The ability of Lactobacillus manihotivorans LMG 18010 to produce l(+)-lactic acid from a high concentration of starch by direct fermentation was investigated. To improve the production of lactic acid from a highly concentrated starch, fed-batch cultures were carried out with different feeding strategies to reduce substrate inhibition. The highest lactic acid production was 71.4 g/L, which was obtained by intermittent feeding of concentrated starch media containing nitrogen sources. After 5 days of fermentation, about 87% of the substrate was consumed with a volumetric productivity of 0.6 g/L·h which is 1.8 times higher than that obtained from batch fermentation. This result clearly shows that intermittant feeding of a highly concentrated substrate solution during fermentation is an efficient way of eliminating substrate inhibition.  相似文献

Conventional lithography methods of gold patterning are based on deposition and lift‐off or deposition and etching. In this letter, we demonstrate a novel method of gold patterning using spin‐coatable gold electron‐beam resist which is functionalized gold nanocrystals with amine ligands. Amine‐stabilized gold electron beam resist exhibits good sensitivity, 3.0 mC/cm2, compared to that of thiol‐stabilized gold electron beam resists. The proposed method reduces the number of processing steps and provides greater freedom in the patterning of complex nanostructures.  相似文献

Band‐like transport behavior of H‐doped transition metal dichalcogenide (TMD) channels in field effect transistors (FET) is studied by conducting low‐temperature electrical measurements, where MoTe2, WSe2, and MoS2 are chosen for channels. Doped with H atoms through atomic layer deposition, those channels show strong n‐type conduction and their mobility increases without losing on‐state current as the measurement temperature decreases. In contrast, the mobility of unintentionally (naturally) doped TMD FETs always drops at low temperatures whether they are p‐ or n‐type. Density functional theory calculations show that H‐doped MoTe2, WSe2, and MoS2 have Fermi levels above conduction band edge. It is thus concluded that the charge transport behavior in H‐doped TMD channels is metallic showing band‐like transport rather than thermal hopping. These results indicate that H‐doped TMD FETs are practically useful even at low‐temperature ranges.  相似文献

Ethereum is arguably the second most popular cryptocurrency-based network after Bitcoin. Both use the distributed ledger technology known as the blockchain, which is considered secure. However, the provided security level is proportional to the number of connected nodes, the number of influential nodes, and the supported amount of hash power. Thus, the knowledge of the network properties and nodes' behavior is helpful to protect the network from possible attacks such as double-spending attacks, DDoS attacks, 51% attacks, and Sybil attacks. This paper proposes a node discovery mechanism, which performs a P2P link discovery on the Ethereum main network. For that, we develop Search-node, a modified version of Ethereum client that searches for all participating nodes in the blockchain network, stores the node information in the Bucket, and then processes the peer discovery method. Based on the collected data, we first visualize the Ethereum network topology and analyze the attributes of the network such as node degree, path length, diameter, and clustering coefficient. We then analyze the node properties and provide analytical results regarding the relationship between nodes, heavily connected nodes, node geo-distribution, security issues, and possible attacks over the influential nodes. As a result, we have identified 68,406 nodes with a total of 642,034 edges. By analyzing the collected data, we have found that the diameter in the Ethereum network is equal to 8. The node degree is over 19, which is two times higher than the default configuration.  相似文献

Abstract

The unstructured grid finite volume method has been applied to predict the linear and nonlinear attenuation characteristics of the expansion chamber type silencer system. In order to achieve grid flexibility and a solution adaptation for geometrically complex flow regions associated with the actual silencers, the unstructured mesh algorithm in context with the node-centered finite volume method has been employed. The validation cases for the linear and nonlinear wave propagation characteristics include the acoustic field of the concentric expansion chamber and the axisymmetric blast flow field with the open end. Effects of the chamber geometry on the nonlinear wave propagation characteristics are discussed in detail.  相似文献

The gastrointestinal (GI) endoscopy has become a standard diagnostic tool for GI ulcers and cancer. In this study we studied endoscopic application of epidermal growth factor-containing chitosan hydrogel (EGF-CS gel) for treatment of GI ulcer. We hypothesized that directional ulcer-coating using EGF-CS gel via endoscope would precipitate ulcer-healing. EGF-CS gel was directly introduced to the ulcer-region after ulceration in acetic acid-induced gastric ulcer (AAU) and mucosal resection-induced gastric ulcer (MRU) rabbit and pig models. The ulcer dimensions and mucosal thicknesses were estimated and compared with those in the control group. Healing efficacy was more closely evaluated by microscopic observation of the ulcer after histological assays. In the AAU model, the normalized ulcer size of the gel-treated group was 2.3 times smaller than that in the non-treated control group on day 3 after ulceration (P < 0.01). In the MRU model, the normalized ulcer size of the gel-treated group was 5.4 times smaller compared to that in the non-treated control group on day 1 after ulceration (P < 0.05). Histological analysis supported the ability of EGF-CS gel to heal ulcers. The present study suggests that EGF-CS gel is a promising candidate for treating gastric bleeding and ulcers.  相似文献

Translationally controlled tumor protein (TCTP), a repressor for Na,K-ATPase has been implicated in the development of systemic hypertension, as proved by TCTP-over-expressing transgenic (TCTP-TG) mice. Aorta of TCTP-TG exhibited hypercontractile response compared to that of non-transgenic mice (NTG) suggesting dys-regulation of signaling pathways involved in the vascular contractility by TCTP. Because dys-regulation of RhoA/Rho kinase pathway is implicated in increased vascular contractility, we examined whether TCTP induces alterations in RhoA pathway in vascular smooth muscle cells (VSMCs). We found that TCTP over-expression by adenovirus infection up-regulated RhoA pathway including the expression of RhoA, and its downstream signalings, phosphorylation of myosin phosphatase target protein (MYPT-1), and myosin light chain (MLC). Conversely, lentiviral silencing of TCTP reduced the RhoA expression and Rho kinase signalings. Using immunohistochemical and Western blotting studies on aortas from TCTP-TG confirmed the elevated expression of RhoA and increase in p-MLC (phosphorylated MLC). In contrast, down-regulation of RhoA and p-MLC were found in aortas from heterozygous mice with deleted allele of TCTP (TCTP+/−). We conclude that up-regulation of TCTP induces RhoA-mediated pathway, and that TCTP-induced RhoA plays a role in the regulation in vasculature. Modulation of TCTP may offer a therapeutic target for hypertension and in vascular contractility dysfunction.  相似文献
