has been successfully employed to prepare frozen rock, thereby assisting in development of improved coring methodology for Alaskan permafrost hydrates exploration. 相似文献

Nanovoid growth was investigated using molecular dynamics to reveal its dependence on void size, strain rate, crystallographic loading orientation, initial nanovoid volume fraction, and simulation cell size. A spherical nanovoid was embedded into a periodic face-centered cubic (fcc) Al lattice, and a remote uniaxial load was applied to elucidate dislocation nucleation and shear loop formation from the void surface as well as the subsequent void growth mechanisms. The nucleation stresses and void growth mechanisms were compared for four different strain rates (107 to 1010 seconds?1), five different simulation cell sizes (4-nm to 28-nm lengths), four different initial nanovoid volume fractions, and seven different tensile loading orientations representative of the variability within the stereographic triangle. The simulation results show an effect of the size scale, crystallographic loading orientation, initial void volume fraction, and strain rate on the incipient yield stress for simulations without a void (single-crystal bulk material). For instance, the crystallographic orientation dependence on yield stress was less pronounced for simulations containing a void. As expected, dislocations and shear loops nucleated on various slip systems for the different loading orientations, which included orientations favored for both single slip and multiple slip. The evolution of the nanovoid volume fraction with increasing strain is relatively insensitive to loading orientations, which suggests that the nanoscale plastic anisotropy caused by the initial lattice orientation has only a minor role in influencing the nanovoid growth rate. In contrast, a significant influence of the initial nanovoid volume fractions was observed on the yield stress, i.e., a ~35 pct decrease in yield stress was caused by introducing a 0.4 pct nanovoid volume fraction. Furthermore, a continuum-scale bridging parameter m—which is a material rate sensitivity parameter in continuum damage mechanics—was calculated and found to be close to 1. Consequently, atomistic simulations of this type can indeed inform continuum void growth models for application in multiscale models. 相似文献

In this paper, the feasibility and efficacy of chromium (Cr(VI)) removal using three different kinds of tree leaves viz. Emblica officinalis, Azadirachta indica, Eucalyptus agglomerata, and the activated carbon is examined through batch and continuous flow experiments. Pretreatments were given to the selected tree leaf powders to remove the natural pigments and lignin present. Batch and continuous flow experiments have been conducted to study the kinetics of adsorption, effects of pH, adsorbent dose, contact time, bed depth, flow rate, and initial Cr(VI) concentration on Cr(VI) adsorption onto the selected adsorbents. The adsorption capacity is observed higher for Emblica officinalis followed by Eucalyptus agglomerata and Azadirachta indica. The adsorption equilibrium is reached in less than 30 min and the maximum Cr(VI) uptake occurred at pH 3.0 under the test conditions. The results are also compared with the commercially available activated carbon. A mathematical model incorporating diffusion, advection, and mass transfer mechanisms available in the literature has been simplified and is then tested to simulate the laboratory and literature data. A simple method for the determination of saturation Cr(VI) concentration along the length of column has been presented. The study reveals that the model incorporating the molecular diffusion and the mass transfer mechanisms simulates better the Cr(VI) adsorption onto tree leaf powders than the literature model and the advection term plays only a negligible role due to low flow rates applied during the experiments. The model parameters, i.e., axial dispersion coefficient, “DL” and the external mass transfer coefficient, “kf” are found in the order of 10?5–10?6?m2/s and 10?9–10?11?m/s, respectively. 相似文献
